Almond is appreciated for its nutraceutical value and for the aromatic profile of the *** this work,an almond collection composed of 96 Sicilian accessions complemented with 10 widely cultivated cultivars was phenotyped for the production of volatile organic compounds using a proton-transfer time-of-flight mass spectrometer and genotyped using the Illumina Infinium®18 K Peach SNP *** profiling of the aroma was carried out on fresh and roasted kernels enabling the detection of 150 mass *** eight,for the most related with sulfur compounds,furan containing compounds,and aldehydes formed by Strecker degradation,significantly increased during roasting,while the concentration of fifty-four mass peaks,for the most belonging to alcohols and terpenes,significantly *** hundred and seventy-one robust SNPs were selected and employed for population genetic *** analysis detected three subpopulations with the Sicilian accessions characterized by a different genetic stratification compared to those collected in Apulia(South Italy)and the International *** linkage-disequilibrium(LD)decay across the genome was equal to r^(2)=***,a high level of collinearity(r^(2)=0.96)between almond and peach was registered confirming the high synteny between the two genomes.A preliminary application of a genome-wide association analysis allowed the detection of significant marker-trait associations for 31 fresh and 33 roasted almond mass peaks *** accurate genetic and phenotypic characterization of novel germplasm can represent a valuable tool for the set-up of marker-assisted selection of novel cultivars with an enhanced aromatic profile.

Background:Skeletal muscle energetics decline with age,and physical activity(PA)has been shown to offset these declines in older ***,many studies reporting these effects were based on self-reported PA or structured exercise ***,we examined the associations of accelerometry-measured and self-reported PA and sedentary behavior(SB)with skeletal muscle energetics and explored the extent to which PA and sedentary behavior would attenuate the associations of age with muscle ***:As part of the Study of Muscle,Mobility and Aging,enrolled older adults(n=879),810(age=76.4±5.0 years old,mean±SD;58%women)had maximal muscle oxidative capacity measured ex vivo via high-resolution re spirometry of permeabilized myofibers(maximal oxidative phosphorylation(maxOXPHOS))and in vivo by ^(31)phosphorus magnetic resonance spectroscopy(maximal adenosine triphosphate(ATP_(max))).Accelerometry-measured sedentary behavior,light activity,and moderate-to-vigorous PA(MVPA)were assessed using a wrist-worn Actigraph gT9X over 7 ***-reported sedentary behavior,MVPA,and all PA were assessed with the Community Healthy Activities Model Program for Seniors(CHAMPS)*** regression models with progressive covariate adjustments evaluated the associations of sedentary behavior and PA with muscle energetics,as well as the attenuation of the age/muscle energetics association by MVP A and sedentary *** a sensitivity analysis,we also examined activPAL-measured daily step count and time spent in sedentary behavior and their associations with muscle ***:Every 30 min/day more of Actigraph-measured MVPA was associated with 0.65 pmol/(s×mg)higher maxOXPHOS and 0.012 mM/s higher ATP_(max)after adjusting for age,site/technician,and sex(pght activity was not associated with maxOXPHOS or ATP_(max).Meanwhile,every 30 min/day spent in Actigraph-measured sedentary behavior was associated with 0.39 pmol/s×mg lower maxOXPHOS and0.

Pancreatic ductal adenocarcinoma(PDAC)is a lethal disease,characterized by an intense desmoplastic reaction that compresses blood vessels and limits nutrient *** aggressiveness largely relies on its extraordinary capability to thrive and progress in a challenging tumor *** of the onco-suppressor miR-15a has been extensively documented in ***,we identified the transcription factor Fos-related antigen-2(Fra-2)as a miR-15a target mediating the adaptive mechanism of PDAC to nutrient *** report that the IgF1 signaling pathway was enhanced in nutrient deprived PDAC cells and that Fra-2 and IgF1R were significantly overexpressed in miR-15a downmodulated PDAC ***,we discovered that miR-15a repressed IgF1R expression via Fra-2 *** miR-15a-low context,IgF1R hyperactivated mTOR,modulated the autophagic flux and sustained PDAC growth in nutrient *** a genetic mouse model,Mir15aKO PDAC showed Fra-2 and Igf1r upregulation and mTOR activation in response to diet ***,nutrient restriction improved the efficacy of IgF1R inhibition in a Fra-2 dependent ***,our results point to a crucial role of Fra-2 in the cellular stress response due to nutrient restriction typical of pancreatic cancer and support IgF1R as a promising and vulnerable target in miR-15a downmodulated PDAC.

