Objective:Several reports have proposed that lnc RNAs,as potential biomarkers,participate in the progression and growth of malignant ***1 A-AS2 is a novel lnc RNA and potential biomarker,involved in the genesis and development of ***,the molecular mechanism of HIF1 A-AS2 in renal carcinoma is ***:The relative expression levels of HIF1 A-AS2 and miR-30 a-5 p were detected using RT-qPCR in renal carcinoma tissues and cell *** loss-of-function and overexpression,the biological effects of HIF1 A-AS2 and miR-30 a-5 p in kidney carcinoma progression were *** luciferase reporter gene analysis and Western blot were used to detect the potential mechanism of HIF1 A-AS2 in renal ***:HIF1 A-AS2 was upregulated in kidney carcinoma tissues when compared with para-carcinoma tissues(P<0.05).In addition,tumor size,tumor node mestastasis stage and differentiation were identified as being closely associated with HIF1 A-AS2 expression(P<0.05).Knockdown or overexpression of HIF1 A-AS2 either restrained or promoted the malignant phenotype and WNT/β-catenin signaling in renal carcinoma cells(P<0.05).Mi R-30 a-5 p was downregulated in renal cancers and partially reversed HIF1 A-AS2 functions in malignant renal tumor ***1 A-AS2 acted as a micro RNA sponge that actively regulated the relative expression of SOX4 in sponging miR-30 a-5 p and subsequently increased the malignant phenotypes of renal ***1 A-AS2 showed a carcinogenic effect and miR-30 a-5 p acted as an antagonist of the anti-oncogene effects in the pathogenesis of renal ***:The HIF1 A-AS2-miR-30 a-5 p-SOX4 axis was associated with the malignant progression and development of renal *** relative expression of HIF1 A-AS2 was negatively correlated with the expression of miR-30 a-5 p,and was closely correlated with SOX4 mRNA levels in renal cancers.

The phenomenon of a target echo peak overlapping with the backscattered echo peak significantly undermines the detection range and precision of underwater laser *** overcome this issue,we propose a four-quadrant dual-beam circumferential scanning laser fuze to distinguish various interference signals and provide more real-time data for the backscatter filtering *** enhances the algorithm loading capability of the *** order to address the problem of insufficient filtering capacity in existing linear backscatter filtering algorithms,we develop a nonlinear backscattering adaptive filter based on the spline adaptive filter least mean square(SAF-LMS)*** also designed an algorithm pause module to retain the original trend of the target echo peak,improving the time discrimination accuracy and anti-interference capability of the ***,experiments are conducted with varying signal-to-noise ratios of the original underwater target echo *** experimental results show that the average signal-to-noise ratio before and after filtering can be improved by more than31 d B,with an increase of up to 76%in extreme detection distance.

为了实现煤层气储层高效开发,需要开展储层改造,水力压裂则是最重要的改造手段。面对深部煤储层加砂困难易砂堵、支撑缝长较短、改造范围较小等问题,常规“活性水+石英砂”的砂液组合已无法满足压裂改造需要,因此从支撑剂角度入手,提出一种自悬浮压裂液体系,以提升压裂改造效果。本文对该体系的悬浮性能、流变性能、破胶性能、压裂液破胶液残渣含量、岩心伤害情况开展室内实验评价,结合裂缝导流能力模拟、裂缝监测结果和实际产气量验证了自悬浮支撑剂与石英砂的组合加砂模式可以提高压裂改造效果,大幅提升煤层气井的产气量,可进一步应用于现场。

