Functional structure and diversity of soil free-living nematodes in a desert environment depend on plant gender and sampling *** objective of this study was to compare the composition,abundance and tropic group of soil free-living nematodes in the upper 0–10 cm soil layer under the male and female Acanthosicyos horridus *** *** and in the inter-shrub open areas(control)in the Namib Desert,Namibia in April *** moisture,organic matter(Om)and pH was also ***-living nematodes were extracted from 100 g soil using the Baermann funnel procedure,and total number of nematodes was counted under a *** composition and diversity of soil free-living nematodes were analyzed using 18S rDNA *** indicated that a total of 67 groups,including 64 species,2 genera and 1 family were *** behavior of 58 species were identified as follows:15 bacteria-feeding species,12 fungi-feeding species,10 plant-parasite species,5 omnivorous-predator species,8 animal-parasite species,5 invertebrate-parasite species and 3 non-free-living nematodes,known as marine ***,soil free-living nematodes were found to be affected by sampling locations and plant gender,and community composition and density of these nematodes were strongly influenced by soil Om *** confirmed that spatial location and plant cover were main factors influencing the diversity of soil free-living ***,molecular tools were found to be very useful in defining the richness of soil non-free-living *** conclusion,the results elucidated the importance of biotic variables in determining the composition and abundance of soil free-living nematodes in the Namib Desert,Namibia.

Characterising the mechanisms of cell death following focal cerebral ischaemia has been hampered by a lack of an in vitro assay emulating both the apoptotic and necrotic features observed in vivo. The present study systematically characterised oxygen-glucose-deprivation (OGD) in primary rat cortical neurones to establish a reproducible model with components of both cell-death endpoints. OGD induced a time-dependent reduction in cell viability, with 80% cell death occurring 24 h after 3 h exposure to 0% O2 and 0.5 mm glucose. Indicative of a necrotic component to OGD induced cell death, N-methyl-D-aspartate (NmDA) receptor inhibition with mK-801 attenuated neuronal loss by 60%. This study assessed the effects of reducing hydraulic retention times （HRTs） from 21 days to 10.5 days when anaerobically co-digesting pig manure and food waste. Continuously stirred tank reactors of 3.75 L working volume were operated in triplicate at 42℃. Digester HRT was progressively decreased from 21 to 15 days to 10.5 days, with an associated increase in organic loading rate （OLR） from 3.1 kg volatile solids （VS）·m^-***^-1 to 5.1 kg VS·m^***-1 to 7.25 kg VS.m^-3·day^-1. Reducing HRT from 21 days to 15 days caused a decrease in specific methane yields and VS removal rates. Operation at a HRT of 10.5 days initially resulted in the accumulation of isobutyric acid in each reactor. High throughput 16S rRNA gene sequencing revealed that this increase coincided with a shift in acidogenic bacterial populations, which most likely resulted in the increased isobutyric acid concentrations. This may in turn have caused the increase in relative abundance of Clocamonaceae bacteria, which syntrophically degrade non-acetate volatile fatty acids （VFAs） into H2 and CO2. This, along with the increase in abundance of other syntrophic VFA oxidizers, such as Spiorchatetes, suggests that VFA oxidation plays a role in digester operation at low HRTs. Reducing the HRT to below 21 days compromised the ability of the anaerobic digestion system to reduce enteric indicator organism counts below regulatory limits.

The seed coat is important for embryo protection, seed hydration, and dispersal. Seed coat composition is also of interest to the agricultural sector, since it impacts the nutritional value for humans and livestock alike. Although some seed coat genes have been identified, the developmental pathways controlling seed coat development are not completely elucidated, and a global genetic program associated with seed coat development has not been reported. This study uses a combination of genetic and genomic approaches in Arabidopsis thaliana to begin to address these knowledge gaps. Seed coat development is a complex process whereby the integuments of the ovule differentiate into specialized cell types. In Arabidopsis, the outermost layer of cells secretes mucilage into the apoplast and develops a secondary cell wall known as a columella. The layer beneath the epidermis, the palisade, synthesizes a secondary cell wall on its inner tangential side. The innermost layer （the pigmented layer or endothelium） produces proanthocyanidins that condense into tannins and oxidize, giving a brown color to mature seeds. Genetic separation of these cell layers was achieved using the ap2-7 and tt16-1 mutants, where the epidermis/palisade and the endothelium do not develop respectively. This genetic ablation was exploited to examine the developmental programs of these cell types by isolating and collecting seed coats at key tran- sitions during development and performing global gene expression analysis. The data indicate that the developmental programs of the epidermis and the pigmented layer proceed relatively independently. Global expression datasets that can be used for identification of new gene candidates for seed coat development were generated. These dataset provide a comprehensive expression profile for developing seed coats in Arabidopsis, and should provide a useful resource and reference for other seed systems.
