Autocatalytic bifunctional supramolecular hydrogels for osteoporotic bone repair

Conventional bone scaffolds, which are mainly ascribed to highly active osteoclasts and an inflammatory microenvironment with high levels of reactive oxygen species and pro-inflammatory factors, barely satisfy osteoporotic defect repair. Herein, multifunctional self-assembled supramolecular fiber hydrogels(Ce–Aln gel) consisting of alendronate(Aln) and cerium(Ce) ions were constructed for osteoporotic bone defect repair. Based on the reversible interaction and polyvalent cerium ions, the Ce–Aln gel, which was mainly composed of ionic coordination and hydrogen bonds, displayed good injectability and autocatalytic amplification of the antioxidant effect. In vitro studies showed that the Ce–Aln gel effectively maintained the biological function of osteoblasts by regulating redox homeostasis and improved the inflammatory microenvironment to enhance the inhibitory effect on osteoclasts. Ribonucleic acid(RNA) sequencing further revealed significant downregulation of various metabolic pathways, including apoptosis signaling,hypoxia metabolism and tumor necrosis factor-alpha(TNF-α) signaling via the nuclear factor kappa-B pathway after treatment with the Ce–Aln gel. In vivo experiments showed that the clinical drug-based Ce–Aln gel effectively promoted the tissue repair of osteoporotic bone defects by improving inflammation and inhibiting osteoclast formation at the defect. Notably, in vivo systemic osteoporosis was significantly ameliorated, highlighting the strong potential of clinical translation for precise therapy of bone defects.

Laser welding study of vacuum sintered HUST-1 lunar regolith simulant

Efforts are underway to establish a permanent lunar base on the Moon. In situ lunar regolith is anticipated to be useful as a building material after sintering. However, sintering lunar regolith into a large-scale structure presents challenges. Therefore, the key to lunar construction lies in assembling multiple small-sized sintered modules into a stable, large-sized structure. This study explored the feasibility of welding the sintered HUST-1 lunar regolith simulant(HLRS) using a laser device and conducted experiments using lasers of varying power. The microstructure, mineral composition, element distribution, and shear strength of the welded joint were investigated. A few low-melting minerals were fused and vaporized during welding, leading to the generation of thermal decomposition gas. Furthermore, the welded joint exhibited numerous micro-cracks, pores, and bubbles,resulting in reduced weld shear strength. Finally, the influence of laser power on weld shear strength was investigated, revealing that the highest shear strength(15.69 N/cm) was achieved at a laser power of 1000 W. This study demonstrates the feasibility of laser welding of sintered HLRS for the first time, with potential applications in lunar base construction.

Role of 5-hydroxytryptamine type 3 receptors in the regulation of anxiety reactions

5-Hydroxytryptamine(5-HT)type 3 receptor(5-HT_(3)R)is the only type of ligand-gated ion channel in the 5-HT receptor *** the high permeability of Na+,K+,and Ca2+and activation of subsequent voltage-gated calcium channels(VGCCs),5-HT_(3)R induces a rapid increase of neuronal excitability or the release of neurotransmitters from axon terminals in the central nervous system(CNS).5-HT_(3)Rs are widely expressed in the medial prefrontal cortex(mPFC),amygdala(AMYG),hippocampus(HIP),periaqueductal gray(PAG),and other brain regions closely associated with anxiety *** have a bidirectional regulatory effect on anxiety reactions by acting on different types of cells in different brain regions.5-HT_(3)Rs mediate the activation of the cholecystokinin(CCK)system in the AMYG,and theγ-aminobutyric acid(GABA)“disinhibition”mechanism in the prelimbic area of the mPFC promotes anxiety by the activation of GABAergic intermediate inhibitory neurons(IINs).In contrast,a 5-HT_(3)R-induced GABA“disinhibition”mechanism in the infralimbic area of the mPFC and the ventral HIP produces anxiolytic effects.5-HT_(2)R-mediated regulation of anxiety reactions are also activated by 5-HT_(3)R-activated 5-HT release in the HIP and *** provides a theoretical basis for the treatment of anxiety disorders or the production of anxiolytic drugs by targeting 5-HT_(3)***,given the circuit specific modulation of 5-HT_(3)Rs on emotion,systemic use of 5-HT_(3)R agonism or antagonism alone seems unlikely to remedy anxiety,which deeply hinders the current clinical application of 5-HT_(3)R ***,the exploitation of circuit targeting methods or a combined drug strategy might be a useful developmental approach in the future.

Distribution of oxide inclusions in H13 castings under super-gravity field with multi-stage rotation speeds

The 42 kg industrial H13 castings were prepared by different super-gravity fields with multi-rotation speeds,and the distribution of oxide inclusions in the castings was *** addition,the inward movement Reynolds number and inward movement time of oxide inclusions as well as the solidification time of molten steel at different positions in the castings were calculated to clarify the removal mechanism of oxide inclusions in super-gravity *** results show that the large size(i.e.,greater than 10μm)oxide inclusions are mainly concentrated in the inner and outer parts of the super-gravity castings with constant rotation speed(500 r min^(-1))and five-stage rotation speeds(500,600,750,850,and 950 r min^(-1)),respectively,while there are no large oxide inclusions in the super-gravity castings with three-stage rotation speeds(500,600,and 750 r min^(-1)).Although an increase in the particle size of inclusion and the rotation speed in super-gravity field is conducive to the increase in the inward movement Reynolds number of oxide inclusions and reduction in the inward movement time of oxide inclusions,it will reduce the local solidification time of molten *** the range of the rotation speed studied,the super-gravity field with three-stage rotation speeds has the best effect on the removal of inclusions in H13 molten steel.

A data-driven model of drop size prediction based on artificial neural networks using small-scale data sets

An artificial neural network(ANN)method is introduced to predict drop size in two kinds of pulsed columns with small-scale data *** training,the deviation between calculate and experimental results are 3.8%and 9.3%,*** ANN model,the influence of interfacial tension and pulsation intensity on the droplet diameter has been *** size gradually increases with the increase of interfacial tension,and decreases with the increase of pulse *** can be seen that the accuracy of ANN model in predicting droplet size outside the training set range is reach the same level as the accuracy of correlation obtained based on experiments within this *** two kinds of columns,the drop size prediction deviations of ANN model are 9.6%and 18.5%and the deviations in correlations are 11%and 15%.

Non-alcoholic fatty liver disease(NAFLD)is a pressing global health concern that is associated with metabolic syndrome and *** the basis of the insights provided by Jiang et al,this editorial presents an exploration of the potential of mesenchymal stem cells(MSCs)for NAFLD *** have numerous desirable characteristics,including immunomodulation,anti-inflammatory pro-perties,and tissue regeneration promotion,rendering them attractive candidates for NAFLD *** preclinical and early clinical studies have high-lighted the efficacy of MSCs in improving liver function and reducing disease severity in NAFLD ***,MSC heterogeneity,long-term safety concerns,and unoptimized therapeutic protocols remain substantial *** these challenges through standardized protocols and rigorous clinical trials is essential to the safe and successful application of MSCs in NAFLD *** research into MSC mechanisms and therapeutic optimization is required to improve treatments for NAFLD and related liver diseases.
