Raltitrexed + irinotecan as second-line chemotherapy in elderly patients with advanced colorectal cancer

Aims and Background: Irinotecan is a standard option for relapsed/refractory advanced colo- rectal cancer. Combination with raltitrexed and irinotecan at lower than MTD doses should preserve disease stabilisation while decreasing toxicity. Patients and Methods: From January 2004 to April 2009, we analyzed, retrospectively, our data on irinotecan + raltitrexed, fixed doses, as a second-line chemotherapy in elderly pa- tients (>70 years) with advanced colorectal can- cer after failure of oxaliplatin based chemothera- py twenty-three patients were evaluated. Iri- notecan 350 mg + raltitrexed 2.6 mg were given every 3 weeks. Tumo r measurements were ob- tained after every third course of therapy. Toxic- ity was assessed weekly using the National Cancer Institute Common Toxicity Criteria, ver- sion 2. Results: The median number of treatment courses received per patient was 4 (range, 1 - 8). All pa-tients were assessable for toxicity and 21 for response. The most frequently observed severetoxicities were diarrhea (grade 2, 13%) No cases of significant neutropenia occurred. Ob- jective partial responses were observed in 3 pa- tients (13%). An additional 10 patients (43%) had stable disease as their best response. To date, 12 patients have progressed with a median time- to-progression of 4.3 months and a median sur- vival of 8.3 months. Conclusions: A three weekly irinotecan + raltitrexed administration can indu- ce tumor control in elderly patients with advanc- ed colorectalcancer that has progressed during or shortly after oxaliplatin-based chemotherapy. Characterization of acquired nutlin-3 resistant non-small cell lung cancer cells

Aim:The purpose of this manuscript is to study the potential characteristics of acquired nutlin-3 resistant non-small cell lung cancer cells(NSCLC).Nutlin-3 is an inhibitor of the murine-double minute 2 protein,the main negative regulator of wild type p53,of which several derivatives are currently in clinical ***:A549 NSCLC cells were exposed to increasing concentrations of nutlin-3 for a period of 18 *** derivates were cultured,and the most resistance subclone was selected for whole transcriptome *** set enrichment analysis was performed on differentially expressed genes between A549 nutlin-3 resistant cancer cells and the parental A549 p53 wild type cancer *** findings were validated at the gene,protein and/or functional ***:All nutlin-3 resistant subclones acquired mutations in the TP53 gene,resulting in overexpression of the mutant p53 *** most resistant subclone was enriched for genes related to epithelial to mesenchymal transition(EMT),resulting in increased migratory and invasive ***,these cells were enriched in genes related to inflammation,tissue remodelling,and ***,expression of several immune checkpoints,including PD-L1 and PD-L2,was significantly upregulated,and cisplatin-induced cell death was ***:Transcriptome analysis of a highly nutlin-3 resistant A549 subclone shows the relevance of studying(1)resistance to standard of care chemotherapy;(2)secretion of immunomodulating chemo-and cytokines;(3)immune checkpoint expression;and(4)EMT and invasion in nutlin-3 resistant cancer cells in addition to acquired mutations in the TP53 gene.

Monitoring of Acoustic Emission Signal of Loaded Axial Bearings

The disclosure of initial stadium of contact fatigue of rotating machines is very difficult. At present, some methods which are suitable for diagnosis exist, such as vibration method, shock pulse method, acoustic emission and others. The article describes laboratory tests with axial bearings which are monitored by acoustic emission method. In the article, experimental stands, measuring devices and chosen types of axial bearings are presented. The exploitation of acoustic emission signal for the lifetime evaluation of selected bearings is shown with the characteristics as number of counts, rise time, maximal amplitude or duration of event. The bearing lifetime is separated to three stages and a representative sample in its time and frequency domain for each of them is chosen. Final failures of the bearings are shown too. The paper is focused also on examples of results obtained with continuous AE signal sensing.

Hydrochar has potential applications in soil improvement and heavy metal *** would undergo the process of aging when introduced into the soil,altering its ***,recent studies have focused mainly on the artificial aging of hydrochar,which could not reveal the cumulative effect of multiple environmental ***,the periodical monitoring of the property and sorption behavior of hydrochar after amending soils is necessary to better understand the multifaceted mechanisms associated with the natural aging of *** study selected the sludge-derived hydrochar(SLHC)as a typical hydrochar and applied a 16-month rice-wheat-rice rotation to mimic the natural aging of hydrochar,focusing on changing properties and cadmium(Cd)sorption and literature contrast between aging strategies and biochar *** porosity,O abundance,and ash content of 16-month aged SLHC increased by 37%,47%,and 8.5%,respectively,facilitating Cd sorption due to surface complexation,pore sorption,and *** sorption percentage of Cd to SLHC was in the range of 11-14%for SLHC-A0 and increased to 17-31%for SLHC-A4 and 20-32%for SLHC-A16 after natural *** natural aging of SLHC induced by ash content played an essential role in Cd sorption site *** regression analysis showed that aging strategies on sorption behavior significantly differed between ***,studies involving natural aging with multiple environmental factors are preferred over those involving chemical or biological *** studies should continue to explore the mechanisms of natural aging-induced heavy metal sorption between hydrochar and *** results improve insights to appraise the potential of SLHC as soil amendments to alleviate the adverse effects of heavy metal contamination and provide an essential basis for researchers and staff in soil management and environmental prevention.

We reviewed the pleiotropic beneficial effects of the stable gastric pentadecapeptide BPC 157,three very recent demonstrations that may be essential in the gut-brain and braingut axis operation,and therapy application in the central nervous system disorders,in ***,given in the reperfusion,BPC 157 counteracted bilateral clamping of the common carotid arteries-induced stroke,sustained brain neuronal damages were resolved in rats as well as disturbed memory,locomotion,and *** therapy effect supports particular gene expression in hippocampal tissues that appeared in BPC 157-treated ***,there are L-NG-nitro arginine methyl ester(L-NAME)-and haloperidol-induced catalepsy as well as the rat acute and chronic models of 'positivelike' schizophrenia symptoms,that BPC 157 counteracted,and resolved the complex relationship of the nitric oxide-system with amphetamine and apomorphine(dopamine agents application),MK-801(non-competitive antagonist of the N-methyl-D-aspartate receptor) and chronic methamphetamine administration(to induce sensitivity).Thirdly,after rat spinal cord compression,there were advanced healing and functional recovery(counteracted tail paralysis).Likewise,in BPC 157 therapy,there is specific support for each of these topics:counteracted encephalopathies;alleviated vascular occlusion disturbances(stroke);counteracted dopamine disturbances(dopamine receptors blockade,receptors super sensitivity development,or receptor activation,over-release,nigrostriatal damage,vesicles depletion),and nitric oxide-system disturbances("L-NAME non-responsive,L-arginine responsive," and "L-NAME responsive,L-arginine responsive")(schizophrenia therapy);inflammation reduction,nerve recovery in addition to alleviated hemostasis and vessels function after compression(spinal cord injury therapy).Thus,these disturbances may be all resolved within the same agent's beneficial activity,i.e.,the stable gastric pentadecapeptide BPC 157.

Cholelithiasis-induced cholestasis is one of the most common causes of hospitalization due to gastrointestinal disease,yet considerable knowledge gaps exist in the pathogenesis of this *** can partially be explained by inadequate characterization of experimental cholestasis ***,we compared the transcriptional profile of commonly used mouse models for obstructive cholestasis and benchmarked them to human disease to identify the model(s)best suited for cholelithiasis-induced cholestasis research and to uncover conserved mechanisms involved in human and murine cholestasis.
