Pro-resolving lipid mediator reduces amyloid-β42–induced gene expression in human monocyte–derived microglia

Specialized pro-resolving lipid mediators including maresin 1 mediate resolution but the levels of these are reduced in Alzheimer's disease brain, suggesting that they constitute a novel target for the treatment of Alzheimer's disease to prevent/stop inflammation and combat disease pathology. Therefore, it is important to clarify whether they counteract the expression of genes and proteins induced by amyloid-β. With this objective, we analyzed the relevance of human monocyte–derived microglia for in vitro modeling of neuroinflammation and its resolution in the context of Alzheimer's disease and investigated the pro-resolving bioactivity of maresin 1 on amyloid-β42–induced Alzheimer's disease–like inflammation. Analysis of RNA-sequencing data and secreted proteins in supernatants from the monocyte-derived microglia showed that the monocyte-derived microglia resembled Alzheimer's disease–like neuroinflammation in human brain microglia after incubation with amyloid-β42. Maresin 1 restored homeostasis by down-regulating inflammatory pathway related gene expression induced by amyloid-β42 in monocyte-derived microglia, protection of maresin 1 against the effects of amyloid-β42 is mediated by a re-balancing of inflammatory transcriptional networks in which modulation of gene transcription in the nuclear factor-kappa B pathway plays a major part. We pinpointed molecular targets that are associated with both neuroinflammation in Alzheimer's disease and therapeutic targets by maresin 1. In conclusion, monocyte-derived microglia represent a relevant in vitro microglial model for studies on Alzheimer's disease-like inflammation and drug response for individual patients. Maresin 1 ameliorates amyloid-β42–induced changes in several genes of importance in Alzheimer's disease, highlighting its potential as a therapeutic target for Alzheimer's disease.

In recent years, cyber attacks have posed great challenges to the development of cyber-physical systems. It is of great significance to study secure state estimation methods to ensure the safe and stable operation of the system. This paper proposes a secure state estimation for multi-input and multi-output continuous-time linear cyber-physical systems with sparse actuator and sensor attacks. First, for sparse sensor attacks, we propose an adaptive switching mechanism to mitigate the impact of sparse sensor attacks by filtering out their attack modes. Second, an unknown input sliding mode observer is designed to not only observe the system states, sensor attack signals, and measurement noise present in the system but also counteract the effects of sparse actuator attacks through an unknown input matrix. Finally, for the design of an unknown input sliding mode state observer, the feasibility of the observing system is demonstrated by means of lyapunov functions. Additionally, simulation experiments are conducted to show the effectiveness of this method.

The global disease burden of diabetes mellitus is high. It is well-established that prediabetes is reversible but it is unclear whether diabetes is reversible once it has been diagnosed. The objective of this narrative review is to review the evidence of reversibility of diabetes me-llitus and stimulate interest in prolonged remission as a treatment target. The current evidence for bariatric surgery is stronger than intensive medical management and the evidence is stronger for type 2 diabetes pa-tients compared with type 1 diabetes patients. It is also unclear whether non obese diabetes patients would benefit from such interventions and the duration of diabetes before diabetes become irreversible. Further research is needed in this area especially with regards to the subgroup of diabetes patient who will benefit from these interventions and the long term safety and efficacy remains unknown especially with intensive me-dical management.

A new technology-rotationally accelerated shot peening（RASP）, was developed to prepare gradient structured materials. By using centrifugal acceleration principle and large steel balls, the RASP technology can produce much higher impact energy compared to conventional shot peening. As a proof-of-concept demonstration, the RASP was utilized to refine the surface layer in pure copper（Cu） with an average grain size of 85 nm. The grain size increases largely from surface downwards the bulk, forming an800 ？m thick gradient-structured surface layer and consequently a micro-hardness gradient. The difference between the RASP technology and other established techniques in preparing gradient structured materials is discussed. The RASP technology exhibits a promoting future for large-scale manufacturing of gradient materials.

Microseismic(MS)event locations are vital aspect of MS monitoring technology used to delineate the damage zone inside the surrounding rock ***,complex geological conditions can impose significantly adverse effects on the final location *** achieve a high-accuracy location in a complex cavern-containing structure,this study develops an MS location method using the fast marching method(FMM)with a second-order difference approach(FMM2).Based on the established velocity model with three-dimensional(3D)discrete grids,the realization of the MS location can be achieved by searching the minimum residual between the theoretical and actual first arrival ***,based on the calculation results of FMM2,the propagation paths from the MS sources to MS sensors can be obtained using the linear interpolation approach and the Runge–Kutta *** methods were validated through a series of numerical *** addition,our proposed method was applied to locate the recorded blasting and MS events that occurred during the excavation period of the underground caverns at the houziyan hydropower *** location results of the blasting activities show that our method can effectively reduce the location error compared with the results based on the uniform velocity ***,the obtained MS location was verified through the occurrence of shotcrete fractures and spalling,and the monitoring results of the in-situ multipoint *** proposed method can offer a more accurate rock fracture location and facilitate the delineation of damage zones inside the surrounding rock mass.

During surveys, water layers may interfere with the detection of oil layers. In order to distinguish between oil and water layers in a porous cracked medium, research on the properties of cracks and oil and water layers and their relation to acoustic logging rules is essential. On the basis of hudson＇s crack theory, we simulated oil and water layers in crack-porous medium with different crack parameters corresponding to the well-field response. We found that in a cracked medium with high crack angle or low number density of cracks, compressional and shear wave velocities are sensitive to crack characteristics; further, these velocities are more sensitive to crack characteristics when the waves propagate through the water layer than when they propagate through the oil layer. Compressional and shear wave velocities increase with an increase in crack angle： in the water layer, the increase is approximately linear. On comparing the full waveforms observed in the oil and water layers, we find that the amplitudes of most waves are higher in the water layer. Among the considered waves, the Stoneley wave suffers maximum amplitude attenuation in the oil layer. The maximum excitation intensity for oil layer is greater than that for the water layer. These results can guide further cracked media logging field exploration work.
