Fatty Acid Binding Protein-4 Silencing Attenuates Atherosclerosis Progression by Affecting Macrophage Apoptosis and Autophagy

Background: Atherosclerosis, which is the principal cause of heart attacks and strokes, is the leading cause of death in the United States. Various biological processes such as apoptosis and autophagy are involved in almost every stage of atherosclerosis, which could lead to plaque instability causing stroke and death. Fatty Acid Binding Protein 4 (FABP4) is an adipokine released by macrophages and is involved in multiple disease conditions including stroke. However, the association of FABP4 with macrophage apoptosis and autophagy in atherosclerosis has not been elucidated. We hypothesize that silencing FABP4 protein could be a novel therapeutic approach to attenuate macrophage apoptosis and autophagy thereby minimizing plaque instability in atherosclerosis. Methods: RAW264 mouse macrophage cells were transfected with siRNA control liposome quantum dots (QD), siFABP4 liposome QD at the concentration of 150 μg/ml total lipids, or TNF-α at 100 ng/ml. Western blot and reverse phase protein array (RPPA) analysis were completed. Results: Inhibiting the translation of FABP4 blocked the apoptotic pathway as evidenced from the increased expression of anti-apoptotic BCl-xl and BCl-2 along with reduced expression of BAX and activated Caspase 3 levels. Beclin-1 and lC3-II levels were also reduced with knocking down FABP4 indicating the attenuation of autophagy. Conclusion: Targeting FABP4 protects against macrophage processes associated with the progression of atherosclerosis.

Foliar application of rare earth elements on soybean(Glycine max(l)):Effects on biometrics and characterization of phytotoxicity

This study aims at investigating the effects of foliar application of aqueous solutions of la3+and Ce3+nitrates on soybean plants(Glycine max).First,we observe that Triton HW 1000 surfactant at 0.01 vol%reduces droplets contact angle and increases their drying *** greenhouse conditions,the foliar treatments do not affect chlorophyll content,plant height,number of leaves,number of pods,number of seeds per pod,and average seed ***,the treatments induce phytotoxicity since foliar injuries appear after the *** X-ray fluorescence spectroscopy combined to scanning electron microscopy shows that the leaf lesions are positively correlated to accumulation of Ce and la on the leaf surface and also promote structural alteration to the epidermal cells.X-ray absorption near edge structure shows that the la and Ce nitrates are partially bio transformed into oxides by the leaves which might explain the harmful effects.

Kabbalah logic and Semantic Foundations for a Postmodern Fuzzy Set and Fuzzy logic Theory

Despite half a century of fuzzy sets and fuzzy logic progress, as fuzzy sets address complex and uncertain information through the lens of human knowledge and subjectivity, more progress is needed in the semantics of fuzzy sets and in exploring the multi-modal aspect of fuzzy logic due to the different cognitive, emotional and behavioral angles of assessing truth. We lay here the foundations of a postmodern fuzzy set and fuzzy logic theory addressing these issues by deconstructing fuzzy truth values and fuzzy set membership functions to re-capture the human knowledge and subjectivity structure in membership function evaluations. We formulate a fractal multi-modal logic of Kabbalah which integrates the cognitive, emotional and behavioral levels of humanistic systems into epistemic and modal, deontic and doxastic and dynamic multi-modal logic. This is done by creating a fractal multi-modal Kabbalah possible worlds semantic frame of Kripke model type. The Kabbalah possible worlds semantic frame integrates together both the multi-modal logic aspects and their Kripke possible worlds model. We will not focus here on modal operators and axiom sets. We constructively define a fractal multi-modal Kabbalistic l-fuzzy set as the central concept of the postmodern fuzzy set theory based on Kabbalah logic and semantics.

