Amino acids and mammary gland development:nutritional implications for milk production and neonatal growth

Milk is synthesized by mammary epithelial cells of lactating mammals. The synthetic capacity of the mammary gland depends largely on the number and efficiency of functional mammary epithelial cells. Structural development of the mammary gland occurs during fetal growth, prepubertal and post-pubertal periods, pregnancy, and lactation under the control of various hormones （particularly estrogen, growth hormone, insulin-like growth factor-I, progesterone, placental lactogen, and prolactin） in a species- and stage-dependent manner. Milk is essential for the growth, development, and health of neonates. Amino acids （AA）, present in both free and peptide-bound forms, are the most abundant organic nutrients in the milk of farm animals. Uptake of AA from the arterial blood of the lactating dam is the ultimate source of proteins （primarily 13-casein and a-lactalbumin） and bioactive nitrogenous metabolites in milk. Results of recent studies indicate extensive catabolism of branched-chain AA （leucine, isoleucine and valine） and arginine to synthesize glutamate, glutamine, alanine, aspartate, asparagine, proline, and polyamines. The formation of polypeptides from AA is regulated not only by hormones （e.g., prolactin, insulin and glucocorticoids） and the rate of blood flow across the lactating mammary gland, but also by concentrations of AA, lipids, glucose, vitamins and minerals in the maternal plasma, as well as the activation of the mechanistic （mammalian） target rapamycin signaling by certain AA （e.g., arginine, branched-chain AA, and glutamine）. Knowledge of AA utilization （including metabolism） by mammary epithelial cells will enhance our fundamental understanding of lactation biology and has important implications for improving the efficiency of livestock production worldwide.

Biochar has been shown to be an effective organic soil amendment for the in-situ immobilization of lead(Pb).Little research has been done on the effects of biochar and zeolite interactions on Pb immobilization in contaminated calcareous ***,the aim of this study was to investigate the effects of different levels of zeolite(2%and 4%,weight:weight)and biochars(3%,weight:weight)prepared from green compost(GB),poultry manure(PB),and municipal waste(MB)on the content of Pb in spinach shoots and Pb stabilization in a greenhouse experiment conducted using a Pb-contaminated calcareous *** of GB and PB significantly decreased the Pb mobility factor(MF)from 2.8%to 4.6%and the cumulative Pb release(diethylenetriamine pentaacetate(DTPA)extraction)from 16.8%to 20.1%in the calcareous soil,which were further enhanced by the combined addition of zeolite,with Pb MF reduction from 4.4%to 8.4%and cumulative Pb release reduction from 24.8%to 28.6%.The enhancement of soil Pb immobilization was attributed to the properties of the amendments,such as high pH,ash content,and phosphorus content(10.5^(-1)1.0,390-435 g kg^(-1),and 2.5-4.3 g kg^(-1),respectively).Results of the soil Pb sequential extraction and Pb release kinetics were highly and significantly correlated with spinach shoot Pb *** chemical data showed that the application of MB with 4%zeolite was the most effective treatment for immobilizing Pb(8.4%reduction in Pb MF and 28.6%reduction in cumulative DTPA-extractable Pb),which is in agreement with the results of the spinach shoot Pb(89.0%reduction in Pb uptake).

AIM： TO determine p15 promoter hypermethylation in gastric tumoral tissue and serum samples, its impact on p16-protein expression, and correlation with clinical and histological features. METHODS： Samples were obtained from 52 histologically confirmed cases of gastric adenocarcinoma. Gastric tissue and serum of 50 age- and sex-matched individuals with normal gastroscopy and biopsy were obtained as control samples. Methylation-specific polymerase chain reaction （MSP） was used to evaluate methylation status of p16 promoter, p16-protein expression was analyzed by immunohistochemical staining on paraffin-embedded sections. RESULTS： Methylation was detected in 44.2% （23/52） of tumoral tissues. 60.9% of them were also methylated in serum, i.e., 26.9% of all patients （14/52）. Methylation was not detected in tissue and sera of control samples. p16-protein expression was decreased in 61.5% of cases （32/52）, and was significantly associated with promoter hypermethylation （P 〈 0.001）. Methylation was significantly more frequent in higher pathological grades （P 〈 0.05）. Methylation was not associated with other clinicopathological features and environmental factors including Hpylori infection and smoking. CONCLUSION： p16 promoter hypermethylation is an important event in gastric carcinogenesis. It is the principle mechanism of p16 gene silencing. It is related to malignant tumor behavior. Detection of DNA methylation in serum may be a biomarker for early detection of gastric cancer.

Surrogate safety measures （SSM） are suitable tools to detect dangerous situations. These indicators can be applied as a warning strategy in collision avoidance systems （CAS）. Timeto-collision （TTC） and post-encroachment time （PET） are two important time-based SSM that identify the probability of a rear-end collision. TIC refers to the imminent danger, and PET implies the potential danger. However, sometimes the results from each indicator are inconsistent. An appropriate warning strategy for CAS can be developed using a new index that combines the properties of both TTC and PET. For this purpose, a new mixed index （MI） is proposed. In order to develop this MI, three main microscopic parameters, clearance, speed and the relative speed, are simultaneously applied to the leading vehicle. To cali- brate MI, based on a fuzzy inference system （FIS）, a value would be determined by a combination of TTC and PET at each instant and then by regression analysis the model parameters would be determined. Finally, MI, TIC and PET values for real car-following scenarios on the 1-80 freeway are determined and compared. The results show that MI may be more suitable in detecting the rear-end collision risk within the proper time and with less errors.
