Whole genome phylogeny of Gallus: introgression and data-type effects

Background: Previous phylogenetic studies that include the four recognized species of Gallus have resulted in a number of distinct topologies, with little agreement. Several factors could lead to the failure to converge on a consistent topology, including introgression, incomplete lineage sorting, different data types, or insufficient ***: We generated three novel whole genome assemblies for Gallus species, which we combined with data from the published genomes of Gallus gallus and Bambusicola thoracicus(a member of the sister genus to Gallus). To determine why previous studies have failed to converge on a single topology, we extracted large numbers of orthologous exons, introns, ultra-conserved elements, and conserved non-exonic elements from the genome assemblies. This provided more than 32 million base pairs of data that we used for concatenated maximum likelihood and multispecies coalescent analyses of ***: All of our analyses, regardless of data type, yielded a single, well-supported topology. We found some evidence for ancient introgression involving specific Gallus lineages as well as modest data type effects that had an impact on support and branch length estimates in specific analyses. However, the estimated gene tree spectra for all data types had a relatively good fit to their expectation given the multispecies ***: Overall, our data suggest that conflicts among previous studies probably reflect the use of smaller datasets(both in terms of number of sites and of loci) in those analyses. Our results demonstrate the importance of sampling large numbers of loci, each of which has a sufficient number of sites to provide robust estimates of gene trees. low-coverage whole genome sequencing, as we did here, represents a cost-effective means to generate the very large data sets that include multiple data types that enabled us to obtain a robust estimate of Gallus phylogeny.

Analysis of equilibrium, kinetic, and thermodynamic parameters for biosorption of fluoride from water onto coconut(Cocos nucifera linn.)root developed adsorbent

Drinking water with higher fluoride levels results in serious irremediable health problems that have attained a startle all over the *** focused towards deflouridation through the application of biosorbents prepared from various plants are finding greater scope and *** research is done on Cocos nucifera linn.(coconut tree) one of the very commonly available plants throughout Kerala and around the *** adsorbent developed from the root portion of *** *** used in the present *** study revealed that the fluoride uptake capacity is quite significant and linearly increases with initial adsorbate *** adsorption data is analyzed for langmuir, Freundlich, Temkin, and Dubinin–Radushkevich isotherm models at varying initial adsorbate concentrations(2–25 mg·l^(-1)).It is found that the adsorption of fluoride onto *** *** adsorbent follows langmuir *** isotherm constants "a" and "b" obtained are 2.037 mg·g^(-1) and 0.823 l·mg^(-1) at an adsorbent dose of 8 g·l^(-1) and temperature(26 ± 1) ℃.The mean free sorption energy, E obtained, is 9.13 kJ ·mol^(-1) which points out that the adsorption of fluoride onto *** *** adsorbent is by chemisorption *** kinetic study also supports chemisorption with adsorption data fitting well with a pseudo-second-order kinetic model with an estimated rate constant K_2 of 0.2935 g·mg^(-1) min at an equilibrium contact time of 90 *** thermodynamic study indicated the spontaneous and endothermic nature(ΔH =12.728 kJ·mol^(-1)) of fluoride adsorption onto the *** *** *** Electron Microscopy(SEM), BET, FTIR, and EDX methods were used to analyze the surface morphology of adsorbent before and after fluoride adsorption *** on deflouridation using *** *** adsorbent application on fluoride contaminated ground water samples from fields showed encouraging results.

