Local corticosterone production and angiotensin-Ⅰconverting enzyme shedding in a mouse model of intestinal inflammation

AIM: To investigate local corticosterone production and angiotensin-Ⅰ converting enzyme(ACE) protein expression and their interaction in healthy and inflamed ***: Acute intestinal inflammation was induced to six weeks old male Balb/c mice by administration of either 3% or 5% dextran sodium sulfate(DSS) in drinking water for 7 d(n = 12 in each group). Healthy controls(n = 12) were given tap water. Corticosterone production and ACE protein shedding were measured from ex vivo incubates of the small and large intestine using EIA and ELISA, respectively. Morphological changes of the intestinal wall were assessed in hematoxylin-eosin stained tissue preparations of jejunum and distal colon. Effects of angiotensin Ⅱ, captopril and metyrapone on corticosterone production was assessed by incubating pieces of small intestine of healthy mice in the presence of 0.1, 1 or 10 μmol/L angiotensin Ⅱ, 1, 10 or 100 μmol/L captopril or 1, 10 or 100 μmol/L metyrapone solutions and measuring corticosterone released to the incubation buffer after 90 min(n = 5 in each group).RESULTS: Both concentrations of DSS induced inflammation and morphological changes in large intestines but not in small intestines. Changes were observed as distortions of the crypt structure, mucosal erosion, immune cell infiltration to the mucosa and submucosal edema. Ex vivo corticosterone production(2.9 ± 1.0 ng/mL vs 2.0 ± 0.8 ng/mL, P = 0.034) and ACE shedding(269.2 ± 97.1 ng/m L vs 175.7 ± 52.2 ng/mL, P = 0.016) were increased in small intestines in 3% DSS group compared to the controls. In large intestine, corticosterone production was increased compared to the controls in both 3% DSS(229 ± 81 pg/mL vs 158 ± 30 pg/mL, P = 0.017) and 5% DSS groups(366 ± 163 pg/mL vs 158 ± 30 pg/mL, P = 0.002). Large intestine ACE shedding was increased in 5% DSS group(41.5 ± 9.0 ng/mL vs 20.9 ± 5.2 ng/mL, P = 0.034). AIM:To study the individual effects of glucocorticoid (GC) therapy on the state ofimmune activation in patient ***:We developed a novel assay in which the effect of corticosteroid-treated patient serum on healthy donor peripheral blood mononuclear cells (target cells) was studied,with a panel of markers for effector [interferon (IFN)γ and interleukin (IL)-5] and regulatory T cells (FOXP3 and glucocorticoid-induced tumor necrosis factor receptor,GITR).The study group comprised 19 children with inflammatory bowel *** individual effect of patient serum on target cells was analyzed prior to GC therapy and 2 wk ***:The effect of GC therapy mediated by patient serum was seen as a decrease in the target cells expression of regulatory T-cell-related markers GITR (median suppression 24%,range of suppression 1%-63%,in 2 cases increase of 6% and 77%,P < 0.01 for mitogen-activated target cells) and FOXP3 (median suppression 33%,range of suppression 0%-79%,in one case an increase of 173%,P < 0.05 for resting cells),and secretion of IFNγ [from a mean of 87 700 pg/mL (SD 33 900 pg/mL) to 60 900 pg/mL (SD 44 200 pg/mL) in mitogen-activated target cells,13 of the cases showed a decrease,P < 0.01].The total or weight-related prednisolone dose did not correlate with the patient-seruminduced changes in the target cell ***:GC response could be monitored at an individual level by studying the effect of patient serum on signaling pathways of target immune cells.

AIM:To study whether immune-activation stage in serum of adult Crohn's disease (CD) patients correlates with disease activity and with treatment response to antitumor necrosis factor-α (TNF-α) ***:Serum samples were obtained from 15 adult CD patients introduced to anti-TNF-α *** individual stage of immune activation was studied applying our new in vitro assay,in which target cells (donor derived peripheral blood mononuclear cells) were cultured with patient serum and the T-cell activation induced by the patient serum was studied using a panel of markers for effector [interferon γ (IFNγ),interleukin (IL)-5] and regulatory T-cells [forkhead transcription factor 3 (FOXP3) and glucocorticoid-induced tumour necrosis factor receptor (GITR)].The endoscopic disease activity was assessed with the Crohn's disease endoscopic index of severity (CDEIS) before and 3 mo after therapy with an anti-TNF-α ***:Low induction of FOXP3 and GITR in target cells cultured in the presence of patient serum was associated with high disease activity *** assessed before therapy (r=-0.621,P=0.013 and r=-0.625,P=0.013,respectively).FOXP3 expression correlated inversely with pre-treatment erythrocyte sedimentation rate (r=-0.548,P=0.034).Low serum induced FOXP3 (r=-0.600,P=0.018) and GITR (r=-0.589,P=0.021) expression and low IFNγ secretion from target cells (r =-0.538,P=0.039) associated with treatment response detected as a decrease in ***:The immune-activation potency in the patient serum prior to anti-TNF-α therapy reflected intestinal inflammation and the therapeutic response.

