Autophagy plays a pivotal role in diverse biological processes,including the maintenance and differentiation of neural stem cells(NSCs).Interestingly,while complete deletion of Fip200 severely impairs NSC maintenance and differentiation,inhibiting canonical autophagy via deletion of core genes,such as Atg5,Atg16l1,and Atg7,or blockade of canonical interactions between FIP200 and ATG13(designated as FIP200-4A mutant or FIP200 KI)does not produce comparable detrimental *** highlights the likely critical involvement of the non-canonical functions of FIP200,the mechanisms of which have remained ***,utilizing genetic mouse models,we demonstrated that FIP200 mediates non-canonical autophagic degradation of p62/sequestome1,primarily via TAX1BP1 in *** deletion of Tax1bp1 in fip200hGFAP conditional knock-in(cKI)mice led to NSC deficiency,resembling the fip200hGFAP conditional knockout(cKO)mouse ***,reintroducing wild-type TAX1BP1 not only restored the maintenance of NSCs derived from tax1bp1-knockout fip200hGFAP cKI mice but also led to a marked reduction in p62 aggregate ***,a TAX1BP1 mutant incapable of binding to FIP200 or NBR1/p62 failed to achieve this ***,conditional deletion of Tax1bp1 in fip200hGFAP cKO mice exacerbated NSC deficiency and p62 aggregate accumulation compared to fip200hGFAP cKO ***,these findings illustrate the essential role of the FIP200-TAX1BP1 axis in mediating the non-canonical autophagic degradation of p62 aggregates towards NSC maintenance and function,presenting novel therapeutic targets for neurodegenerative diseases.

BACKGROUND Chlamydia psittaci(***)is a gram-negative intracellular parasitic pathogenic bacterium that can infect avian and mammalian hosts,including *** detection of *** infections typically relies on traditional antigen-based immunoassays or serological testing that often lack sensitivity and/or *** next generation sequencing(mNGS)is an emerging tool for *** To demonstrate that mNGS represents a valuable tool for rapid,sensitive,and accurate pathogen detection including *** *** Four cases of psittacosis pneumonia and one case of pediatric psittacosis meningitis were diagnosed between December 2019 and May 2020 using mNGS at Changzhou Second People’s Hospital affiliated to Nanjing Medical ***’clinical characteristics,manifestations,and treatment histories were retrospectively *** All five patients had a history of exposure to wild(psittacine or other birds)or domesticated birds(chickens).All patients had a high fever(>39℃)and three of them(60%)experienced organ insufficiency during the *** laboratory data showed normal to slightly increased leucocyte and neutrophil counts,and elevated procalcitonin levels in all five cases,and very high C-reactive protein levels in psittacosis pneumonia *** identified a potential pathogen,***,in patients’bronchoalveolar lavage fluid or cerebrospinal *** tomography revealed lung air-space consolidation,pleural thickening,and effusion fluid buildup in psittacosis pneumonia cases,and an arachnoid cyst in the right temporal lobe of the pediatric psittacosis meningitis *** patients experienced complete recovery following the administration of targeted antichlamydia *** This study not only demonstrated that mNGS represents a valuable tool for rapid,sensitive,and accurate pathogen detection,but also raised public health concerns over *** infections.

