Contr.lled gene r.gulation dur.ng gamete development is vital for.maintaining r.pr.ductive potential. Dur.ng the complex pr.cess of mammalian sper.atogenesis, male ger. cells exper.ence extended per.ods of the inactive tr.nscr.ption despite heavy tr.nslational r.quir.ments for.continued gr.wth and differ.ntiation. Hence, sper.atogenesis is highly r.liant on mechanisms of posttr.nscr.ptional r.gulation of gene expr.ssion, facilitated by r.A binding pr.teins （r.Ps）, which r.main abundantly expr.ssed thr.ughout this pr.cess. One such gr.up of pr.teins is the Musashi family, pr.viously identified as cr.tical r.gulator. of testis ger. cell development and meiosis in Dr.sophila, and also shown to be vital to sper. development and r.pr.ductive potential in the mouse. This r.view descr.bes the r.le and function of r.Ps our.r.cent knowledge of the Musashi pr.teins in sper.atogenesis. within the scope of male ger. cell development, focusing on The functional mechanisms utilized by r.Ps within the cell ar. outlined in depth, and the significance of sub-cellular.localization and stage-specific expr.ssion in r.lation to the mode and impact of posttr.nscr.ptional r.gulation is also highlighted. We emphasize the histor.cal r.le of the Musashi family of r.Ps in stem cell function and cell fate deter.ination, as or.ginally char.cter.zed in Dr.sophila and Xenopus, and conclude with our.cur.ent under.tanding of the differ.ntial r.les and functions of the mammalian Musashi pr.teins, Musashi-1 and Musashi-2, with a pr.mar. focus on our.findings in sper.atogenesis. This r.view highlights both the essential contr.bution of r.Ps to posttr.nscr.ptional r.gulation and the impor.ance of the Musashi family as master.r.gulator. of male gamete development.
