the pancreas became one of the first objects of regenerative medicine,since other possibilities of dealing with the pancreatic endocrine insufficiency were clearly *** number of people living with diabetes mellitus is currently approaching half a billion,hence the crucial relevance of new methods to stimulate regeneration of the insulin-secretingβ-cells of the islets of *** restrictions on the islet regeneration are very tight;nevertheless,the islets are capable of physiological regeneration viaβ-cell self-replication,direct differentiation of multipotent progenitor cells and spontaneousα-toβ-orδ-toβ-cell conversion(trans-differentiation).the existing preclinical models ofβ-cell dysfunction or ablation(induced surgically,chemically or genetically)have significantly expanded our understanding of reparative regeneration of the islets and possible ways of its *** ultimate goal,sufficient level of functional activity ofβ-cells or their substitutes can be achieved by two prospective broad strategies:β-cell replacement andβ-cell ***“regeneration”strategy aims to maintain a preserved population ofβ-cells through in situ exposure to biologically active substances that improveβ-cell survival,replication and insulin secretion,or to evoke the intrinsic adaptive mechanisms triggering the spontaneous non-β-toβ-cell ***“replacement”strategy implies transplantation ofβ-cells(as non-disintegrated pancreatic material or isolated donor islets)orβ-like cells obtained ex vivo from progenitors or mature somatic cells(for example,hepatocytes orα-cells)under the action of small-molecule inducers or by genetic *** believe that the huge volume of experimental and clinical studies will finally allow a safe and effective solution to a seemingly simple goal-restoration of the functionally activeβ-cells,the innermost hope of millions of people globally.

Glucose transporters expression in the placental terminal villi of preeclampsia and intrauterine growth retardation complicated pregnancies

Introduction: transmembrane sodium-independent glucose transporters (GLUts) play an important role in both placental and fetal development by the providing transplacental glucose transport. Abnormal GLUt-1 and GLUt-3 placental expression could play a role in such late pregnancy complications development as preeclampsia and intrauterine growth retardation. Materials and Methods: Immunohistochemistry was performed to reveal patterns of GLUt-1 and GLUt-3 expression in the placental terminal villi compartments. Results: GLUt-1 syncytial expression in the terminal villi of severe PE cases (both with and without IUGR) was significantly lower in compare to control group. We also report about GLUt-3 both syncytial and endothelial expression in the near term and term placental terminal villi without significant difference between the study and control groups. Conclusion: other study should be performed to establish the nature of GLUt-1 downregulation in severe preeclampsia cases and whether GLUt-3 is expressed in the syncytiotrophoblast of near term and term placentas or not.

Multipotent stromal cells stimulate liver regeneration by influencing the macrophage polarization in rat

AIM to investigate the influence of the umbilical cordderived multipotent stromal cells(MSCs) on recovery of the liver after the subtotal resection, that is, removal of 80% of the organ mass, a renowned model of the small-for-size liver remnant syndrome. MEtHODS the MSCs were obtained from the intervascular tissue of umbilical cords, dissected from rat fetuses, by the explant culture technique. the vital labeling of MSCs with РКН26 was carried out on the 3 rd passage. the subtotal resection was performed on male Sprague-Dawley rats. the experimental group animals received a transplant 106 MSCs infused into the spleen. Hepatocyte proliferation was assessed by counting of either mitotic figures or Ki67-positive cells in microscopic images. MSC differentiation was assessed with antibodies to hepatocyte-specific marker cytokeratin 18(CK18), cholangiocyte-specific protein CK19, smooth muscle cell-specific protein α-SMA, the endothelial cell marker CD31, or the active fibroblast marker FAPα. total macrophages of the liver were selectively stained in cryosections incubated with antiCD68 antibodies(1:100, Abcam), while the M2 a and M2 c macrophage populations were selectively stained with anti-CD206 antibodies. Expression of interleukin and growth factor genes was evaluated with *** Intrasplenic allogeneic transplantation of the umbilical cord-derived multipotent stromal cells stimulates reparative processes within the residual liver tissue after subtotal resection(removal of 80% of the organ mass), as indicated by increased rates of hepatocyte proliferation and accelerated organ mass recovery. these effects may result from paracrine influence of the transplanted cells on the resident macrophage population of the liver. the transplantation favors polarization of macrophages to M2 phenotype(the M2-polarized macrophages specifically express CD206; they are known to suppress inflammation and support tissue repair). the Study on the Cycloids of Moving Loops

the infinite set of cycloids is created. Each cycloid of this set is defined as a movement trajectory of a point when this point circulates on the convex closed contour of arbitrary form when this contour moves rectilinearly without rotation on the plane with a velocity equal to the tangential velocity of a point on circulation contour. the classical cycloid is elements of this set. the differential equation of a cycloid set is derived and its solution in quadratures is received. the inverse problem when for the given cycloid it is necessary to fine the form of a circulation contour is solved. the problem of differential equation of the second order with boundary conditions about a bend of big curvature of an elastic rod of infinite length is solved in quadratures. Geometry of the loop which is formed at such bend is investigated. It is discovered that at movement of an elastic loop on a rod when the form and the size of a loop don’t change, each point of a loop moves on a trajectory which named by us the cycloid and which represents a circumference arch.

