Optical tissue clearing enables rapid,precise and comprehensive assessment of three-dimensional morphology in experimental nerve regeneration research

Morphological analyses are key outcome assessments for nerve regeneration studies but are historically limited to tissue *** optical tissue clearing techniques enabling three-dimensional imaging of entire organs at a subcellular resolution have revolutionized morphological studies of the *** extend their applicability to experimental nerve repair studies we adapted these techniques to nerves and their motor and sensory targets in *** solvent-based protocols rendered harvested peripheral nerves and their target organs transparent within 24 hours while preserving tissue architecture and *** optical clearing was compatible with conventional laboratory techniques,including retrograde labeling studies,and computational image segmentation,providing fast and precise cell ***,optically cleared organs enabled three-dimensional morphometry at an unprecedented scale including dermatome-wide innervation studies,tracing of intramuscular nerve branches or mapping of neurovascular *** their wide-ranging applicability,rapid processing times,and low costs,tissue clearing techniques are likely to be a key technology for next-generation nerve repair *** procedures were approved by the Hospital for Sick children’s Laboratory Animal Services committee(49871/9)on November 9,2019.

changes in feeding selectivity of freshwater invertebrates across a natural thermal gradient

Environmental warming places physiological constraints on organisms, which may be mitigated by their feeding behavior. Theory predicts that consumers should increase their feeding selectivity for more energetically valuable resources in warmer environments to offset the disproportionate increase in metabolic demand relative to ingestion rate. This may also result in a change in feeding strategy or a shift towards a more specialist diet. This study used a natural warming experiment to investigate temperature effects on the feeding selectivity of three freshwater invertebrate grazers： the snail Radix balthica, the blackfly larva Simulium aureum, and the midgefly larva Eukiefferiella minor. chesson＇s Selectivity Index was used to compare the proportional abundance of diatom species in the guts of each invertebrate species with corresponding rock biofilms sampled from streams of different tem- perature. The snails became more selective in warmer streams, choosing high profile epilithic diatoms over other guilds and feeding on a lower diversity of diatom species. The blackfly larvae appeared to switch from active collector gathering of sessile high profile diatoms to more passive filter feeding of motile diatoms in warmer streams. No changes in selectivity were observed for the midgefly larvae, whose diet was representative of resource availability in the environment. These results suggest that key primary consumers in freshwater streams, which constitute a major portion of invertebrate biomass, can change their feeding behavior in warmer waters in a range of different ways. These patterns could potentially lead to fundamental changes in the flow of energy through freshwater food webs.

Unlearning Descartes:Sentient AI is a Political Problem

The emergence of Large Language Models(LLMs)has renewed debate about whether Artificial Intelligence(AI)can be conscious or *** paper identifies two approaches to the topic and argues:(1)A“cartesian”approach treats consciousness,sentience,and personhood as very similar terms,and treats language use as evidence that an entity is *** approach,which has been dominant in AI research,is primarily interested in what consciousness is,and whether an entity possesses it.(2)An alternative“Hobbesian”approach treats consciousness as a sociopolitical issue and is concerned with what the implications are for labeling something sentient or *** both enables a political disambiguation of language,consciousness,and personhood and allows regulation to proceed in the face of intractable problems in deciding if something“really is”sentient.(3)AI systems should not be treated as conscious,for at least two reasons:(a)treating the system as an origin point tends to mask competing interests in creating it,at the expense of the most vulnerable people involved;and(b)it will tend to hinder efforts at holding someone accountable for the behavior of the systems.A major objective of this paper is accordingly to encourage a shift in *** place of the cartesian question-is AI sentient?-I propose that we confront the more Hobbesian one:Does it make sense to regulate developments in which AI systems behave as if they were sentient?

Disruption of bone and skeletal muscle in severe burns

Severe burn injury triggers the body＇s nonspecific adaptive responses to acute insult, including the systemic inflammatory and stress responses, as well as the sympathetic response to immobilization. These responses trigger inflammatory bone resorption followed by glucocorticoid-induced apoptosis of osteoblasts and probably osteocytes. Because these patients are catabolic, they suffer concomitant muscle wasting and negative nitrogen balance. The use of anabolic agents such as recombinant human growth hormone and oxandrolone results in improved bone mineral content and muscle strength after approximately I year. Use of bisphosphonates within the first 10 days of a severe burn completely blocks the resorptive bone loss and has the added advantage of appearing to preserve muscle protein from excessive breakdown. The mechanism for the protective effect on muscle is not currently known. However, if the effect of bisphosphonates on muscle can be confirmed, it raises the possibility that bone communicates with muscle.

