Objective: The purpose of this study was to investigate the effects of desensitized nicotinic receptors(n ACh Rs) on striatal dopaminergic system in the hemiparkinsonian rats treated with 6-hydroxydopamine(6-OHDA). Methods: We examined the effects of desensitized n ACh Rs on the levels of dopamine(DA) and its metabolites, m RNA expression of dopamine receptor D1,D2 and monoamine oxidase B(MAO-B) in the striatum of 6-OHDA-lesioned rats using high-performance liquid chromatography and reverse transcription-polymerase chain reaction. Results: The results showed that n ACh Rs desensitization following repeated nicotine stimulation could reverse significantly the decrease of striatal DA and its metabolites levels and the increase in DA turnover in lesioned side striatum of hemiparkinsonian rats. Dopamine D1 receptor m RNA expression increased significantly, whereas dopamine D2 receptor m RNA expression remained unchanged in lesioned side striatum of nicotine-treated rats compared to 6-OHDA-lesioned rats when n ACh Rs were desensitized. Meanwhile, nicotine-treated rats displayed a significant decrease in MAO-B m RNA expression in lesioned side striatum compared to 6-OHDA-lesioned rats after n ACh Rs desensitization. Conclusion: These results suggest that n ACh Rs desensitization could promote DA level, upregulate dopamine D1 receptor expression and downregulate MAO-B expression in striatum of hemiparkinsonian rats.

Network localization serves as a fundamental component for enabling various position based operations in multi-agent systems,facilitating tasks like target searching and formation control by providing accurate position information for all nodes in the network.Network localization focuses on the challenge of determining the positions of nodes within a network,relying on the known positions of anchor nodes and internode relative measurements.Over the past few decades,distributed network localization has garnered significant attention from researchers.This paper aims to provide a review of main results and advancements in the field of distributed network localization,with a particular focus on the perspective of graph Laplacian.Owning to its favorable characteristics,graph Laplacian unifies various network localization,even when dealing with diverse types of internode relative measurements,into a unified protocol framework,which can be constructed by a linear method and ensure the global convergence.

Introduction:Human immunodeficiency virus(HIV)voluntary counseling and testing(VCT)clinics play a critical role in identifying and diagnosing HIV cases.This study aimed to describe the trend of HIV positivity rate(HPR)among Chinese VCT clinics between 2015 and 2022.Methods:This study utilized data from the China Information System for Disease Control and Prevention to analyze the trend in the HPR for VCT clinics from 2015 to 2022.The HPR was calculated by dividing the number of newly-reported HIV cases by the number of HIV tests,multiplied by 100%.To identify temporal and spatial trends in the HPR,we employed joinpoint regression analysis and the Getis-Ord hotspot analysis.Results:From 2015 to 2022,VCT clinics in China performed a total of 22,075,386 HIV tests,leading to the identification of 260,353 HIV cases,resulting in a HPR of 1.18%.The HPR consistently declined over the study period,with an average annual percent change(AAPC)of−7.5%(95%confidence interval:−12.6%,−2.2%,Pfurther improving the utilization of VCT clinics for HIV testing.

The profound influence of microbiota in cancer initiation and progression has been under the spotlight for years,leading to numerous researches on cancer microbiome entering clinical evaluation.As promising biomarkers and therapeutic targets,the critical involvement of microbiota in cancer clinical practice has been increasingly appreciated.Here,recent progress in this field is reviewed.We describe the potential of tumor-associated microbiota as effective diagnostic and prognostic biomarkers,respectively.In addition,we highlight the relationship between microbiota and the therapeutic efficacy,toxicity,or side effects of commonly utilized treatments for cancer,including chemotherapy,radiotherapy,and immunotherapy.Given that microbial factors influence the cancer treatment outcome,we further summarize some dominating microbial interventions and discuss the hidden risks of these strategies.This review aims to provide an overview of the applications and advancements of microbes in cancer clinical relevance.

Splenic infarction(SI)is a rare clinical phenomenon that occurs when the blood supply to the spleen is interrupted,resulting in tissue ischemia and necrosis[1].There are many causes of SI;in adult patients,cardioembolism,hematological malignancy,and infectious disease are the most common[2].Rheumatic diseases,especially systemic lupus erythematosus(SLE)and granulomatosis with polyangitis,can also lead to SI[3];however,only a few cases have been reported.There are currently no statistically meaningful data on the etiology of SI in children.Although most patients with SI can recover after conservative treatment,a small number of patients develop complications,such as splenic abscess,splenic rupture,splenic hemorrhage,and hemoperitoneum,and may even undergo splenectomy in severe cases.