A new Multilocus Sequence Analysis Scheme for Mycobacterium tuberculosis

Objective Tuberculosis remains one of the most serious infectious diseases in the world. In this study, a scheme of Mycobacterium tuberculosis （M. tuberculosis） multilocus sequence analysis （MLSA） was established for the phylogenetic and epidemiology analysis. Methods To establish the scheme of M. tuberculosis MLSA, the genome of H37Rv, CCDC5079 and CCDC5180 were compared, and some variable genes were chosen to be the MLSA typing scheme. 44 M. tuberculosis clinical isolates were typed by MLSA, IS6110-RFLP, and soligotyping, to evaluate the MLSA methods. Results After comparison of the genome, seven high discrimination gene loci （recX, rpsL, rmlC, rpmG1, mprA, gcvH, ideR） were chosen to be the MLSA typing scheme finally. 11 variable SNP sites of those seven genes were found among the 44 M. tuberculosis isolate strains and 11 sequence types （STs） were identified. Based on the Hunter-Gaston Index （HGI）, MLSA typing was not as good for discrimination at the strain level as IS6110-RFLP, but the HGI was much better than that of spoligotyping. In addition, the MEGA analysis result of MLSA data was similar to spoligotyping/PGG lineage, showing a strong phylogenetic signal in the modern strains of M. tuberculosis. The MLSA data analysis by eBURST revealed that 4 sequence types （ST） came into a main cluster, showing the major clonal complexes in those 44 strains. Conclusion MLSA genotyping not only can be used for molecular typing, but also is an ideal method for the phylogenetic analysis for M. tuberculosis.

The counterion-mediated hydrogen bonding(CMHB)is related to the hydrogen bonding between bound counterions and polyelectrolyte chains in polyelectrolyte systems,where the counterions can both electrostatically bind to the charged groups of polyelectrolyte chains and act as hydrogen bond donors or acceptors to form hydrogen bonds with the hydrogen bond sites associated with polyelectrolyte chains simultaneously.A large number of literatures illustrate that strong polyelectrolytes(SPs)are insensitive to pH,which severely limmits the applications of SPs as smart ***,our studies have demonstrated that the CMHB makes SPs *** perspective discusses the mechanism of pH responsiveness of SPs and the pH-tunable properties of SPs,based on the pH-controlled CMHB *** future research directions on the pH responsiveness of SPs are also discussed *** is anticipated that the study of the pH responsiveness of SPs not only will provide a new understanding of the fundamental properties of SPs,but also will greatly expand the applications of SPs in the field of smart materials.

Currently,the enhancement in electromagnetic interference(EMI)performance of polymeric composite generally relies on either improving electrical conductivity(σ)for stronger electromagnetic(EM)reflections or tailoring structure for higher EM ***,we proposed a novel technique called cyclic pulsating pressure enhanced segregating structuration(CPP-SS),which can reinforce these two factors *** structural information was supplied by optical microscopy(OM)and scanning electron microscopy(SEM),both of which confirmed the formation and evolution of segregate structured ultra-high molecular weight polyethylene(UHMWPE)/graphene ***,the result showed that CPP-SS can significantly improve theσof ***,advanced specific EMI shielding efficiency of 31.1 d B/mm was achieved for UHMWPE/graphene composite at 1-mm thickness and a low graphene loading of 5 wt%.Meanwhile,it also confirmed that the intrinsic disadvantage of poor mechanical properties of conventional segregated structure composites can be *** work is believed to provide a fundamental understanding of the structural and performance evolutions of segregated structured composites prepared under CPPSS,and to bring us a simple and efficient approach for fabricating high-performance,strong and light-weight polymeric EMI shields.

Detection of H pylori antibody profile in serum by protein array

AIM： To detect multiple H pylori antibodies in serum samples of individuals who carryHpylori by protein array. METHODS： Recombinant H pylori antigens, urease B subunit （UreB）, vacuolating toxin A （VacA） and cytotoxin associated gene A protein （CagA）, were prepared and immobilized in matrixes on nitrocellulose membrane by robotics to bind the specific immunoglobulin G （IgG） antibodies in serum. Staphylococcus protein A （SPA） labeled by colloid gold was used to integrate the immuno-complex and gave red color signal. The scanner based on charge-coupled device （CCD） could collect the image signal and convert it into digital signal. RESULTS： When human IgG was printed on the membrane in increasing concentrations and incubated with immunogold, a linear dose response curve was obtained and the detection limit for IgG was about 0.025 ng. The cutoff values, which were defined as the mean grey level plus 3 times of standard deviation, were 27.183, 28.546 and 27.402, for anti-UreB IgG, anti- CagA IgG and anti-VacA IgG, respectively, as 400 human serum samples with negative H pylori antibodies were detected by the protein array. When 180 serum samples from patients in hospital were employed for detection of IgG against UreB, CagA and VacA, the sensitivity of the protein array was 93.4%, 95.4%, 96.0%, and the specificity was 94.8%, 94.4% and 97.5%, respectively, as compared with the results obtained by ELISA. The assay also showed high reproducibility, uniformity and stability, and the results were available within 30 min. CONCLUSION： The protein array is a very practical method for rapid detection of multiple antibodies in serum samples. It is especially useful for large scale epidemiological investigation of the infection of Hpylori.

