Induction of chronic cholestasis without liver cirrhosis-Creation of an animal model

To analyze time intervals of inflammation and regeneration in a cholestatic rat liver *** 36 Lewis rats, divided into six groups of 6 animals (postoperative observation periods: 1, 2, 3, 4, 6, 8 wk), the main bile duct was ligated with two ligatures and observed for the periods mentioned above. For laboratory evaluation, cholestasis parameters (bilirubin, γ-GT), liver cell parameters (ASAT, ALAT) and liver synthesis parameters (quick, albumin) were determined. For histological analysis, HE, EvG, ASDCL and HMGB-1 stainings were performed. Furthermore, we used the mRNA of IL-33, GADD45a and p-21 for analyzing cellular stress and regeneration in cholestatic *** chemical laboratory and histological evaluation, a distinction between acute and chronic cholestatic liver injury with identification of inflammation and regeneration could be demonstrated by an increase in cholestasis (bilirubin: 1-wk group, 156.83 ± 34.12 μmol/L, P = 0.004) and liver cell parameters (ASAT: 2-wk group, 2.1 ± 2.19 μmol/L.s, P = 0.03; ALAT: 2-wk group, 1.03 ± 0.38 μmol/L.s, P = 0.03) after bile duct ligation (BDL). Histological evaluation showed an increase of bile ducts per portal field (3-wk group, 48 ± 6.13, P = 0.004) during the first four weeks after bile duct ligation. In addition to inflammation, which is an expression of acute cholestasis, there was an increase of necrotic areas in the histological sections (2-wk group, 1.38% ± 2.28% per slide, P = 0.002). Furthermore, the inflammation could be verified by ASDCL (4-wk group, 22 ± 5.93 positive cells per portal field, P = 0.041) and HMGB-1 [2-wk group, 13 ± 8.18 positive cells per field of view (FoV), P = 0.065] staining. Therefore, in summary of the laboratory evaluation and histological studies, acute cholestasis could be found during the first four weeks after bile duct ligation. Effect of Yttrium Implantation on Microstructure and Adhesive Property of Nickel Oxide Film

Isothermal and cyclic oxidation behaviors of pure and yttrium-implanted nickel were studied at 1000 ℃ in the air. Scanning electronic microscopy (SEM) and transmission electronic microscopy (TEM) were used to examine the micro-morphology and structure of oxide scales formed on nickel substrate. It was found that Y-implantation greatly improved the anti-oxidation ability of nickel both in isothermal and cyclic oxidizing experiments. Laser Raman Microscopy was also used to study the stress status of oxide scales formed on nickel with and without yttrium. The main reason for the improvement in anti-oxidation of nickel was that Y-implantation greatly reduced the growing speed and grain size of NiO. This fine-grained NiO oxide film might have better high temperature plasticity and could relieve parts of compressive stress by means of creeping, and maintained ridge character and relatively lower internal stress level, hence remarkably enhanced the adhesion of protective NiO oxide scale formed on nickel substrate. The actual existing form of yttrium in oxide can be Y2O3 and NiY2O4 spinel nanometric particles, and even be Y3+ ions segregated at NiO grain boundaries.

Is laparoscopy an advantage in the diagnosis of cirrhosis in chronic hepatitis C virus infection?

AIM:To evaluate the potential of laparoscopy in the diagnosis of cirrhosis and outcome of interferon treatment in HCV-infected patients. METHODS:In this retrospective study,diagnostic laparoscopy with laparoscopic liver biopsy was performed in 72 consecutive patients with chronic HCV *** presence or absence of drrhosis was analyzed macroscopically by laparoscopy and microscopically by liver biopsy *** and laboratory data and outcome of interferon-alfa treatment were compared between cirrhotic and noncirrhotic patients. RESULTS:Laparoscopically,cirrhosis was seen in 29.2 % (21/72)and non-cirrhosis in 70.8 %(51/72)of patients. Cirrhotic patients were significantly older with a significant longer duration of HCV infection than noncirrhotic patients. Laboratory parameters(AST,y-GT,y-globulin fraction)were measured significantly higher as well as significantly lower (prothrombin index,platelet count)in cirrhotic patients than in non-cirrhotic ***,cirrhosis was confirmed in 11.1%(8/72)and non cirrhosis in 88.9 %(64/72).Patients with macroscopically confirmed cirrhosis(n=21)showed histologically cirrhosis in 38.2 %(8/21)and histologically non- cirrhosis in 61.9 %(13/21).In contrast,patients with macroscopically non-cirrhosis(n=51)showed histologically non cirrhosis in all cases(51/51).Thirty-nine of 72 patients were treated with interferon-alfa,resulting in 35.9 %(14/39) patients with sustained response and 64.1%(25/39)with non ***-responders showed significantly more macroscopically cirrhosis than sustained *** contrast,there were no significant histological differences between non-responders and sustained responders. In-situ investigation on real-time suspended sediment measurement techniques:Turbidimetry, acoustic attenuation,laser diffraction (LISST) and vibrating tube densimetry

