AAV for Gene Therapy in Ocular Diseases:Progress and Prospects

Owing to the promising therapeutic effect and one-time treatment advantage, gene therapy may completely change the management of eye diseases, especially retinal diseases. Adeno-associated virus (AAV) is considered one of the most promising viral gene delivery tools because it can infect various types of tissues and is considered as a relatively safe gene delivery vector. The eye is one of the most popular organs for gene therapy, since its limited volume is suitable for small doses of AAV stably transduction. Recently, an increasing number of clinical trials of AAV-mediated gene therapy are underway. This review summarizes the biological functions of AAV and its application in the treatment of various ocular diseases, as well as the characteristics of different AAV delivery routes in clinical applications. Here, the latest research progresses in AAV-mediated gene editing and silencing strategies to modify that the genetic ocular diseases are systematically outlined, especially by base editing and prime editing. We discuss the progress of AAV in ocular optogenetic therapy. We also summarize the application of AAV-mediated gene therapy in animal models and the difficulties in its clinical transformation.

Sodium-ion batteries(SIBs) and hybrid capacitors(SIHCs) have garnered significant attention in energy storage due to their inherent advantages,including high energy density,cost-effectiveness,and enhanced ***,developing high-performance anode materials to improve sodium storage performa nce still remains a major ***,a facile one-pot method has been developed to fabricate a hybrid of MoSeTe nanosheets implanted within the N,F co-doped honeycomb carbon skeleton(MoSeTe/N,F@C).Experimental results demonstrate that the incorporation of large-sized Te atoms into MoSeTe nanosheets enlarges the layer spacing and creates abundant anion vacancies,which effectively facilitate the insertion/extraction of Na^(+) and provide numerous ion adsorption sites for rapid surface capacitive ***,the heteroatoms N,F co-doped honeycomb carbon skeleton with a highly conductive network can restrain the volume expansion and boost reaction kinetics within the *** anticipated,the MoSeTe/N,F@C anode exhibits high reversible capacities along with exceptional cycle *** coupled with Na_(3)V_(2)(PO_(4))_(3)@C(NVPF@C) to form SIB full cells,the anode delivers a reversible specific capacity of 126 ma h g^(-1) after 100 cycles at 0.1 A g^(-1).furthermore,when combined with AC to form SIHC full cells,the anode demonstrates excellent cycling stability with a reversible specific capacity of50 ma h g^(-1) keeping over 3700 cycles at 1.0 A g^(-1).In situ XRD,ex situ TEM characterization,and theoretical calculations(DFT) further confirm the reversibility of sodium storage in MoSeTe/N,F@C anode materials during electrochemical reactions,highlighting their potential for widespread practical *** work provides new insights into the promising utilization of advanced transition metal dichalcogenides as anode materials for Na^(+)-based energy storage devices.

VOCs can exert great harm to both human and environment,and catalytic oxidation is believed to be an effective technique to eliminate these *** this paper,Ag-Mn bimetal catalysts with 10 wt.%of silver were synthesized using doping,impregnation,and reduction methods respectively,and then they were applied to the catalytic oxidation of *** series of characterizations it showed that the loading of silver using reduction method significantly resulted in improved physico-chemical properties of manganese oxides,such as larger surface area and pore volume,higher proportion of surface Mn~(3+)and Mn~(4+),stronger reducibility and more active of surface oxygen species,which were all beneficial to its catalytic *** a result,the Ag-Mn catalysts synthesized by reduction method showed a lower T_(90)value(equals to the temperature at which 90%of initial benzene was removed)of 203℃.Besides,both the used and fresh Ag-Mn catalysts synthesized by reduction method showed preferable stability in this research.

