Type 1 diabetes(T1D),also referred to as insulin-dependent diabetes mellitus,is a debilitating disease that follows the destruction of pancreatic insulin-producingβcells by autologous T-cells.T1D primarily affects children and has a strong genetic component,with>50 susceptibility loci identified,including HLA-DQβchains.1 However,the striking differences between European regions with similar genetic backgrounds and a sharp rise in T1D incidence in developed countries over the last several decades suggest that environmental factor(s)also play a relevant etiopathogenic role.2 Given the accumulating evidence linking the gut microbiota to protection against metabolic diseases,3 Mariño et al.4 recently tested the hypothesis that short-chain fatty acids(SCFAs),which are the end products of fermentation of dietary fibers by anaerobic intestinal microbiota,can protect genetically susceptible mice from developing ***,the authors found that diets enriched in acetate or butyrate(two of the main SCFAs)can protect animals from developing diabetes through different and complementary cellular *** findings significantly enhance our understanding of the role of diet and gut microbiota in the development of autoimmune diseases and indicate that the use of medicinal foods may be a cost-effective treatment against T1D and other autoimmune diseases with a cellular *** that current anti-T1D approaches(which focus on prevention or modulation of the adaptive specific immune response against autoantigens)have been generally disappointing,5 such a new and refreshing strategy has attracted a great deal of attention(reviewed in Ref.2).

Invasive fungal diseases(IFD)impact morbidity,mortality,hospital stay and healthcare costs in critically ill patients,constituting an unmet medical need.1 Pathogenic fungi rarely cause IFD in immunocompetent individuals but become life-threatening in cases of immunodeficiency,young infants,disrupted mucosal barriers or polyantibiotic therapy,highlighting the role of immune surveillance in IFD control.^(2)

Apoptosis inhibitor of macrophages （AIMs）, a homologue of human Spa, is a mouse soluble member of the scavenger receptor cysteine-rich superfamily （SRCR-SF）. This family integrates a group of proteins expressed by innate and adaptive immune cells for which no unifying function has yet been described. Pleiotropic functions have been ascribed to AIM, from viability support in lymphocytes during thymic selection to lipid metabolism and anti-inflammatory effects in autoimmune pathologies. In the present report, the pathogen binding properties of AIM have been explored. By using a recombinant form of AIM （rAIM） expressed in mammalian cells, it is shown that this protein is able to bind and aggregate Gram-positive and Gram-negative bacteria, as well as pathogenic and saprophytic fungal species. Importantly, endogenous AIM from mouse serum also binds to microorganisms and secretion of AIM was rapidly induced in mouse spleen macrophages following exposure to conserved microbial cell wall components. Cytokine release induced by well-known bacterial and fungal Toll-like receptor （TLR） ligands on mouse splenocytes was also inhibited in the presence of rAIM. Furthermore, mouse models of pathogen-associated molecular patterns （PAMPs）-induced septic shock of bacterial and fungal origin showed that serum AIM levels changed in a time-dependent manner. Altogether, these data suggest that AIM plays a general homeostatic role by supporting innate humoral defense during pathogen aggression.

Cadmium Distribution in Tobacco Growing Soil Fractions: Its Influence on Dried Leaf Contents

Many authors have studied cadmium concentrations in tobacco leaves. However, crops such as tobacco store Cd mainly in their leaves and these are for human consumption. The objective of this study is to establish the cadmium levels present in each of the soils fractions throughout the growing period of tobacco plants, and to examine the influence of these fractions on the total content cadmium in dried leaves. The field select is situated near ofGranadacity (soils from Vega de Granada) loamy calcareous thermic typic Xerorthent. The plant (Nicotiana tabacum, cvs. RJ36 variety), and the soil dedicated to the tobacco crop were contaminated with a fixed amount of Cd (100 mg/kg of CdCl2) in the proximity of the root influence area. The contents of Cd are analysed in channel waters, in different soil fractions and dried tobacco leaves (mg/kg) with the different treatments. In conclusion, the water for irrigation is not a contamination risk for the soil even when user over lengthy periods of time. The fraction presenting higher average values is the one of Cd bound to CaCO3 with 0.161 ppm, and the Cd bioavailable fractions is second in importance, and propose a Cd content sequence in fractions, higher to lower: Cd-Carbonates > Cd-Bioavailable > Cd-Organically bound > Cd-Exchangeable > Cd-Soluble.

Psoriasis is a chronic inflammatory skin disease with a strong genetic background and is triggered by environmental *** evidence supports CD6,a lymphocyte surface receptor mostly expressed by T cells,as a putative target in ***,a humanized anti-CD6 antibody has been assayed for the treatment of certain autoimmune disorders,including ***,we present novel evidence in mice and humans for a direct involvement of CD6 in psoriasis ***,an attenuated form of imiquimod-induced psoriasis-like skin inflammation was demonstrated in CD6-deficient mice,as deduced from lower epidermal thickness and local reduced production of pro-inflammatory cytokines,namely,***,isolated CD4+CD62L+T cells from CD6-deficient mice displayed decreased in vitro T-helper type 17 ***,a statistically significant association between CD6 single-nucleotide polymorphisms(rs17824933,rs11230563 and rs12360861)and more severe forms of psoriasis was demonstrated in a cohort of 304 patients at three public hospitals from the metropolitan area of *** together,these results provide new supportive evidence of the contribution of the CD6 lymphocyte receptor in psoriasis at both experimental and clinical levels.