Distribution of atrial cardiomyopathy markers and association with atrial fibrillation detected after ischaemic stroke in the SAFAS study

Background Atrial cardiomyopathy(AC)is an emerging concept explaining the pathophysiology of cardioembolic strokes in absence of atrial fibrillation(AF).A definition based on the presence of electrical abnormality(P-wave terminal force in lead V1(PTFV1)>5000µV×ms),N-Terminal pro-B-type natriuretic peptide(NT pro BNP)>250pg/ml and/or indexed left atrial diameter(lADI)>3cm/m^(2)is currently tested in the ARCADIA(AtRial Cardiopathy and Antithrombotic Drugs In prevention After cryptogenic stroke)*** set out to estimate the prevalence of AC as defined in the ARCADIA trial,its determinants and its association with AF detected after stroke(AFDAS).Methods Stepwise screening for silent Atrial Fibrillation After Stroke(SAFAS)study prospectively included 240 ischaemic stroke *** markers were complete for 192 of them and 9 were not included in this analysis because AF had been diagnosed on *** A total of 183 patients were analysed,of whom 57%(104 patients)met the AC criteria(79 NT-proBNP,47 PTFV1,4 lADI).In the multivariate logistic regression,C reactive protein>3mg/l(OR(95%CI)2.60(1.30 to 5.21),p=0.007)and age(OR(95%CI)1.07(1.04 to 1.10),ply associated with *** 6 months of follow-up,AFDAS was detected in 33%of AC patients and in 14%of the remaining ones(p=0.003).However,AC was not independently associated with AFDAS,contrary to left atrial volume index(>34ml/m^(2),OR 2.35(CI 1.09 to 5.06)p=0029).Conclusion AC as defined in ARCADIA is mostly based on NT pro BNP elevation(76%of patients)and is associated with age and ***,AC was not independently associated with AFDAS at *** ARCADIA trial,which compares aspirin to apixaban in patients with embolic strokes of undetermined source with AC markers and must,therefore be analysed in the light of these limitations.

Variable Chlorine Decay Rate Modeling of the Matsapha Town Water Network Using EPANET Program

A variable chlorine decay rate modeling of the Matsapha town water network was developed based on initial chlorine dosages. The model was adequately described by a second order rate function of the chlorine decay rate with respect to the initial chlorine dose applied. Simulations of chlorine residuals within the Matsapha water distribution network were run using the EPANET 2.0 program at different initial chlorine dosages and using the variable decay rate as described by the second order model. The measurement results indicated that the use of constant decay rate tended to underestimate chlorine residuals leading to potentially excess dosages with the associated chemical cost and side effects. The error between the two rate models varied between 0% and 15%. It is suggested that the use of water quality simulation programs such as EPANET be enhanced through the extension programs that accommodate variable rate modeling of chlorine residuals within distribution systems.

liposomes effectively transport fatty proteins to targeted tissues. laboratory experiments use multiple methods to administer liposomes, but comparison of these methods is not available. In this retrospective study, we characterized and compared four intravenous administration routes (tail vein, jugular catheter, femoral vein and percutaneous retro-orbital injections) in murine models. ApoE-/- mice were used to compare administration routes. Results indicate that the jugular catheter route delivered the highest amount of liposomes to tissues due to longer period of injections compared to other routes;however, this route failed to remain patent for 8/10 animals. Delivery via tail vein, femoral vein and percutaneous retro-orbital injections resulted in similar accumulation in the organs. When including technical difficulty and expense, percutaneous retro-orbital injections of liposomes are the most convenient and efficacious approach.

Quantifying Isotibolone in Raw Materials of Tibolone

Tibolone, a synthetic steroid, is used in the treatment of natural or surgical menopause disturbs resultant of estrogenic deficiency. Isotibolone (Δ4-tibolone) is one of the three active metabolic degradation products of tibolone that displays progestagenic effects on carcinoma cell growth and gene regulation. Isotibolone can be present in raw material of tibolone due to some inadequate synthesis or storage. Its presence is necessary to be identified and quantified in active pharmaceutical ingredients (API), before its use in the manufacturing of medicines. After a recent study on the crystal structure determination of isotibolone, quantitative phase analyses of both tibolone and isotibolone in raw materials and tablets became possible to be conducted. X-ray powder diffraction is one recommended tool for this purpose, but it can be highly frustrating due to the extreme peak overlap when conventional laboratory equipments are used. In this work we show that the use of Brazilian Synchrotron light Source X-ray powder diffraction data and the Rietveld method can be successfully applied to identify and quantify the isotibolone in two samples of tibolone raw materials.