A semantic segmentation-based underwater acoustic image transmission framework for cooperative SLAM

With the development of underwater sonar detection technology,simultaneous localization and mapping(SLAM)approach has attracted much attention in underwater navigation field in recent years.But the weak detection ability of a single vehicle limits the SLAM performance in wide areas.Thereby,cooperative SLAM using multiple vehicles has become an important research direction.The key factor of cooperative SLAM is timely and efficient sonar image transmission among underwater vehicles.However,the limited bandwidth of underwater acoustic channels contradicts a large amount of sonar image data.It is essential to compress the images before transmission.Recently,deep neural networks have great value in image compression by virtue of the powerful learning ability of neural networks,but the existing sonar image compression methods based on neural network usually focus on the pixel-level information without the semantic-level information.In this paper,we propose a novel underwater acoustic transmission scheme called UAT-SSIC that includes semantic segmentation-based sonar image compression(SSIC)framework and the joint source-channel codec,to improve the accuracy of the semantic information of the reconstructed sonar image at the receiver.The SSIC framework consists of Auto-Encoder structure-based sonar image compression network,which is measured by a semantic segmentation network's residual.Considering that sonar images have the characteristics of blurred target edges,the semantic segmentation network used a special dilated convolution neural network(DiCNN)to enhance segmentation accuracy by expanding the range of receptive fields.The joint source-channel codec with unequal error protection is proposed that adjusts the power level of the transmitted data,which deal with sonar image transmission error caused by the serious underwater acoustic channel.Experiment results demonstrate that our method preserves more semantic information,with advantages over existing methods at the same compression ratio.It also improves the error

Artificial CO_(2)removal from the atmosphere(also referred to as negative CO_(2)emissions)has been proposed as a potential means to counteract anthropogenic climate change.Here we use an Earth system model to examine the response of ocean acidification to idealized atmospheric CO_(2)removal scenarios.In our simulations,atmospheric CO_(2)is assumed to increase at a rate of 1%per year to four times its pre-industrial value and then decreases to the pre-industrial level at a rate of 0.5%,1%,2%per year,respectively.Our results show that the annual mean state of surface ocean carbonate chemistry fields including hydrogen ion concentration([H^(+)]),pH and aragonite saturation state respond quickly to removal of atmospheric CO_(2).However,the change of seasonal cycle in carbonate chemistry lags behind the decline in atmospheric CO_(2).When CO_(2)returns to the pre-industrial level,over some parts of the ocean,relative to the pre-industrial state,the seasonal amplitude of carbonate chemistry fields is substantially larger.Simulation results also show that changes in deep ocean carbonate chemistry substantially lag behind atmospheric CO_(2)change.When CO_(2)returns to its pre-industrial value,the whole-ocean acidity measured by[H^(+)]is 15%-18%larger than the pre-industrial level,depending on the rate of CO_(2)decrease.Our study demonstrates that even if atmospheric CO_(2)can be lowered in the future as a result of net negative CO_(2)emissions,the recovery of some aspects of ocean acidification would take decades to centuries,which would have important implications for the resilience of marine ecosystems.

Screening of aplastic anaemia-related genes in bone marrow CD4^+ T cells by suppressive subtractive hybridization

Background CD4^＋ T cells play a crucial role in the pathogenesis of aplastic anaemia. However, the mechanisms of over-proliferation, activation, infiltration of bone marrow and damage to haematopoietic ceils of CD4^＋ T cells in aplastic anaemia are unclear. Therefore, we screened differentially expressed genes of bone marrow CD4^＋ T cells of aplastic anaemia patients and normal donors by suppressive subtractive hybridization to investigate the pathogenesis of aplastic anaemia. Methods The bone marrow mononuclear cells of a first visit aplastic anaemia patient and a healthy donor of the same age and sex were isolated using lymphocyte separating medium by density gradient centrifugation. With the patients as ＂tester＂ and donor as ＂driver＂, their CD4^＋T cells were separated with magnetic bead sorting and a cDNA library established by suppressive subtractive hybridization. Then 15 of the resulting subtracted cDNA clones were randomly selected for DNA sequencing and homological analysis. With semiquantitative RT-PCR, bone marrow samples from 20 patients with aplastic anaemia and 20 healthy donors assessed the expression levels of differentially expressed genes from SSH library. Results PCR detected 89 clones in the library containing an inserted fragment of 100 bp to 700 bp. Among 15 sequenced clones, 12 were known genes including 3 repeated genes. Compared with normal donors, there were 9/12 genes over-expressed in bone marrow CD4^＋T cells of patients with aplastic anaemia. The effects of these genes included protein synthesis, biology oxidation, signal transduction, proliferative regulation and cell migration. Not all these genes had been reported in the mechanisms of haematopoietic damage mediated by CD4^＋ T cells in aplastic anaemia. Conclusions Screening and cloning genes, which regulate functions of CD4^＋ T cells, are helpful in elucidating the mechanisms of over proliferation, activation, infiltrating bone marrow and damaging haematopoietic cells of CD4^＋ T cells in aplasUc anaemia.