Ethnobotanical database based screening and identification of potential plant species with antiplasmodial activity against chloroquine-sensitive(3D7) strain of Plasmodium falciparum

Objective: To evaluate the antiplasmodial activity of aqueous-methanolic plant extracts of nine plant species selected, based on ethnobotanical data. Methods: Based on ethnobotanical database, the selected plants were tested for their antiplasmodial activity against chloroquinesensitive(3 D7) strain of Plasmodium falciparum. Qualitative tests and high performance thin layer chromatography analysis were carried out to explore the phytocomponents present in the plant extracts. 1,1-diphenyl-2-picrylhydrazyl antioxidant activity was also determined to check the antioxidant activity of the plant extracts. Results: Moringa oleifera(IC_(50): 3.906 μg/ml),Acalypha indica(IC_(50): 3.906 μg/ml), Hyptis suaveolens(IC_(50): 3.906 μg/ml), Mangifera indica(IC_(50): 4.150 μg/ml) and Averrhoa bilimbi(IC_(50): 4.881 μg/ml) showed very good antiplasmodial activity. Conclusions: Crude extracts of Mangifera indica and Hyptis suaveolens demonstrated the most efficacious antimalarial activity. A bioassay-guided fractionation of these extracts to identify the lead compound is proved to be useful. The results validate the traditional use of the selected plants as antimalarials.

Compared with bare-metal stents (BMS),drug-eluting stems (DES)have shown better clinical outcomes for pa- tients undergoing percutaneous coronary intervention (PCI) by inhibition of neo-intirnal hyperplasia.[1]However,early- generation DES produced late thrombotic events,more than l-year,by delaying arterial healing of stented vessels,[2-5] New-generation DES have been developed'with thinner stent struts,more biocompatible polymer coatings for drug release,and a variety of antiproliferative agents with similar or superior anti-restenotic efficacy.[6]This development has led to a significant improvement in the efficacy and safety of new-generation DES,and consistently lower rates of very late stent thrombosis (VlST).[7,8]In fact,use of new-ge- neration DES is the standard treatment in contemporary PCI practice.[9]

No evidence demonstrating hepatotoxicity associated with hydroxycitric acid

Although a number of cases of hepatotoxicity are associated with the use of Hydroxycut weight management products, it has been alleged that their effects are primarily due to the presence of hydroxycitric acid (HCA, as Super CitriMax) in the formulations. However, while these products contain up to 20 different ingredients, some do not contain HCA. Case studies reported to date have not considered in depth the literature on the numerous animal and human studies that have been conducted on the safety and effi cacy of HCA. No HCA-associated hepatotoxicity or treatment-related adverse effects have been reported in these studies, and thus it is premature to make the assumptions presented in the recent case studies regarding Hydroxycut. If it is established in well controlled studies that the use of these formulations with and/or without HCA can result in the occurrence or progression of hepatotoxicity, additional studies should be conducted to characterize the causative factor(s).

Early ileocolonoscopy with biopsy for the evaluation of persistent post-transplantation diarrhea

AIM: To investigate the signifi cance of ileocolonoscopy with histology in the evaluation of post-transplantation persistent diarrhea (PD). METHODS: We retrospectively reviewed all records of renal transplant patients with PD, over a 3-year period. All patients were referred for ileocolonoscopy with biopsy, following a negative initial diagnostic work up. Clinical and epidemiological data were compared between cases with infectious or drug-induced diarrhea. RESUlTS: We identif ied 30 episodes of PD in 23 renaltransplant patients (1-3 cases per patient). There were 16 male patients and the mean age at the time of PD was 51.4 years. The average time from transplantation to a PD episode was 62.3 ± 53.2 mo (range 1-199 mo). Ileocolonoscopy detected mucosal abnormalities in 19 cases, whereas the intestinal mucosa appeared normal in 11 cases. Histological examination achieved a specific diagnosis in 19/30 cases (63.3%). In nine out of 11 cases (82%) with normal endoscopic appearance of the mucosa, histological examination of blinded biopsies provided a specif ic diagnosis. The etiology of PD was infectious in 11 cases (36.6%), drug-related in 10 (33.3%), of other causes in three (10%), and of unknown origin in six cases (20%). Infectious diarrhea occurred in significantly longer intervals from transplantation compared to drug-related PD (85.5 ± 47.6 mo vs 40.5 ± 44.8 mo, P ly, PD due to drug-toxicity was rarely seen after the f irst year post-transplantation. Clinical improvement followed therapeutic intervention in 90% of cases. Modif ication of immunosuppressive regimen was avoided in 57% of patients. CONClUSION: Early ileocolonoscopy with biopsies from both affected and normal mucosa is an important adjunctive tool for the etiological diagnosis of PD in renal transplant patients.

