Intra-tum.r h.terogeneity is now arguably one of th. m.st-studied topics in tum.r biology,as it represents a m.jor obstacle to effective cancer *** tum.r cells are h.gh.y diverse at genetic,epigenetic,and ph.notypic levels,intra-tum.r h.terogeneity can be assum.d as an im.ortant contributing factor to th. nullification of ch.m.th.rapeutic effects,and recurrence of th. *** on th. role of h.terogeneous subpopulations of cancer cells with.varying cell-cycle dynam.cs and beh.vior during cancer progression and treatm.nt;h.rein,we aim.to establish.a com.reh.nsive definition for adaptation of neoplastic cells against *** discuss two parallel and yet distinct subpopulations of tum.r cells th.t play pivotal roles in reducing th. effects of ch.m.th.rapy:"resistant"and"tolerant"***,th.s review also h.gh.igh.s th. im.act of th. quiescent ph.se of th. cell cycle as a survival m.ch.nism.for cancer *** understanding th. m.ch.nism. under-lying th. quiescence,it provides an insigh.ful perspective on cancer stem.cells(CsCs)and th.ir dual and intertwined functions based on th.ir cell cycle state in response to ***-over,CSCs,epith.lial-m.sench.m.l transform.d cells,circulating tum.r cells(CTCs),and dissem.nated tum.r cells(DTCs),wh.ch.are m.stly in a quiescent state of th. cell cycle are proved to h.ve m.ltiple biological links and can be im.licated in our viewpoint of cell cycle h.terogeneity in ***,increasing our knowledge of cell cycle h.terogeneity is a key to identifying new th.rapeutic solutions,and th.s em.rging concept m.y provide us with.new opportunities to prevent th. dreadful cancer recurrence.

Th.s cross-sectional study investigates a connection between fem.le infertility and obesity across various regions of th. United States, utilizing data from.th. NIh.s “All of Us” Research.r Workbench. Analysis suggested th.t th. South.rn region exh.bited th. h.gh.st percentage of concurrently infertile and obese fem.les at 32.3%, in contrast to th. lowest in th. Western region at 28.7%. Th. findings underscore th. significant regional disparities in obesity-related infertility, particularly in m.dically underserved areas wh.re h.alth.are access is lim.ted. Th. study h.gh.igh.s th. urgent need for targeted public h.alth.interventions, particularly in th. South.rn United States, to address th. rising prevalence of obesity and its im.act on infertility. Furth.r, th.se results advocate for enh.nced public h.alth.education and policies aim.d at im.roving h.alth.are accessibility, with.th. goal of m.tigating th. adverse effects of obesity on reproductive h.alth.

NSUN2 m.diates distinct path.ays to regulate enterovirus 71 replication

Increasing evidences suggest th.t th. m.th.ltransferase NSUN2 catalyzes 5-m.th.lcytosine (m.C) m.difications on viral RNAs, wh.ch.are essential for th. replication of various viruses. Despite th. function of m.C deposition is well ch.racterized, oth.r potential roles of NSUN2 in regulating viral replication rem.in largely unknown. In th.s study, th. m.C m.dified residues catalyzed by NSUN2 on enterovirus 71 (EV71) RNAs were m.pped. NSUN2, along with.m.C m.difications, played m.ltiple roles during th. EV71 life cycle. Functional m.C m.dified nucleotides increased th. translational efficiency and stability of EV71 RNAs. Additionally, NSUN2 was found to target th. viral protein VP1 for binding and prom.te its stability by inh.biting th. ubiquitination. Furth.rm.re, both.viral replication and path.genicity in m.ce were largely attenuated wh.n functional m.C residues were m.tated. Taken togeth.r, th.s study ch.racterizes distinct path.ays m.diated by NSUN2 in regulating EV71 replication, and h.gh.igh.s th. im.ortance of its catalyzed m.C m.difications on EV71 RNAs for th. viral replication and path.genicity. © 2024 Th. Norovirus (NoV) infection is a m.jor cause of gastroenteritis worldwide. Th. virus poses great ch.llenges in developing vaccines with.broad im.une protection due to its genetic and antigenic diversity. To date, th.re are no approved NoV vaccines for clinical use. h.re, we aim.d to develop a broad-acting quadrivalent NoV vaccine based on a ch.m.anzee adenovirus vector, AdC68, carrying th. m.jor capsid protein (VP1) of noroviral GI and GII genotypes. Com.ared to intram.scular (i.m.), intranasal (i.n.), or oth.r prim.-boost im.unization regim.ns (i.m. + i.m., i.m. + i.n., i.n. + i.m.), AdC68-GI.1-GII.3 (E1)-GII.4-GII.17 (E3), adm.nistered via i.n. + i.n. induced h.gh.r titers of serum.IgG antibodies and h.gh.r IgA antibodies in bronch.alveolar lavage fluid (BALF) and saliva against th. four h.m.logous VP1s in m.ce. It also significantly stim.lated th. production of blocking antibodies against th. four genotypes. In response to re-stim.lation with.virus-like particles (VLP)-GI.1, VLP-GII.3, VLP-GII.4, and VLP-GII.17, th. quadrivalent vaccine adm.nistered according to th. i.n. + i.n. regim.n effectively triggered specific cell-m.diated im.une responses, prim.rily ch.racterized by IFN-γ secretion. Furth.rm.re, th. preparation of th.s novel quadrivalent NoV vaccine requires only a single recom.inant adenovirus to provide broad preventive im.unity against th. m.jor GI/GII epidem.c strains, m.king it a prom.sing vaccine candidate for furth.r developm.nt. © 2024 Th.
