A robust methodology for the quantitative assessmentof the rat jawbone microstructure

Micro-computed tomography can be applied for the assessment of the micro-architectural characteristics of the cortical and trabecular bones in either physiological or disease conditions. However, reports often lack a detailed description of the methodological steps used to analyse these images, such as the volumes of interest, the algorithms used for image filtration, the approach used for image segmentation, and the bone parameters quantified, thereby making it difficult to compare or reproduce the studies. This study addresses this critical need and aims to provide standardized assessment and consistent parameter reporting related to quantitative jawbone image analysis. Various regions of the rat jawbones were screened for their potential for standardized micro-computed tomography analysis. Furthermore, the volumes of interest that were anticipated to be most susceptible to bone structural changes in response to experimental interventions were defined. In the mandible, two volumes of interest were selected, namely, the condyle and the trabecular bone surrounding the three molars. In the maxilla, the maxillary tuberosity region and the inter-radicular septum of the second molar were considered as volumes of interest. The presented protocol provides a standardized and reproducible methodology for the analysis of relevant jawbone volumes of interest and is intended to ensure global, accurate, and consistent reporting of its morphometry. Furthermore, the proposed methodology has potential, as a variety of rodent animal models would benefit from its implementation.

Pine-fruit shell (PFS) is a lignocellulosic residue derived from the fruit of Araucaria angustifolia, a coniferoustree native of South America, part of a whole vegetation of the Atlantic Forest, found in the South and Southwestof Brazil. In this work PFS will be characterized and used in the production of PFS-based polyols throughoxypropylation. Three series were chosen (PFS/propylene oxide (PO) (w/v, g/mL) of 30/70, 20/80 and 10/90)with four catalyst levels (5%, 10%, 15% and 20%, (w/w, PFS based)). Oxypropylation occurred at moderateconditions of temperature, pressure and time giving rise to liquid polyols with a homopolymer content (PPO)ranging from 4-65%, a hydroxyl number (IOH) between 257-605 mg KOH/g and viscosities (V) varying from0.76 Pa.s to 373.90 Pa.s (20°C) for the series 20/80 and 10/90, while for the series 30/70, the viscosity reachedvalues higher than 500 Pa.s, 20°C. The unreacted PFS (UR) varied between 3.6% and 77.4% (PFS-basis). Afterthe PFS-based polyols production and characterization, a principal component analysis (PCA) was performed inorder to evaluate the established interactions between the used formulation variables and the obtained polyolproperties. The PCA analysis allowed to clarify the interactions between PFS and PO contents and the finalbiopolyol properties (PPO, IOH, V and UR). This approach showed to be a simple method to rationally analyzethe influence of the input formulation variables on the final polyol properties.

Trauma to the peripheral nervous system often results in loss of motor and sensory functions of the affected area of the body,leading to a series of functional impairments(Allodi et al.,2012).Injuries to peripheral nerves initiate a series of complex events,known as Wallerian degeneration,which allows the injured axons to regenerate and reinnervate their targets(Allodi et al.,2012).Damage to a nerve induces multiple alterations,which include:axonal degeneration,breakdown of myelin sheath,Schwann cells(SC)proliferation and conversion to a repair phenotype which express cytokines and chemokines that allow the infiltration and activation of macrophages(Mietto et al.,2015;Jessen and Mirsky,2016).

Experimental approaches to select tree species for forest restoration:effects of light,water availability and interspecific competition in degraded areas

Knowing what native trees can recr uit on degraded areas allows selecting the best species to restore these ***,as this information is not often available,experimentation is required before large-scale *** study used ex situ experiments to make these decisions on *** with r-strategist plants,excessive solar radiation and water shortage commonly impair tree recruitment in open *** experiments focused on the interactions among these factors and were conducted with three pioneer species from seasonally dry forests of northwest Argentina,Anadenanthera colubrina,Ceiba chodatii and Jacaranda *** of each species were sown at two light levels(sunlight/shade),two rainfall levels(full/reduced)and two levels of interspecific competition(with/without competitor)in a tree *** emergence and survival were monitored over a year and the results indicate that species differentially respond to varying levels of light and *** of *** tolerated water shortages under elevated solar radiation,which are desirable features for forest *** of *** tolerated shade and drought,suggesting that they require shading for establishing in open ***,*** seedlings neither tolerated full sunlight nor water shortages,suggesting that this species requires shading and regular watering if used in *** of the effects of light and water,the survival of all species was reduced by interspecific *** results highlight the importance of experimentation for selecting the best species for forest restoration and can enhance the cost/benefit ratios of these actions.