Introduction: The abdominal approach for the treatment of rectal tumors is associated with a considerable rate of morbidity. Transanal Endoscopic Microsurgery (TEM) is an alternative technique that is less invasive than radical surgery, and therefore has a lower associated morbidity. Moreover, with proper patient selection, TEM presents oncological outcomes comparable to radical surgery. The aim of this study is to review our results obtained with TEM and discuss its role in the treatment of malignant rectal lesions. Patients and Methods: A prospective descriptive study from June 2008 until February 2011. The indications for TEM were: early rectal neoplastic lesions (T1N0M0) with good prognostic factors;neoplastic lesions in more advanced stages in selected patients (high surgical risk, refusal of radical surgery or stoma, and palliative intention). Results: Resection by TEM was performed on 19 patients. The average hospital stay was 5.7 days with an associated morbidity of 16.7%. R0 resection was 88.8%. During the follow-up of 15 (3 - 31) months, no recurrence has been shown. Conclusions: TEM is a safe and effective procedure for the treatment of selected early malignant rectal lesions and is associated with low morbidity. It is a therapeutic strategy based on a multidisciplinary team, careful patient selection, an audited surgical technique and a strict follow-up protocol.

Genetic Diversity of the Endemic Xantus’ Hummingbird Using 16 Novel Polymorphic Microsatellite Loci, and Their Cross Amplification between Six Related Species

A set of 16 microsatellite loci was developed and characterized for the Xantus’ hummingbird (Hylocharis xantusii) by using 454 next-generation sequencing. Twenty-five H. xantusii samples from one population were genotyped;all loci were polymorphic, with the number of alleles ranging from three to ten. The mean observed heterozygosity was 0.681 for all loci. No significant linkage disequilibrium was detected, but five loci (Hxan05, 06, 09, 13 and 14) showed deviation from Hardy-Weinberg equilibrium. These microsatellite loci are the first to be characterized for H. xantusii. A moderate to high level of cross-species amplification was observed across the six hummingbird species (31% - 87.5%), with the best cross amplification results observed in the closest related species (H. leucotis, Cynanthus latirostris, Calypte costae). The availability of these molecular tools allows assessing questions integrating population genetics, ecology, conservation, and evolutionary history for H. xantusii and for other phyogenetically related species.

The use of molecular markers has improved the ecological and evolutionary research in the case of clonal reproduction species, allowing the identification of boundaries among clonal modules (ramets), genetic individuals (genets), and populations. Microsatellite markers were developed for Stenocereus gummosus, a columnar cactus with both sexual and clonal reproduction which is distributed in the Sonoran Desert, Mexico. 454-Pyrosequencing reads were analyzed to detect microsatellite markers. Forty primer pairs were screened to look for polymorphism. Nine loci were genotyped for two S. gummosus localities. Sampling strategy was intended to avoid collecting a genetic individual more than once, considering that clonal architecture for this cactus was previously deduced as clumped. Polymorphic loci exhibited low allele count, ranging from 2 - 7 (mean of 3.78 ± 0.62 SE);mean heterozygosity values were 0.221 and 0.234 HO and 0.408 and 0.306 HE, with FIS of 0.383 and 0.299, for peninsular and continental localities respectively. Unexpectedly, some multilocus genotypes were found repeated within locality, which were assumed as clones since data was evaluated as sufficient (clonal richness R of 0.966 and 0.897). These results were different from those previously reported: the distribution of clones might as well be intermingled, having a minimum ramet dispersion distance of 30 m. This characteristic was also consistent with the recent colonization proposed for this and other arid lands plants. A wider genetic neighborhood, due to clone dispersion might affect diversity indexes while increasing the chance of geitonogamy and mating among relatives. The markers isolation and its population characterization allowed addressing new questions about S. gummosus ecology, clonal reproduction and reproductive biology.

Massive Vesical Hernias and Its Management: Case Series and Literature Review

Background: Bladder hernias are infrequent and should be treated by the urologist, or an experienced surgeon. An inguinoscrotal hernia of the bladder should be suspected in male patients with voiding symptoms and a fluctuant mass in the scrotum that changes its volume during urination. The most common characteristics found on the series were patients with advanced age, sedentary, and overweight. Case Presentation: We present a case series of 4 patients with inguinoscrotal herniation of the bladder, the surgical management, and outcome. Conclusion: The surgical technique used was inguinal plasty with Lichtenstein technique and either laparoscopic or open surgery mesh-plug placement. None had surgical complications, and none had recurrence at their follow-up.