Predicting lower limb periprosthetic joint infections: A review of risk factors and their classification

AIM To undertook a systematic review to determine factors that increase a patient's risk of developing lower limb periprosthetic joint infections(PJI).METHODS This systematic review included full-text studies that reviewed risk factors of developing either a hip or knee PJI following a primary arthroplasty published from January 1998 to November 2016. A variety of keywords were used to identify studies through international databases referencing hip arthroplasty, knee arthroplasty, infection, and risk factors. Studies were only included if they included greater than 20 patients in their study cohort, and there was clear documentation of the statistical parameter used; specifically P-value, hazard ratio, relative risk, or/and odds ratio(OR). Furthermore a quality assessment criteria for the individual studies was undertaken to evaluate the presence of record and reporting bias. RESUlTS Twenty-seven original studies reviewing risk factors relating to primary total hip and knee arthroplasty infections were included. Four studies(14.8%) reviewed PJI of the hip, 3(11.21%) of the knee, and 20(74.1%) reviewed both joints. Nineteen studies(70.4%) were retrospective and 8(29.6%) prospective. Record bias was identified in the majority of studies(66.7%). The definition of PJI varied amongst the studies but there was a general consensus to define infection by previously validated methods. The most significant risks were the use of preoperative high dose steroids(OR = 21.0, 95%CI: 3.5-127.2, P low 20(OR = 6.00, 95%CI: 1.2-30.9, P = 0.033), diabetes(OR = 5.47, 95%CI: 1.77-16.97, P = 0.003), and coronary artery disease(OR = 5.10, 95%CI: 1.3-19.8, P = 0.017).CONClUSION We have highlighted the need for the provider to optimise modifiable risk factors, and develop strategies to limit the impact of non-modifiable factors.

Interferon-free regimens for the treatment of hepatitis C virus in liver transplant candidates or recipients

The goal of therapy in chronic hepatitis C virus(HCV) infection is sustained virological response(SVR) which reflects HCV eradication. Treatment against HCV has dramatically improved with the recent availability of direct-acting antivirals(DAAs) including sofosbuvir, simeprevir, daclatasvir, ledipasvir/sofosbuvir, paritaprevir/ombitasvir and dasabuvir. Carefully selected combinations of these DAAs offer the potential for highly effective all-oral safe regimens even for patients with decompensated cirrhosis or liver transplant(lT) recipients. like all current protease inhibitors, simeprevir and paritaprevir should not be used in patients with Child C cirrhosis, while sofosbuvir and ledipasvir/sofosbuvir should not be given in patients with severe renal impairment and glomerular filtration rate less than 30 m l/min. Drug-drug interactions may still occur with the current DAAs particularly in postlT patients, in whom simeprevir should not be coadministered with cyclosporine and dose adjustments of calcineurin inhibitors are required in case of regimens including the ritonavir boosted paritaprevir. Phase Ⅱ clinical trials and real life cohort studies have shown that sofosbuvir based combinations are safe and can achieve improvements of clinical status, high SVR rates and even prevention of post-lT HCV recurrence in patients with decompensated cirrhosis or lT-candidates. In the post-lT setting, sofosbuvir based regimens and the combination of paritaprevir/ombitasvir and dasabuvir have been reported to be safe and achieve high SVR rates, similar to those in non-transplantpatients, being effective even in cases with cholestatic fibrosing hepatitis. Ongoing clinical trials and rapidly emerging real life data will further clarify the safety and efficacy of the new regimens in these settings.
