建立一种可同时快速检测大肠杆菌O157:h7、单增李斯特菌和蜡样芽孢杆菌的多重实时定量PCR(qPCR)方法。依据大肠杆菌O157:h7 tir基因、单增李斯特菌mpl基因和蜡样芽孢杆菌entFM基因的保守序列分别设计特异性引物和TaqMan探针,建立多重qPCR反应体系,进行灵敏度、特异性和稳定性试验,同步检测人工染菌牛奶样品中的病原菌并与国家标准方法作对比。结果表明,建立的多重qPCR方法灵敏度高,最低检出限为12 CFU/mL;特异性强,只对3种目标菌进行PCR扩增;稳定性好,各重复性试验中Ct值的变异系数h。本研究建立的TaqMan多重qPCR方法能同时快速、准确地检测乳品中的大肠杆菌O157:h7、单增李斯特菌和蜡样芽孢杆菌,为食品安全提供技术支撑。

Metabolic flexibility has emerged as a critical determinant of CD8+T-cell antitumor activity,yet the mechanisms driving the metabolic flexibility of T cells have not been *** this study,we investigated the influence of the nuclear cap-binding complex(CBC)adaptor protein ARS2 on mature T *** doing so,we discovered a novel signaling axis that endows activated CD8+T cells with flexibility of glucose ***2 upregulation driven by CD28 signaling reinforced splicing factor recruitment to pre-mRNAs and affected approximately one-third of T-cell activation-induced alternative splicing *** these effects,the CD28-ARS2 axis suppressed the expression of the M1 isoform of pyruvate kinase in favor of PKM2,a key determinant of CD8+T-cell glucose utilization,interferon gamma production,and antitumor effector ***,PKM alternative splicing occurred independently of CD28-driven PI3K pathway activation,revealing a novel means by which costimulation reprograms glucose metabolism in CD8+T cells.

Toxicological effects of PBDEs on carassius aurats

In this study the effects of polybrominated diphenyl ether compounds (PBDEs), BDE-47 and BDE-99 on carassius aurats were discussed. The reproductive capacity viability and proliferation percent decreased with in-creasing BDE-47and BDE-99 concentrations (physiological bio-markers such as cholesterol (ChL), acetylcholinesterase (AChE), ethoxyresorufin-O-deethylase (EROD) and vitamin were studied to reveal the toxicological effects of BDE-47 and BDE-99 exposure. ChL levels generally decrease with increasing BDE-47 exposure. however, there were no significant differences between the samples and the con-trol. As to BDE-99, the ChL level decreased as compared with the control level when the BDE-99 concentrations reached 20 μg/L (phe degree of inhibition of the AChE increased with the increase of DE-47and BDE-99 concentrations. The EROD activity during 72 hours' exposure significantly increased to 20 μg/L for BDE-47 (phowever, EROD activity was not significantly different at different concentrations of BDE-99 (phepatocytes reacted to the BDE-47 exposure with increasing EROD activities in the dose depend-ent manner. Although at lower concentrations, the induced effects of BDE-47 and BDE-99 were not significantly different, though BDE-47 showed strong induced effects on the EROD activity with increasing concentrations in carassius aurats hepatocytes. In contrast, BDE-99 did not influence EROD activity consistently. The VA level of carassius aurats was significantly different from that of the controls for BDE-99 (phe VA level was significantly lower of carassius aurats than the controls for BDE-47 (phL, AChE, EROD, and VA levels in carassius aurats were differently affected by BDE-47 and BDE-99 exposures, therefore, they would be good indicators for exposure.

为了分析新风热回收系统在夏热冬冷地区住宅建筑中应用时的节能性能,对该系统在各季节的实际节能效果作出判断。通过现场测试计算出新风热回收系统的热交换效率、热回收量以及室内的新风负荷,运用PKPM能耗模拟软件建立两种建筑模型,计算分析某住宅建筑使用新风热回收系统前后的用电能耗和相对节能率。研究结果显示,使用新风热回收系统后,建筑相对节能率由2.60%提高到6.09%,建筑冬季供暖用电节能3948.35 kW·h,夏季空调用电节能2036.22 kW·h,总用电能耗降低了5984.57 kW·h,实现能源节约3.57%。上述研究内容为新风热回收系统的设计、运用及研究工作提供了借鉴帮助。