Quasi-type II heterostructured nanocrystals(NCs)have been of particular interest due to their great potential for controlling the interplay of charge ***,the lack of material choices for quasi-type II NCs restricts the accessible emission wavelength from red to near-infrared(NIR),which hinders their use in light-emitting applications that demand a wide range of visible ***,we demonstrate a new class of quasi-type II nanoemitters formulated in ZnSe/ZnSe_(1-X)Te_(X)/ZnSe seed/spherical quantum well/shell heterostructures(SQWs)whose emission wavelength ranges from blue to *** a given geometry,ZnSe_(1-X)Te_(X) emissive layers grown between the ZnSe seed and the shell layer are strained to fit into the surrounding media,and thus,the lattice mismatch between ZnSe_(1-X)Te_(X) and ZnSe is effectively *** addition,composition of the ZnSe_(1-X)Te_(X) emissive layer and the dimension of the ZnSe shell layer are engineered to tailor the distribution and energy of electron and hole wave *** from the capabilities to tune the charge carriers on demand and to form defectfree heterojunctions,ZnSe/ZnSe_(1-X)Te_(X)/ZnSe/ZnS NCs show near-unity photoluminescence quantum yield(PlQY>90%)in a broad range of emission wavelengths(peak Pl from 450nm to 600 nm).Finally,we exemplify dichromatic white NC-based light-emitting diodes(NC-lEDs)employing the mixed layer of blue-and yellow-emitting ZnSe/ZnSe_(1-X)TeX/ZnSe/ZnS SQW NCs.

Obesity and cardiometabolic disease risk factors among US adolescents with disabilities

AIM: To generate prevalence estimates of weight status and cardiometabolic disease risk factors among adolescents with and without ***: Analysis of the 1999-2010 National Health and Nutrition Examination Survey data was conducted among 12-18 years old with(n = 256) and without disabilities(n = 5020). Mean values of waist circumference, fasting glucose, high-density-lipoprotein cholesterol, triglycerides, systolic and diastolic blood pressure and metabolic syndrome(Met S, ≥ 3 risk factors present) were examined by the following standardized body mass index(BMI) categories for those with and without disabilities; overweight(BMI ≥ 85th- le for age and sex), obesity(BMI ≥ 95 th percentile) and severe obesity(BMI ≥35 kg/m2). linear regression models were fit with each cardiometabolic disease risk factor independently as continuous outcomes to show relationships with disability status. RESUlTS: Adolescents with disabilities were significantlymore likely to be overweight(49.3%), obese(27.6%) and severely obese(12%) vs their peers without disabilities(33.1%, 17.5% and 3.6%, respectively, P ≤ 0.01 for all). A higher proportion of overweight, obese and severely obese children with disabilities had abnormal SBP, fasting lipids and glucose as well as Met S(18.9% of overweight, 32.3% of obese, 55% of severely obese) vs their peers without disabilities(9.7%, 16.8%, 36.3%, respectively). US adolescents with disabilities are over three times as likely to have Met S(OR = 3.45, 95%CI: 1.08-10.99, P = 0.03) vs their peers with no ***: Results show that adolescents with disabilities are disproportionately affected by obesity and poor cardiometabolic health vs their peers with no disabilities. Health care professionals should monitor the cardiometabolic health of adolescents with disabilities.

Mountains are paramount for exploring biodiversity patterns due to the mosaic of topographies and climates encompassed over short *** research has traditionally focused on taxonomic diversity when investigating changes along elevational gradients,but other facets should be *** first time,we simultaneously assessed elevational trends in taxonomic,functional,and phylogenetic diversity of woody plants in Andean tropical montane forests and explored their underlying ecological and evolutionary *** investigation covered four transects(traversing ca.2200 m a.s.l.) encompassing 114 plots of 0.1 ha across a broad latitudinal range(ca.10°).Using Hill numbers to quantify abundance-based diversity among 37,869 individuals we observed a consistent decrease in taxonomic,functional,and phylogenetic diversity as elevation increased,although the decrease was less pronounced for higher Hill *** exception was a slight increase in phylogenetic diversity when dominant species were *** decrease in taxonomic and functional diversity might be attributed to an environmental filtering process towards highlands,where the increasingly harsher conditions exclude species and functional ***,the differences in steepness decrease between Hill orders suggest that rare species disproportionately contribute to functional *** phylogenetic diversity the shifting elevational trend between Hill orders indicates a greater than previously considered influence in central Andean highlands of tropical lowlands originated species with strong niche conservatism relative to distantly related temperate *** could be explained by a decreasing presence and abundance of temperate,extratropical taxa towards the central Andes relative to northern or southern Andes,where they are more prevalent.
