Effect of adipose-derived mesenchymal stem cells on hepatocellular carcinoma: In vitro inhibition of carcinogenesis

AIM To investigate the effect of adipose-derived mesenchymal stem cells(ADMSCs)and their conditioned media(CM) on hepatocellular carcinoma(HCC) cell *** The proliferation rate of HepG2 and PlC-PRF-5 HCC cancer cells was measured using the trypan blue exclusion method and confirmed using the cell-counting kit8(commonly known as CCK-8) assay. Apoptosis was detected by flow cytometry using annexin V-FITC. Protein and mRNA expression was quantified by ElISA and real time PCR, respectively. Migration and invasion rates were performed by Transwell migration and invasion assays. Wound healing was examined to confirm the data obtained from the migration *** Our data demonstrated that when co-culturing HCC cell lines with ADMSCs or treating them with ADMSC CM, the HCC cell proliferation rate was significantly inhibited and the apoptosis rate increased. The decreased proliferation rate was accompanied by an upregulation of P53 and Retinoblastoma mRNA and a downregulation of c-Myc and hTERT mRNA levels. More notably, ADMSCs and their CM suppressed the expression of the two important markers of HCC carcinogenicity, alpha-fetoprotein and Des-gamma-carboxyprothrombin. In addition, the migration and invasion levels of HepG2 and PlC-PRF-5 cells significantly decreased, potentially through increased expression of the tissue inhibitor metalloproteinases TIMP-1, TIMP-2 and *** These findings shed new light on a protective and therapeutic role for ADMSCs and their CM in controlling HCC invasiveness and carcinogenesis.

A Model Study on Accelerated Consolidation of Coir Reinforced lateritic lithomarge Soil Blends with Vertical Sand Drains for Pavement Foundations

Sub-grade soils of lateritic origin encountered in the construction of highway embankments in various regions of India, often comprise intrusions of soft lithomargic soils that result in large settlements during constructions, and differential settlements at later stages. This necessitates the use of appropriate soil improvement techniques to improve the load-carrying capacity of pavements. This work deals with accelerated consolidation of un-reinforced and coir-rein- forced lateritic lithomargic soil blends, provided with three vertical sand drains. The load-settlement characteristics were studied for various preloads ranging from 50 kg (0.0013 N/mm2) to 500 kg (0.013N/mm2) on soil specimens prepared in circular ferro-cement moulds. It was observed that at lower preloads up to 200kg, across the blends, the relative increase in consolidation (Rct) for randomly reinforced soil with vertical drains was sig-nificantly higher than that of un-reinforced soil without vertical drains, with an average value of 124.8%. Also, the Rct for un-reinforced soil with vertical drains was quite higher than that of un-reinforced soil without vertical drains, with an average value of 103.9%. In the case of higher preloads, the Rct values for randomly reinforced soil with vertical drains were moderate with an average value of 30.88%, while the same for un-reinforced soil with vertical drains was about 20.4%. The aspect-ratio of coir fibers used was 1:275.

Plants have a perception system triggered by pathogen and pest signals to initiate *** signals include evolutionarily conserved molecules from microbes and insects termed pathogen/herbivore-associated molecular patterns(PAMPs/HAMPs).Here we showed that hexaacetyl-chitohexaose(HC),an oligosaccharide from chitin,a structural component in insect exoskeletons and fungi cell walls,upregulated defense-associated genes WRKY22,GST1,RAR1,EDS1,PAl1 and NPR2,and downregulated ICS1 at 1 h after HC treatment in Sun Chu Sha mandarin *** effect was transient as defense gene transcriptional changes were not observed at 18 h after the *** penetration graph(EPG)recordings were used to study the feeding behavior of Asian citrus psyllid(ACP)following the HC *** is the hemipteran vector of Candidatus liberibacter asiaticus(Clas),the pathogen associated with huanglongbing(HlB).Adult ACP displayed reduced intercellular probing,reduced xylem feeding count and duration,and increased non-probing activity on HC-treated citrus compared to *** an 18-h recording,percentage for total duration of xylem ingestion,phloem ingestion,intercellular probing were lower,and the percentage of nonprobing behavior was higher in HC-treated leaves than in *** host-selection behavior studies,HC treatment did not alter the attractiveness of citrus leaves under light or dark *** addition,ACP feeding on HC-treated leaves did not show differences in mortality for up to 10 day of *** summary,we report that HC induced a transient defense in citrus and an inhibitory effect on ACP feeding but did not affect host selection or the insect fitness under the tested conditions.

S-Nitrosothiols or thionitrites with the general formula RSNO are formally composed of the nitrosyl cation(NOt) and a thiolate(RSà), the base of the corresponding acids RSH. The smallest S-nitrosothiol is HSNO and derives from hydrogen sulfide(HSH, H_2S). The most common physiological S-nitrosothiols are derived from the amino acid l-cysteine(Cys SH). Thus, the simplest S-nitrosothiol is S-nitroso-l-cysteine(Cys SNO). Cys SNO is a spontaneous potent donor of nitric oxide(NO) which activates soluble guanylyl cyclase to form cyclic guanosine monophosphate(c GMP). This activation is associated with multiple biological actions that include relaxation of smooth muscle cells and inhibition of platelet *** NO, Cys SNO is a short-lived species and occurs physiologically at concentrations around 1 n M in human blood. Cys SNO can be formed from Cys SH and higher oxides of NO including nitrous acid(HONO)and its anhydride(N_2O_3). The most characteristic feature of RSNO is the S-transnitrosation reaction by which the NOtgroup is reversibly transferred to another thiolate. By this way numerous RSNO can be formed such as the low-molecular-mass S-nitroso-N-acetyl-l-cysteine(SNAC) and S-nitroso-glutathione(GSNO), and the high-molecular-mass S-nitrosol-l-cysteine hemoglobin(Hb Cys SNO) present in erythrocytes and S-nitrosol-l-cysteine albumin(Alb Cys SNO) present in plasma at concentrations of the order of 200 n M. All above mentioned RSNO exert NO-related biological activity, but they must be administered intravenously. This important drawback can be overcome by lipophilic charge-free ***, we prepared the ethyl ester of SNAC, the S-nitroso-N-acetyl-l-cysteine ethyl ester(SNACET), from synthetic N-acetyl-l-cysteine ethyl ester(NACET). Intracranial Hemangiopericytoma Focally Recurrent to the Pelvis

A 42-year-old male with a history of recurrent intracranial hemangiopericytoma presented to the Emergency Department with urinary retention. Ensuing work-up revealed a 10-centimeter circumscribed pelvic mass that appeared anatomically distinct from the sacrum. This metastasis was found 10 years after the initial resection of his intracranial lesion. An abdominal resection was performed, and the patient recovered uneventfully. Final pathology and staining findings were consistent with metastatic hemangiopericytoma. Hemangiopericytoma is a rare mesenchymal soft tissue neoplasm with potential for late local recurrence and metastasis. Here, we review the case and discuss the clinical relevance of narrowing the diagnosis to HPC or the growing umbrella of Solitary Fibrous Tumor (SFT). To our knowledge, this is the first report of intracranial HPC metastatic to the pelvis.