Tissue-resident memory T(T_(RM))cells are increasingly associated with the outcomes of health and disease.T_(RM) cells can mediate local immune protection against infections and cancer,which has led to interest in T_(RM) cells as targets for vaccination and ***,these cells have also been implicated in mediating detrimental pro-inflammatory responses in autoimmune skin diseases such as psoriasis,alopecia areata,and ***,we summarize the biology of T_(RM) cells established in animal models and in translational human *** review the beneficial effects of T_(RM) cells in mediating protective responses against infection and cancer and the adverse role of T_(RM) cells in driving pathology in autoimmunity.A further understanding of the breadth and mechanisms of T_(RM) cell activity is essential for the safe design of strategies that manipulate T_(RM) cells,such that protective responses can be enhanced without unwanted tissue damage,and pathogenic T_(RM) cells can be eliminated without losing local immunity.

Physician approaches to drug shortages: Results of a national survey of pediatric hematologist/oncologists

AIM To evaluate personnel involved in scarce drug prioritization and distribution and the criteria used to inform drug distribution during times of shortage among pediatric hematologists/oncologists. METHODS Using the American Society of Pediatric Hematology/Oncology(ASPHO) membership list, a 20 question survey of pediatric hematologists/oncologists was conducted via email to evaluate personnel involved in scarce drug prioritization and distribution and criteria used to inform scarce drug distribution. RESULTS Nearly 65% of the 191 study respondents had patients directly affected by drug shortages. Most physicians find out about shortages from the pharmacist(n = 179, 98%) or other doctors(n = 75, 41%). One third of respondents do not know if there is a program or policy for handling drug shortages at their institution. The pharmacist was the most commonly cited decision maker for shortage drug distribution(n = 128, 70%), followed by physicians(n = 109, 60%). One fourth of respondents did not know who makes decisions about shortage drug distribution at their institution. The highest priority criterion among respondents was use of the shortage drug for curative, rather than palliative intent and lowest priority criterion was order of arrival or first-come *** Despite pediatric hematology/oncology physicians and patients being heavily impacted by drug shortages, institutional processes for handling shortages are lacking. There is significant disparity between how decisions for distribution of shortage drugs are currently made and how study respondents felt those decisions should be made. An institution-based, and more importantly, a societalapproach to drug shortages is necessary to reconcile these disparities.

Synergistic Multilayer Adsorption for Low concentration Dyestuffs by Biomass

Due to the high cost of adsorbents and their thermal regeneration in recent years, much research has fo- cused on the search for cheaper adsorbents for treating wastewater from textile industry. The single component ad-sorption of an acidic dye, Acid Yellow 117, and a basic dye, Methylene Blue, onto several adsoroents-bamooo, waste wood, bamboo char, waste wood char, bamboo activated carbon, wood activated carbon and active carbon F400 were conducted. Based on a Langmuir analysis, the monolayer adsorption capacities were determined. Three of the adsorbents were selected for binary layer adsorption to check the multilayer concept and the potential application for better adsorbent usage. The two cheapest adsorbents, bamboo and wood are compared with the commer-cial activated carbon F400, and all three systems were successful.

On embedding infinite cyclic covers in compact 3-manifolds

Nonalcoholic fatty liver disease （NAFLD）, an pathologies characterized by fatty accumulation in escalating health problem worldwide, covers a spectrum of hepatocytes in early stages, with potential progression to liver inflammation, fibrosis, and failure. A close, yet poorly understood link exists between NAFLD and dyslipidemia, a constellation of abnormalities in plasma lipoproteins including triglyceride-rich very low density lipoproteins. Apolipoproteins are a group of primarily liver-derived proteins found in serum lipoproteins; they not only play an extracellular role in lipid transport between vital organs through circulation, but also play an important intracellu- lar role in hepatic lipoprotein assembly and secretion. The liver functions as the central hub for lipoprotein metab- olism, as it dictates lipoprotein production and to a significant extent modulates lipoprotein clearance. Lipoprotein metabolism is an integral component of hepatocellular lipid homeostasis and is implicated in the pathogenesis, potential diagnosis, and treatment of NAFLD.

In recent times there has been a great deal of interest in the conversion of carbon dioxide into more useful chemical *** the other hand,the translation of these developments in electrochemical reduction of carbon dioxide from the laboratory bench to practical scale remains an underexplored *** we examine some of the major challenges,demonstrating some promising strategies towards such scale-up,including increased electrode area and stacking of electrode pairs in different *** observed that increasing the electrode area from 1 to 10 cm^(2) led to only a 4%drop in current density,with similarly small penalties realised when stacking sub-cells together.