The organosulfur compound sulforaphane(SFN;C6H11NOS2) is a potent cytoprotective agent promoting antioxidant, anti-inflammatory, antiglycative, and antimicrobial effects in in vitro and in vivo experimental models. Mitochondria are the major site of adenosine triphosphate(ATP) production due to the work of the oxidative phosphorylation(OXPHOS) system. They are also the main site of reactive oxygen species(ROS) production in nucleated human cells. Mitochondrial impairment is central in several human diseases, including neurodegeneration and metabolic disorders. In this paper, we describe and discuss the effects and mechanisms of action by which SFN modulates mitochondrial function and dynamics in mammalian cells. Mitochondria-related pro-apoptotic effects promoted by SFN in tumor cells are also discussed. SFN may be considered a cytoprotective agent, at least in part, because of the effects this organosulfur agent induces in mitochondria. Nonetheless, there are certain points that should be addressed in further experiments, indicated here as future directions, which may help researchers in this field of research.

Clinicopathological Description of a Chronic Lymphocytic Leukemia in a Dog

Hematopoietic tumors are associated with diagnostic and therapeutic challenges and a poor prognosis. Furthermore, their pathophysiology in dogs is not well understood. Chronic lymphocytic leukemia has been rarely reported in golden retrievers, perhaps due to difficulties in diagnosis and care. An 8-year-old spayed female golden retriever was referred to our hospital with a 2-month history of fatigue, weakness, and selective appetite. The dog was diagnosed with chronic lymphocytic leukemia based on blood smear evaluation. Despite treatment, the dog deteriorated and the owners opted for euthanasia. Histopathological examination revealed atypical lymphocytes in the spleen, extramedullary hematopoiesis in the spleen and liver, bone marrow depletion, moderate neutrophilic myocarditis, and acute renal tubular necrosis. In an immunohistochemical panel, cells in the spleen and bone marrow were positive for anti-CD20, anti-CD45, anti-CD3, anti-IBA-1, anti-Ki67, anti-MUM-1, and anti-PAX-5. In conclusion, the leukemia diagnosis was confirmed through reactive histiocytosis, marked extramedullary hematopoiesis, and other clinical and laboratory findings. Herein, we report a case of chronic lymphocytic leukemia in a female dog with clinical, laboratory, and necroscopy findings.

Endoscopic ultrasound-guided celiac plexus neurolysis using a reverse phase polymer

AIM:To assess the feasibility of endoscopic ultrasound(EUS)guided celiac plexus neurolysis(CPN) using a poloxamer. METHODS:In this prospective evaluation,six Yorkshire pigs underwent EUS-guided *** received an injection of 10 mL of 0.25%Lidocaine plus methylene blue(group 1) and three received an injection of 10 mL of 0.25%Lidocaine plus blue colored poloxamer(PS137-25)(group 2) .Necropsy was performed immediately after the animals were *** abdominal and pelvic cavities were examined for the presence of methylene blue and the blue colored ***:EUS-guided CPN was successfully performed in all 6 pigs without immediate *** blue was identified throughout the peritoneal and retroperitoneal cavity in group *** blue colored poloxamer was found in the retroperitoneal cavity immediately adjacent to the aorta,in the exact location of the celiac plexus in group ***:EUS-guided CPN using a reverse phase polymer in a non-survival porcine model was technically *** presence of a poloxamer gel at the site of the celiac plexus at necropsy indicates a precise delivery of the neurolytic agent.