Concerning spikes in emission and absorption in the microwave range

In some events, weak fast solar bursts （near the level of the quiet Sun） were observed in the background of numerous spikes in emission and absorption. In such a case, the background contains the noise signals of the receiver. In events on 2005 September 16 and 2002 April 14, the solar origin of fast bursts was confirmed by simultaneous recording of the bursts at several remote observatories. the noisy background pixels in emission and absorption can be excluded by subtracting a higher level of continuum when constructing the spectra. the wavelet spectrum, noisy pro- files in different polarization channels and a spectrum with continuum level greater than zero demonstrates the noisy character of pixels with the lowest levels of emission and absorption. thus, in each case, in order to judge the solar origin of all spikes, it is necessary to determine the level of continuum against the background of which the solar bursts are observed. Several models of microwave spikes are discussed. the electron cyclotron maser emission mechanism runs into serious problems with the in- terpretation of microwave millisecond spikes： the main obstacles are too high values of the magnetic field strength in the source （Pe 〈 uB）. the probable mechanism is the interaction of plasma Langmuir waves with ion-sound waves （l ＋ s → t） in a source related to shock fronts in the reconnection region.

the measurement of positions and sizes of radio sources in observations is important for un- derstanding of the flare evolution. For the first time, solar radio spectral fine structures in an M6.5 flare that occurred on 2013 April 11 were observed simultaneously by several radio instruments at four different observatories： Chinese Solar Broadband Radio Spectrometer at Huairou （SBRS/Huairou）, Ondrejov Radio Spectrograph in the Czech Republic （ORSC/Ondrejov）, Badary Broadband Microwave Spectropolarimeter （BMS/Irkutsk）, and spectrograph/IZMIRAN （Moscow, troitsk）. the fine structures included microwave zebra patterns （ZPs）, fast pulsations and fiber bursts. they were observed during the flare brightening lo- cated at the tops of a loop arcade as shown in images taken by the extreme ultraviolet （EUV） telescope onboard NASA＇s satellite Solar Dynamics Observatory （SDO）. the flare occurred at 06：58-07：26 Ut in solar active region NOAA 11719 located close to the solar disk center. ZPs appeared near high frequency boundaries of the pulsations, and their spectra observed in Huairou and Ondrejov agreed with each other in terms of details. At the beginning of the flare＇s impulsive phase, a strong narrowband ZP burst occurred with a moderate left-handed circular polarization. then a series of pulsations and ZPs were observed in almost unpolarized emission. After 07：00 Ut a ZP appeared with a moderate right-handed polarization. In the flare decay phase （at about 07：25 Ut）, ZPs and fiber bursts become strongly right-hand polarized. BMS/Irkutsk spectral observations indicated that the background emission showed a left-handed circular polarization （similar to SBRS/Huairou spectra around 3 GHz）. However, the fine structure appeared in the right-handed polarization. the dynamics of the polarization was associated with the motion of the flare ex- citer, which was observed in EUV images at 171 A and 131 A by the SDO Atmospheric Imaging Assembly （AIA）. transposable element insertions(tEIs) are an important source of genomic innovation by contributing to plant adaptation, speciation, and the production of new varieties. the often large,complex plant genomes make identifying tEIs from short reads difficult and expensive. Moreover, rare somatic insertions that refect mobilome dynamics are difficult to track using short *** address these challenges, we combined Cas9-targeted Nanopore sequencing(CANS) with the novel pipeline Nano Cas tE to trace both genetically inherited and somatic tEIs in plants. We performed CANS of the EVADé(EVD) retrotransposon in wild-type Arabidopsis thaliana and rapidly obtained up to 40× sequence *** of hemizygous t-DNA insertion sites and genetically inherited insertions of the EVD transposon in the ddm1(decrease in DNA methylation1) genome uncovered the crucial role of DNA methylation in shaping EVD insertion *** also investigated somatic transposition events of the ONSEN transposon family, finding that genes that are downregulated during heat stress are preferentially targeted by ONSENs. Finally, we detected hypomethylation of novel somatic insertions for two ONSENs. CANS and Nano Cas tE are effective tools for detecting tEIs and exploring mobilome organization in plants in response to stress and in different genetic backgrounds, as well as screening t-DNA insertion mutants and transgenic plants.