目的研究交趾黄檀Dalbergia cochinchinensis Pierre ex Laness的新黄酮类成分及其抗h9c2心肌细胞缺氧/复氧损伤活性。方法交趾黄檀70%乙醇提取物采用硅胶、Sephadex Lh-20、反相制备hPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用CCK-8法检测其对h9c2心肌细胞的活性及对h9c2细胞缺氧/复氧损伤的保护作用,并分析其构效关系。结果从中分离得到12个化合物,分别鉴定为阔叶黄檀酚(1)、5-O-methyllatifolin(2)、mimosifoliol(3)、5-O-methydalbergiphenol(4)、dalbergiphenol(5)、cearoin(6)、2,4-dihydroxy-5-methoxy-benzophenone(7)、2-hydroxy-4,5-dimethoxybenzophenone(8)、melannoin(9)、2,2′,5-trihydroxy-4-methoxybenzophenone(10)、黄檀素(11)、4-甲氧基黄檀醌(12)。黄檀酚及黄檀内酯类化合物对h9c2细胞毒性较小,黄檀酚类化合物抗h9c2心肌细胞缺氧/复氧损伤活性较强。结论化合物8为新天然产物,化合物4、9为首次从该植物中分离得到。黄檀酚类化合物可能是抗h9c2细胞缺氧/复氧损伤的主要新黄酮类成分。

Combinations of metal and lanthanide oxides have been done through casted films for potential medical applications. In this regard, samarium(Ⅲ) oxide/chromium(Ⅲ) oxide/graphene oxide(GO)/polycaprolactone(PCL) based films nano-composites(NCs) were fabricated, pointing their utilization as a biological scaffold for wound dressing purposes. Also, samarium(Ⅲ) oxide and chromium(Ⅲ) oxide have been merged as promising optical constituents due to their unique optical behavior. The structural and compositional examination of the studied NCs was executed by X-ray diffraction(XRD), Raman, and field emission scanning electron microscopy(FESEM). The Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL NC exhibits a surface with a lower roughness degree owing to the presence of GO. Cr_(2)O_(3)shows size reduction upon GO insertion to reach 1.2 μm as the average grain size, whilst Sm_(2)O_(3)records an average grain size of less than 1 μm. As well, the polymeric nano-compositions exhibit variation in contact angle values that hit 29.76°± 3.52°for Sm_(2)O_(3)/PCL, and 48.62°± 1.37°for Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL as the second lowest contact angle. The optical behavior contributes to absorption edge relocation along the x-axis from 1.7 eV for pure PCL, to 2.65 eV for Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL. Regarding biological responses, the cell exposed to 2.5 μg/m L of Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL shows cell viability of 119.31%, while 5 μg/m L hits 99.6%. Additionally, the resulting cell attachment micrographs show layers of fibroblast tissue, besides the proliferation and growth of cultivated cells. Thus, the Sm_(2)O_(3)/Cr_(2)O_(3)/GO/PCL scaffold provides 3D proliferation of fibroblast cells endorsing the wound healing process.

Recently,great attention has been paid to geopolymer concrete due to its advantageous mechanical and environmentally friendly *** effort has been made in experimental studies to advance the understanding of geopolymer concrete,in which compressive strength is one of the most important *** facilitate engineering work on the material,an efficient predicting model is *** this study,three machine learning(ML)-based models,namely deep neural network(DNN),K-nearest neighbors(KNN),and support vector machines(SVM),are developed for forecasting the compressive strength of the geopolymer concrete.A total of 375 experimental samples are collected from the literature to build a database for the development of the predicting models.A careful procedure for data preprocessing is implemented,by which outliers are examined and removed from the database and input variables are standardized before feeding to the fitting *** standard K-fold cross-validation approach is applied for evaluating the performance of the models so that overfitting status is well managed,thus the generalizability of the models is *** effectiveness of the models is assessed via statistical metrics including root mean squared error(RMSE),mean absolute error(MAE),correlation coefficient(R),and the recently proposed performance index(PI).The basic mean square error(MSE)is used as the loss function to be minimized during the model fitting *** three ML-based models are successfully developed for estimating the compressive strength,for which good correlations between the predicted and the true values are obtained for DNN,KNN,and *** numerical results suggest that the DNN model generally outperforms the other two models.

Extension of the theory of adherence to treatment in patients with coronary heart disease

Background: Increased knowledge of the concept of adherence is needed for development patient-centered care, nursing interventions, and guidelines for patients with coronary heart disease (ChD). The aim of this study was to test and extend the Theory of Adherence to Treatment regarding informational support in patients with ***: The study utilized an explanatory and descriptive survey. The study was conducted in 2013 and involved 416 patients in five hospitals in Finland. The Adherence of Patients with Chronic Disease instrument and the Social Support for People with ChD instrument were used. The model was tested using structural equation modeling (SEM).Results: SEM confirmed direct associations between motivation (β = 0.49,p herence to a healthy lifestyle and medication. Informational support included information and advice on ChD risk factors, physical exercise, chest pain, medication, continuum of care, and rehabilitation. Indirect standardized path coefficients varied between 0.14 and 0.45. The model explained 45% of adherence to a healthy lifestyle and ***: The results of this study showed that informational support is a justified extension to the original Theory of Adherence to Treatment in Patients with ChD. Informational support seems to offer a new perspective that can be used to develop patient-centered nursing interventions and thus support adherence to treatment by patients with a lifelong disease such as ChD.