目的探讨共刺激分子B7H3(costimulatory molecule B7H3,B7H3)通过调控DNA拓扑异构酶Ⅱα(DNA topoisom eraseⅡalpha,TOP2A)的表达对乳腺癌多柔比星化疗敏感性的影响。方法收集临床标本,采用免疫组织化学染色验证B7H3表达与多柔比星化疗敏感性的相关性。采用成簇规律间隔短回文重复/Cas9基因编辑技术敲除B7H3,通过蛋白质印迹法和流式细胞术验证敲除结果。采用半抑制浓度值分析、集落形成、蛋白质印迹法检测凋亡蛋白、荧光双染色检测凋亡细胞以及细胞周期测定等方法验证B7H3表达与多柔比星作用的敏感性。小干扰TOP2A及其过表达TOP2A回复实验检测细胞凋亡。构建小鼠皮下成瘤模型验证体内B7H3表达与多柔比星化疗的作用。结果高表达B7H3的肿瘤组织对多柔比星治疗更加敏感。成功构建B7H3敲除的乳腺癌细胞。B7H3高表达的乳腺癌细胞对多柔比星作用更敏感。B7H3通过激活核因子κB(nuclear factor-κB,NF-κB)途径促进TOP2A的表达,进而增加乳腺癌细胞对多柔比星作用的敏感性。小鼠体内实验证明,B7H3高表达可增加乳腺癌对多柔比星化疗的敏感性。结论B7H3通过激活NF-κB通路调控TOP2A的表达使乳腺癌对多柔比星化疗更敏感。

Myeloid-derived suppressor cells(MDSC) potently inhibit antitumor immune responses, and thereby promoting tumor progression and metastasis. However, the nature of human tumor-infiltrating MDSC remains poorly characterized. Here, we found that B7-H3 was exclusively expressed on a subset of intratumoral CD14+HLA-DR-/low MDSC but not on those from adjacent normal lung tissues of the cancer patients. Cytokine analysis revealed that B7-H3+CD14+HLA-DR-/low MDSC(B7-H3+MDSC) produced higher levels of IL-10 and TNF-α but lower levels of the IL-1β and IL-6 when compared with B7-H3-CD14+HLA-DR-/low MDSC(B7-H3-MDSC). In a murine lung cancer model, B7-H3+MDSC were found only in the tumor microenvironment and their frequencies increased during tumor progression. Clinical data analysis indicated that higher frequencies of B7-H3+MDSC were associated with reduced recurrence-free survival in patients with NSCLC. Taken together, we identify a novel subset of MDSC within the tumor microenvironment that leads to tumor progression.

为解决倾斜摄影测量生产的实景三维模型集中统一管理难度大、可视化程度低、数据共享困难等问题,本文研究了基于Cesium开源三维地球引擎库的数字高程模型和倾斜实景模型的数据融合处理技术方法,并构建了基于B/S架构的面向倾斜实景模型的三维可视化WebGIS平台,实现了倾斜实景模型与多源空间数据的叠加及动态展示、查询定位、空间量测等功能,可为基于3DGIS的实景三维城市、三维自然资源管理等系统开发提供技术参考。

The Chinese herb Ephedra(also known as Mahuang)has been extensively utilized for the prevention and treatment of coronavirus-induced diseases,including coronavirus disease 2019(COVID-19).However,the specific anti-SARS-CoV-2 compounds and mechanisms have not been fully *** main protease(M~(pro))of SARS-CoV-2 is a highly conserved enzyme responsible for proteolytic processing during the viral life cycle,making it a critical target for the development of antiviral *** study aimed to identify naturally occurring covalent inhibitors of SARS-CoV-2 M~(pro)from Ephedra and to investigate their covalent binding *** results demonstrated that the non-alkaloid fraction of Ephedra(ENA)exhibited a potent inhibitory effect against the SARS-CoV-2 M~(pro)effect,whereas the alkaloid fraction did ***,the chemical constituents in ENA were identified,and the major constituents'anti-SARS-CoV-2 M~(pro)effects were *** the tested constituents,herbacetin(HE)and gallic acid(GA)were found to inhibit SARS-CoV-2 M~(pro)in a time-and dose-dependent *** combination displayed a significant synergistic effect on this key ***,various techniques,including inhibition kinetic assays,chemoproteomic methods,and molecular dynamics simulations,were employed to further elucidate the synergistic anti-M~(pro)mechanisms of the combination of HE and ***,this study deciphers the naturally occurring covalent inhibitors of SARS-CoV-2 M~(pro)from Ephedra and characterizes their synergistic anti-M~(pro)synergistic effect,providing robust evidence to support the anti-coronavirus efficacy of Ephedra.