The energy spectrum of energetic electrons is a key factor representing the dynamic variations of Earth’s Van Allen radiation *** measurements have indicated that the commonly used maxwellian and Kappa distributions are inadequate for capturing the realistic spectral distributions of radiation belt *** we adopt the Kappa-type(KT)distribution as the fitting function and perform a statistical analysis to investigate the radiation belt electron flux spectra observed by the Van Allen *** calculating the optimal values of the key KT distribution parameters(i.e.,κandθ2)from the observed spectral shapes,we fit the radiation belt electron fluxes at different L-shells under different geomagnetic *** this manner,we obtain typical values of the KT distribution parameters,which are statistically feasible for modeling the radiation belt electron flux profiles during either geomagnetically quiet or active periods.A comparison of the KT distribution model results with those using the maxwellian or Kappa distribution reveals the advantage of the KT distribution for studying the overall properties of the radiation belt electron spectral distribution,which has important implications for deepening the current understanding of the radiation belt electron dynamics under evolving geomagnetic conditions.

The microstructures and corrosion behaviors of as-cast,T4-treated,and T6-treated Mg-6Gd-3Y-0.5Zr alloys were systematically investigated by SEM,TEM,immersion test,and electrochemical corrosion *** results show that the microstructure of the as-cast alloy is composed ofα-Mg and Mg_(24)(Gd,Y)_(5)eutectic phase,and in T4-treated alloy,Mg_(24)(Gd,Y)_(5)phase dissolves into theα-Mg matrix,leading to an increase in the(Y,Gd)H_(2)*** T6 treatment,nanoscale Mg_(24)(Gd,Y)_(5)phase dispersedly precipitates from theα-Mg matrix,and exhibits a specific orientation relationship with theα-Mg:(332)Mg_((24)(Gd,Y)_(5))//(1011)_(α-Mg),[136]Mg_((24)(Gd,Y)_(5))//[1210]_(α-Mg).The corrosion resistance of the Mg-6Gd-3Y-0.5Zr alloys can be ranked in the following order:T6-treated alloy exhibits the highest corrosion resistance,followed by the T4-treated alloy,and finally,the as-cast *** corrosion products of the alloys are all composed of MgO,Mg(OH)_(2),Gd_(2)O_(3),Y_(2)O_(3),and MgCl_(2).The corrosion behavior of Mg-6Gd-3Y-0.5Zr alloy is closely related to the precipitated *** establishing the relationship between corrosion rate,hydrogen evolution rate,and corrosion potential,it is further demonstrated that during the micro galvanic corrosion process,the coarse Mg_(24)(Gd,Y)_(5)phase in the as-cast alloy undergoes extensive dissolution,and(Y,Gd)H_(2)phase promotes the dissolution of theα-Mg matrix in the T4-treated alloy,intensifying the hydrogen evolution *** T6-treated alloy,with dispersive precipitation of nanoscale Mg_(24)(Gd,Y)_(5)phase,exhibits better corrosion resistance performance.

Bulk nanoporous （np） metallic actuators have attracted increasing attention due to their large strain and low stimulation ***,studies focusing upon the combined effect of composition and structure on the actuation performance of metallic actuators are relatively scarce,and its underlying mechanism needs to be clarified Herein,a series of bulk np-NiPd samples with differen compositions and microstructures were fabricated using a dealloying-coarsening-dealloying strategy and chargecontrolled electrochemical dealloying,and the process involves only one component of precursor *** has been found that the np-NiPd cubes show a composition/structure-dependent mechanical property and electrochemical actuation ***,the npNi70Pd30sample with a homogeneously porous structure and good network connectivity exhibits significantly larger strain amplitude and faster strain rate than other hierarchically porous NiPd samples (np-Ni50Pd50and npNi20Pd80).Moreover,the np-Ni70Pd30sample demonstrates good actuation stability with high strain retention after hundreds of ***,the maximum strain amplitude （1.17%） is even comparable to that of advanced leadfree piezoceramic,and the maximum strain rate exceeds those of many reported metallic actuator *** work indicates that good network connectivity plays a vita role in facilitating large/fast strain response in metallic actuators.
