Neurotherapeutic potential of erythropoietin after ischemic injury of the central nervous system

Erythropoietin (EPO) is one of the most successful biopharmaceuticals in history and is used for treating anemia of different origins. However, it became clear that EPO could also work in a neuroprotective, antiapoptotic, antioxidative, angiogenetic and neurotropic way. It causes stimulation of cells to delay cell apoptosis, especially in the central nervous system. In rodent models of focal cerebral ischemia, EPO showed an impressive reduction of infarct size by 30% and improvement of neurobehavioral outcome by nearly 40%. A large animal model dealing with ischemia and reperfusion of the spinal cord showed that EPO could reduce the risk of spinal cord injury significantly. In addition, some clinical studies tested whether EPO works in real live clinical settings. One of the most promising studies showed the innocuousness and improvements in follow-up, outcome scales and in infarct size, of EPO-use in humans suffering from ischemic stroke. Another study ended unfortunately in a negative outcome and an increased overall death rate in the EPO group. The most possible reason was the involvement of patients undergoing simultaneously systemic thrombolysis with recombinant tissue plasminogen activator. An experimental study on rats demonstrated that administration of EPO might exacerbate tissue plasminogen activator-induced brain hemorrhage without reducing the ischemic brain damage. This case shows clearly how useful animal models can be to check negative side effects of a treatment before going into clinical trials. Other groups looked in human trials at the effects of EPO on the outcome after ischemic stroke, relation to circulating endothelial progerdtor cells, aneurysmal subarachnoid hemorrhage, traumatic brain injury, hemoglobin transfusion thresholds and elective first-time coronary artery bypass surgery. Forecasting the spatial and temporal charging demand of fully electrified urban private car transportation based on large-scale traffic simulation

To support power grid operators to detect and evaluate potential power grid congestions due to the electrification of urban private cars,accurate models are needed to determine the charging energy and power demand of battery electric vehicles(bEVs)with high spatial and temporal ***,e-mobility traffic simulations are used for this *** particular,activity-based mobility models are used because they individually model the activity and travel patterns of each person in the considered geographical *** addition to inaccuracies in determining the spatial distribution of bEV charging demand,one main limitation of the activity-based models proposed in the literature is that they rely on data describing traffic flow in the considered ***,these data are not available for most places in the ***,this paper proposes a novel approach to develop an activity-based model that overcomes the spatial limitations and does not require traffic flow data as an input ***,a route assignment procedure assigns a destination to each bEV trip based on the evaluation of all possible *** basis of this evaluation is the travel distance and speed between the origin of the trip and the destination,as well as the car-access attractiveness and the availability of parking spots at the *** applicability of this model is demonstrated for the urban area of berlin,Germany,and its 448 *** each district in berlin,both the required daily bEV charging energy demand and the power demand are *** addition,the load shifting potential is investigated for an exemplary *** results show that peak power demand can be reduced by up to 31.7%in comparison to uncontrolled charging.

An approach for the coupled simulation of machining processes using multibody system and smoothed particle hydrodynamics algorithms

The prediction accuracy of a simulation method is limited by its theoretical background. This fact can lead to disadvantages regarding the simulation quality when investigating systems of high complexity, e.g. containing components showing a fairly different behavior. To overcome this limitation, co-simulation approaches are used more and more, combining the advantages of different simulation disciplines. That is why we propose a new strategy for the dynamic simulation of cutting processes. The method couples Lagrangian particle methods, such as the smoothed particle hydrodynamics （SPH） method, and multibody system （MbS） tools using co-simulations. We demonstrate the capability of the new approach by providing simulation results of an orthogonal cutting process and comparing them with experimental data. @ 2013 The Chinese Society of Theoretical and Applied Mechanics. Limitations of cancer margin delineation and surgical guidance by means of autofluorescence imaging under conditions of laser ablation were investigated and preliminary results are ***^(TM)(Novadaq Technologies Inc.,Canada)was used to capture digital images and Er:YAG laser(2.94μm,Glissando,WaveLight^(TM),Germany)was exploited to cause laser ablation on both normal and cancer sites of the *** was shown that changes of the autofluorescence image after ablation extend beyond the actual sizes of the ablation *** tumor tissue after the laser ablation starts to emit fluorescent light within the green wavelength band(490-550nm)similar to normal tissue stating that the current technology of in-process tissue classification ***,when the autofluorescence was collected in the red range(600-750nm),then the abnormal/normal contrast was reduced,but still present even after the laser *** present study highlights the importance of finding a proper technology for surgical navigation of cancer removal under conditions of high power effects in biological tissues.

Use of human pluripotent stem cells to study and treat retinopathies

Human cell types affected by retinal diseases(such as age-related macular degeneration or retinitis pimentosa) are limited in cell number and of reduced accessibility. As a consequence, their isolation for in vitro studies of disease mechanisms or for drug screening efforts is fastidious. Human pluripotent stem cells(h PSCs), either of embryonic origin or through reprogramming of adult somatic cells,represent a new promising way to generate models of human retinopathies, explore the physiopathological mechanisms and develop novel therapeutic strategies. Disease-specific human embryonic stem cells were the first source of material to be used to study certain disease states. The recent demonstration that human somatic cells, such as fibroblasts or blood cells, can be genetically converted to induce pluripotent stem cells together with the continuous improvement of methods to differentiate these cells into disease-affected cellular subtypes opens new perspectives to model and understand a large number of human pathologies, including retinopathies. This review focuses on the added value of h PSCs for the disease modeling of human retinopathies and the study of their molecular pathological mechanisms. We also discuss the recent use of these cells for establishing the validation studies for therapeutic intervention and for the screening of large compound libraries to identify candidate drugs.

Cuticular hydrocarbon profiles differ between ant body parts:implications for communication and our understanding of CHC diffusion

Insect cuticular hydrocarbons(CHCs)serve as communication signals and protect against *** form complex blends of up to 150 different *** to differences in molecular packing,CHC classes differ in melting *** is especially important in social insects like ants,which use CHCs to communicate within the colony and to recognize *** recognition models often assume a homogenous colony odor,where CHCs are collected,mixed,and redistributed in the postpharyngeal gland(PPG).Via diffusion,recognition cues should evenly spread over the body ***,CHC composition should be similar across body parts and in the *** test this,we compared CHC composition among whole-body extracts,PPG,legs,thorax,and gaster,across 17 ant species from 3 *** CHC composition differed between body parts,with consistent patterns across species and CHC ***-melting CHC classes were most abundant in the *** contrast,whole body,gaster,thorax,and legs had increasing proportions of CHC classes with higher melting *** CHC variation was highest for rather solid,late-melting CHC classes,suggesting that CHCs differ in their diffusion rates across the body *** results show that body parts strongly differ in CHC composition,either being rich in rather solid,late-melting,or rather liquid,early-melting *** implies that recognition cues are not homogenously present across the insect ***,the unequal diffusion of different CHCs represents a biophysical mechanism that enables caste differences despite continuous CHC exchange among colony members.