Objective To investigate the anti-tumor effects of Pien Tze Huang(PZH)in mouse models of B16–F10 melanoma,MC38 colorectal cancer,Hep1-6 hepatocellular carcinoma and chemically induced hepatocellular carcinoma model.Methods Various tumor models,including B16–F10,MC38 and Hep1-6 tumor hypodermic inoculation models,B16–F10 and Hep1-6 pulmonary metastasis models,Hep1-6 orthotopic implantation model,and chemically induced hepatocellular carcinoma model,were utilized to evaluate the anti-tumor function of PZH.Tumor growth was assessed by measuring tumor size and weight of solid tumors isolated from C57BL/6 mice.For cell proliferation and death of tumor cells in vitro,as well as T cell activation markers,cytokine production and immune checkpoints analysis,single-cell suspensions were prepared from mouse spleen,lymph nodes,and tumors after PZH treatment.Results PZH demonstrated significant therapeutic efficacy in inhibiting tumor growth(P0.05).Nevertheless,PZH exhibited a remarkable ability to enhance T cell activation and the production of interferon gamma,tumor necrosis factor alpha,and interleukin 2 in CD4^(+)T cells in vitro(Pfunction of PZH in T cell-mediated anti-tumor immunity,indicating that PZH holds promise as a potential therapeutic agent for cancer treatment.

Effects of Acupotomy on Immobilization-Induced Gastrocnemius Contracture and Fibrosis in Rats via Wnt/β-Catenin Signaling

Objective: To determine whether acupotomy ameliorates immobilization-induced muscle contracture and fibrosis via Wnt/β-catenin signaling pathway. Methods: Thirty Wistar rats were randomly divided into 5 groups(n=6) by a random number table, including control, immobilization, passive stretching, acupotomy, and acupotomy 3 weeks(3-w) groups. The rat model of gastrocnemius contracture was established by immobilizing the right hind limb in plantar flexion for 4 weeks. Rats in the passive stretching group received passive stretching at gastrocnemius, a daily series of 10 repetitions for 30 s each at 30-s intervals for 10 consecutive days. Rats in the acupotomy and acupotomy 3-w groups received acupotomy once and combined with passive stretching at gastrocnemius a daily series of 10 repetitions for 30 s each at 30-s intervals for 10 consecutive days. Additionally, rats in the acupotomy 3-w group were allowed to walk freely for 3 weeks after 10-day therapy. After treatment, range of motion(ROM), gait analysis [i.e., paw area, stance/swing and maximum ratio of paw area to paw area duration(Max dA/dT)], gastrocnemius wet weight and the ratio of muscle wet weight to body weight(MWW/BW) were tested. Gastrocnemius morphometric and muscle fiber cross-sectional area(CSA) were assessed by hematoxylin-eosin staining. Fibrosis-related mRNA expressions(i.e., Wnt 1, β-catenin, axin-2, α-smooth muscle actin, fibronectin, and types Ⅰ and Ⅲ collagen) were measured using real-time quantitative polymerase chain reactions. Wnt 1, β-catenin and fibronectin concentrations were measured by enzyme-linked immunosorbent assay. Types Ⅰ and Ⅲ collagen in the perimysium and endomysium were analyzed using immunofluorescence. Results: Compared with the control group, ROM, gait function, muscle weight, MWW/BW and CSA were significantly decreased in the immobilization group(all P <0.01), while protein levels of types Ⅰ and Ⅲ collagen, Wnt 1, β-catenin, fibronectin and mRNA levels of fibrosis-related genes were obviously increased(all P<0.01).
