In vitro antioxidant activities of selected seaweeds from Southeast coast of India

Objective:In vitro antioxidant activities of three selected Indian seaweeds viz.,Halimeda tuna(***),Turbinaria conoides(***) and Gracilaria foliifera(***) were ***:Total antioxidant activity,total phenolic content,and reducing power of crude methanol and diethyl ether extracts were ***:Total phenolic content and total antioxidant activity were higher(1.231±0.173 mg GAE/g,1.675±0.361 mg GAE/g) in *** *** power of crude methanol extract increased with concentrations of the *** Fourier transform-infra red spectrum analysis revealed the presence of polyphenolic *** seaweed extracts displayed moderate antioxidant activity compared to gallic acid ***:The seaweeds could be considered for curing diseases from oxidative deteriorations.

AIMS: Sulfated polysaccharide extracted from the brown algae Sargassum swartzii was studied for antioxidant potential. METHODS: The extracted sulfated polysaccharide was analyzed for physico-chemical characteristics, TAC, reducing power, free radical scavenging potentials (DPPH, ABTS, H2O2 radical) and antibacterial properties. RESULTS: The extract showed a high percentage of carbohydrate (7.40 ± 0.63) %, followed by sulfate (5.3 ± 1.54) %. The highest antioxidant activity was observed in ABTS (55 ± 3.61) %, followed by H2O2 (47.23 ± 2.81) % and DPPH (25.33 ± 2.52) %; significant differences were observed at (P ≥ 0.05). Among the ten human pathogenic strains tested, E. coli was the more sensitive. The characterization and mobility of the sulfated polysaccharide was examined by the FT-IR spectrum and assayed by agarose gel electrophoresis which showed highest mobility at higher pH buffer in carbonate-bicarbonate (pH 10) buffer. The molecular weight of the sulfated polysaccharide was determined by gradient PAGE and was found to be 50 KDa. Finally, GC-MS analysis revealed the presence of peaks corresponding to dimethyl-4-nitroaniline (26.34%). CONCLUSIONS: It is suggested that the sulfated polysaccharide from Sargassum swartzii could be a better source of natural antioxidant, as well as an antibacterial agent.

The effects of the dried macroalga, Gracilaria tenuistipitata （Rhodophyta）, on photosynthesis of a highly stress-resistant microalga, ***., were studied. Pyramimonas sp. was cultured with dried G. lemaneiformis under controlled laboratory conditions. The chlorophyll a （Chl a） fluorescence transient O-J-I-P curve coupled with its specific parameters was established. The Chl a fluorescence transients were recorded in vivo at high time resolution and analyzed according to the JIP-test which can quantify photosystem II activity. A clear dose-dependent relationship was observed between the dried biomass of G. tenuistipitata and its inhibitory effect on Pyramimonas sp. A decrease in the O-J-I-P curve expressed as Chl a fluorescence intensity along with its specific parameters were observed, which was also time-dependent. The main photosynthetic inhibitory targets of the macroalga on the microalga, according to the JIP-test, can be expressed as a decrease in the number of active reaction centers and the blocking of the electron transport chain. The results of the present study suggest that dried fragments of G. tenuistipitata effectively inhibit photosynthesis in Pyramimonas sp. and could thus be potential candidates for use in the control and mitigation of harmful algal blooms.