m^(6)A methylation is the most frequent modification of mRNA in eukaryotes and plays a crucial role in cancer progression by regulating biological ***-like growth factor 2 mRNA-binding proteins(IGF2BP)are newly identified m^(6)A‘readers’.They belong to a family of RNA-binding proteins,which bind to the m^(6)A sites on different RNA sequences and stabilize them to promote cancer *** this review,we summarize the mechanisms by which different upstream factors regulate IGF2BP in *** current literature analyzed here reveals that the IGF2BP family proteins promote cancer cell proliferation,survival,and chemoresistance,inhibit apoptosis,and are also associated with cancer glycolysis,angiogenesis,and the immune response in the tumor ***,with the discovery of their role as‘readers’of m^(6)A and the characteristic re-expression of IGF2BPs in cancers,it is important to elucidate their mechanism of action in the immunosuppressive tumor *** also describe in detail the regulatory and interaction network of the IGF2BP family in downstream target RNAs and discuss their potential clinical applications as diagnostic and prognostic markers,as well as recent advances in IGF2BP biology and associated therapeutic value.

Research on shell-side heat and mass transfer with multi-component in LNG spiral-wound heat exchanger under sloshing conditions

The spiral-wound heat exchanger(SWHE) is the primary low-temperature heat exchanger for large-scale LNG plants due to its high-pressure resistance, compact structure, and high heat exchange efficiency. This paper studied the shell-side heat and mass transfer characteristics of vapor-liquid two-phase mixed refrigerants in an SWHE by combining a multi-component model in FLUENT software with a customized multicomponent mass transfer model. Besides, the mathematical model under the sloshing condition was obtained through mathematical derivation, and the corresponding UDF code was loaded into FLUENT as the momentum source term. The results under the sloshing conditions were compared with the relevant parameters under the steady-state condition. The shell-side heat and mass transfer characteristics of the SWHE were investigated by adjusting the component ratio and other working conditions. It was found that the sloshing conditions enhance the heat transfer performance and sometimes have insignificant effects. The sloshing condition is beneficial to reduce the flow resistance. The comprehensive performance of multi-component refrigerants has been improved and the improvement is more significant under sloshing conditions, considering both the heat transfer and pressure *** results will provide theoretical support for the research and design of multi-component heat and mass transfer enhancement of LNG SWHE under ocean sloshing conditions.

The low-wavenumber components in the gradient of full waveform inversion(FWI)play a vital role in the stability of the ***,when FWI is implemented in some high frequencies and current models are not far away from the real velocity model,an excessive number of low-wavenumber components in the gradient will also reduce the convergence rate and inversion *** solve this problem,this paper firstly derives a formula of scattering angle weighted gradient in FWI,then proposes a hybrid *** hybrid gradient combines the conventional gradient of FWI with the scattering angle weighted gradient in each inversion frequency band based on an empirical formula derived *** weighted hybrid mode,we can retain some low-wavenumber components in the initial lowfrequency inversion to ensure the stability of the inversion,and use more high-wavenumber components in the high-frequency inversion to improve the convergence *** results of synthetic data experiment demonstrate that compared to the conventional FWI,the FWI based on the proposed hybrid gradient can effectively reduce the low-wavenumber components in the gradient under the premise of ensuring inversion *** also greatly enhances the convergence rate and inversion accuracy,especially in the deep part of the *** the field marine seismic data experiment also illustrates that the FWI based on hybrid gradient(HGFWI)has good stability and adaptability.