Smartphones have ubiquitously integrated into our home and work environments,however,users normally rely on explicit but inefficient identification processes in a controlled ***,when a device is stolen,a thief can have access to the owner’s personal information and services against the stored *** a result of this potential scenario,this work proposes an automatic legitimate user identification system based on gait biometrics extracted from user walking patterns captured by smartphone sensors.A set of preprocessing schemes are applied to calibrate noisy and invalid samples and augment the gait-induced time and frequency domain features,then further optimized using a non-linear unsupervised feature selection *** selected features create an underlying gait biometric representation able to discriminate among individuals and identify them *** classifiers are adopted to achieve accurate legitimate user *** experiments on a group of 16 individuals in an indoor environment show the effectiveness of the proposed solution:with 5 to 70 samples per window,KNN and bagging classifiers achieve 87–99%accuracy,82–98%for ELM,and 81–94%for *** proposed pipeline achieves a 100%true positive and 0%false-negative rate for almost all classifiers.

Osteoarthritis action alliance consensus opinion-best practice features of anterior cruciate ligament and lower limb injury prevention programs

AIM To identify best practice features of an anterior cruciate ligament(ACL) and lower limb injury prevention programs(IPPs) to reduce osteoarthritis(OA).METHODS This consensus statement started with us performing a systematic literature search for all relevant articles from 1960 through January 2017 in PubM ed, Web of Science and CINAHL. The search strategy combined the Medical Subject Heading(Me SH) and keywords for terms:(1) ACL OR "knee injury" OR "anterior cruciate ligament";(2) "prevention and control" OR "risk reduction" OR "injury prevention" OR "neuromuscular training"; and(3) meta-analysis OR "systematic review" OR "cohort study" OR randomized. We found 166 different titles. The abstracts were reviewed for pertinent papers. The papers were reviewed by at least two authors and consensus of best practice for IPP to prevent OA was obtained by conference calls and e-mail discussions. All authors participated in the *** The best practice features of an IPP have the following six components:(1) lower extremity and core strengthening;(2) plyometrics;(3) continual feedback to athletes regarding proper technique;(4) sufficient dosage;(5) minimal-to-no additional equipment; and(6) balance training to help prevent injuries. Exercises focused on preventing ankle sprains, hamstring injuries and lateral trunk movements are important. Plyometric exercises should focus on correcting knee valgus *** should focus on optimizing the hamstring to quadriceps strength ratio. In order for IPP to be successful, there should be increased education and verbal feedback along with increased athletic compliance. Additional equipment is not necessary. Balance training alone does not significantly reduce injuries, but is beneficial with other exercises. Not enough evidence to recommend stretching and agility exercises, with no ill effects identified. Therefore, we suggest making these optional *** Best practice features for ACL and lower limb

Rapid radiations in Fungi are only beginning to be studied with phylogenomic *** evolutionary history of the lichenized fungal order Peltigerales has not been well resolved,particularly for the ***,we used concatenation and coalescent-based species tree methods to reconstruct the phylogeny of the Peltigerales based on sequences of 125 nuclear single-copy exon sequences among 60 samples,representing 58 *** uneven,lineage-specific missing data and significant topological incongruence of individual exon trees,the resulting phylogenies were concordant and successfully resolved the phylogenetic relationships of the *** in the Collematineae were defined by short branches and lower nodal support than in other parts of the tree,due in part to conflicting signal in exon trees,suggesting rapid diversification events in the early evolution of the *** tree distance measures,we were able to identify a minimum subset of exons that could reconstruct phylogenetic relationships in Peltigerales with higher support than the 125-exon *** between the minimum and complete datasets in species tree inferences,bipartition analyses,and divergence time estimations displayed similar results,although the minimum dataset was characterized by higher levels of error in estimations of divergence *** our inferences from the complete and minimum datasets to those derived from few nuclear and mitochondrial loci reveal that our topology is concordant with topologies reconstructed using the nuclear large subunit and mitochondrial small subunit ribosomal DNA markers,but the target capture datasets had much higher support *** demonstrated how target capture approaches can effectively decipher ancient rapid radiations in cases where well resolved individual exon trees are sufficiently sampled and how to identify subsets of loci that are appropriate for fungal order-level phylogenetics.

Tumor progression and metastasis are the major causes of death among cancer associated *** cells acquire features of migration and invasion and usually undergo epithelia-mesenchymal transition(EMT).Acquirement of these various hallmarks rely on different cellular pathways,including TgF-βand Wnt ***,we reported that WW domain-containing oxidoreductase(WWOX)acts as a tumor suppressor and has anti-metastatic activities involving regulation of several key microRNAs(miRNAs)in triple-negative breast cancer(TNBC).Here,we report that WWOX restoration in highly metastatic MDA-MB435S cancer cells alters mRNA expression profiles;further,WWOX interacts with various proteins to exert its tumor suppressor *** alignment and analysis of gene and miRNA expression in these cells revealed profound changes in cellular pathways mediating adhesion,invasion and *** further demonstrate that WWOX,through regulation of miR-146a levels,regulates SMAD3,which is a member of the TgF-βsignaling ***,proteomic analysis of WWOX partners revealed regulation of the Wnt-signaling activation through physical interaction with ***,these findings underscore a significant role for WWOX in antagonizing metastasis,further highlighting its role and therapeutic potential in suppressing tumor progression.