Modern pig production contributes to many environmental problems that relate to manure, especially in areas with highly intensive production systems and in regions like Asia where the regulative control is not effective. Therefore, the objective of this study was to use three different pig diets varying in dietary protein, fibre and fat as representative for Danish （DK）, Thai （TH） and Vietnamese （VN） pig production to develop and evaluate different approaches to predict/calculate excretion from growing pigs in comparison with the experimentally determined values. Nine female growing pigs were used in a digestibility and balance experiment. Excretion of dry matter （DM）, nitrogen （N）, phosphorus （P） and carbon （C） of the experimental diets were determined. Due to the highest dietary fibre content, VN had the lowest digestibility of N, P and C （73, 49, and 73%, respectively） compared with the DK and TH pig diets. From the known diet composition using standard table values on chemical and nutrient digestibly, high accuracy （bias） and low variation was found and the results could be used for prediction on chemical composition and excretion in faeces and urine in growing pigs. Calculation based on standard values regarding nutrient retention in the pig body as used in the Danish manure normative system （DMNS） showed likewise to be quite useful for quantifying the total excretion of N and P. Overall, the results demonstrate that simple models that require cheap and normally available information on dietary nutrients can give useful information on nutrient excretion in growing pigs.

Strains of live Saccharomyces cerevisiae yeast have exhibited probiotic effects in ruminants. This study investigated the effects of the dietary yeast supplement, S. cerevisiae(Yea-Sacc^(1026)), on primiparous(PP)and multiparous(MP) Egyptian buffaloes in early to mid-lactation. Lactating buffaloes were fed either a basal total mixed ration(TMR, control; 4 PP and 8 MP) or the basal TMR plus 10 g Yea-Sacc^(1026) per buffalo cow per day(yeast; 4 PP and 8 MP). The feeds were given from 15 days prepartum to 180 days postpartum. Feed intake, body weight, and milk yields(MY) were recorded, and milk and blood samples were collected for analyses. Feces were collected from days 45 to 47 during early lactation and from days 90 to92 during mid-lactation to determine apparent digestibility of dry matter(DM), organic matter(OM),crude protein(CP) and crude fiber(CF). Energy corrected milk yield(ECM), feed conversion, and energy and nitrogen conversion efficiency were calculated. Yeast treated MP buffaloes consumed more DM(P < 0.041) and CP than the untreated control group. Apparent digestibility of DM and OM were significantly greater at mid-lactation for treated versus control group(P = 0.001). Crude fiber digestibility was greater in MP than in PP buffaloes(P = 0.049), and yeast supplemented MP cows had a greater CF digestibility than control MP buffaloes at mid-lactation(P = 0.010). Total blood lipids decreased after yeast supplementation(P= 0.029). Milk yields, ECM, fat and protein yields increased for yeast treated MP buffaloes(P < 0.039). The study concluded that the response to yeast supplementation in buffalo cows is parity dependent. Multiparous buffaloes respond to yeast supplementation with an increased DM intake and CF digestibility without significant weight gains, allowing a greater ECM yield with less fat mobilization. The objective of this literature review was to identify and examine research where Patient Participation was used as a part of intervention targeting general life style among patients who comes in contact with a nurse. A literature search were conducted and included papers where judged by the researcher using recommendation from The Danish Centre for Clinical Guidelines. Analysis of the papers was carried out using Per?kyl? and Ruusuvauoris five components of Patient Participation as a theoretical template. It was concluded that the clinical effects of Patient Participation still needs to be clarified.

Comparative methane estimation from cattle based on total CO_2 production using different techniques

The objective of this study was to compare the precision of CH4 estimates using calculated CO_2(HP) by the CO_2 method(CO_2 T) and measured CO_2 in the respiration chamber(CO_2 R). The CO_2 R and CO_2 T study was conducted as a 3 × 3 Latin square design where 3 Dexter heifers were allocated to metabolic cages for 3 periods. Each period consisted of 2 weeks of adaptation followed by 1 week of measurement with the CO_2 R and CO_2 T. The average body weight of the heifer was 226 ± 11 kg(means±SD). They were fed a total mixed ration, twice daily, with 1 of 3 supplements: wheat(W), molasses(M), or molasses mixed with sodium bicarbonate(Mbic). The dry mater intake(DMI: kg/day) was significantly greater(P < 0.001)in the metabolic cage compared with that in the respiration chamber. The daily CH4(L/day) emission was strongly correlated(r=0.78) between CO_2 T and CO_2 R. The daily CH4(L/kg DMI) emission by the CO_2 T was in the same magnitude as by the CO_2 R. The measured CO_2(L/day) production in the respiration chamber was not different(P = 0.39) from the calculated CO_2 production using the CO_2 T. This result concludes a reasonable accuracy and precision of CH4 estimation by the CO_2 T compared with the CO_2 R.