Real-time measurements of Suspended Sediment mass Concentration（SSC） and Particle Size Distribution（PSD） are of prime importance for the investigation and management of fine-sediment related processes in surface water systems and hydraulic schemes. In a field study at the waterway of the hydropower plant（HPP） Fieschertal in the Swiss Alps, the real-time measurement performance of the following techniques and instruments were assessed based on measurements in the sediment seasons of the years 2013 and 2014:（1） turbidimetry,（2） single-frequency acoustic attenuation using a standard acoustic discharge measurement（ADM） installation,（3） laser diffraction（Laser In-Situ Scattering and Transmissometry, LISST）, and（4） vibrating tube densimetry using a Coriolis Flow and Density Meter（CFDM）.Reference SSCs were obtained from gravimetric analysis of 219 automatically pumped water *** additionally supplied PSD every minute. The median particle diameter, d50, was usually 15μm and increased occasionally to 100μm. The turbidimeter and the ADM underestimated the SSC when the transported particles were coarser than usual. Such temporary biases resulted from the poor correlation between d50and SSC at this study site. The SSCs from CFDM and LISST were not or less biased by PSD variations. Mainly due to angular and flaky particle shapes, SSC from LISST needed a correction by 79% on average. With the usually prevailing silt particles, an optical path length of 5 mm and no dilution, the SSC measurement range of LISST was limited to about 1.5 g/l. The CFDM allowed measuring higher SSC than the other investigated instruments（e.g. up to 13.5 g/l）. With a periodic offset correction, its relative SSC measurement uncertainty was < 20% for SSC≥ 1.5 g/I. To reliably measure a wide range of SSC with temporarily variable PSD, a combination of instruments is recommended: e.g. a standard LISST, a CFDM,and an automatic water sampler for gravimetric reference measurements.

Complex Effective Relative Permittivity of Soil Samples from the Taunus Region (Germany)

The most important parameter affecting ground-penetrating radar （GPR） measurements is the complex effective relative permittivity εr^＊,eff because it controls the propagation velocity and the reflection of GPR pulses. Knowing εr^＊,eff of soils passed through by electromagnetic waves increases accuracy in soil thickness and interface identification. Complex effective relative permittivity εr^＊,eff= εr^＊,eff - jεr^＊,effof 25 soil samples with textures ranging from loamy sand to silty clay was measured using the two-electrode parallelplate method. The measurements were conducted at defined water contents for frequencies from 1 MHz to 3 GHz. The results confirm the frequency dependence of εr^＊,eff and show that the dielectric behavior of soil-water mixtures is a function of water content. Applying the experimental data of this study with predictions based on the empirical model by Toppet aL （1980）, we find that Topp et aL＇s curve tends to underestimate the real part of εr^＊,eff measured. Along with frequency and water content, soil texture and organic matter affect soil permittivity. Moreover, the real part of εr^＊,eff increases at higher dry bulk densities. Output from our calibration model enables us to predict εr^＊,eff for the soil samples which were tested under the actual in situ soil water content. This results in high accuracy of soil thickness prediction.

Combination of "low-dose" ribavirin and interferon alfa-2a therapy followed by interferon alfa-2a monotherapy in chronic HCV-infected nonresponders and relapsers after interferon alfa-2a monotherapy

AIM To report on the efficacy, safety and tolerability of interferon alfa-2a combined with a "low dose" of ribavirin for relapsers and non responders to alpha interferon *** Thirty-four chronic hepatitis C virus-infected non-responders to interferon alfa2a monotherapy (a course of at least 3 months treatment) and 13 relapsers to interferon alfa 2a monotherapy (a dose of 3 to 6 million units three times per week for at least 20 weeks but not more than 18 months) were treated with the same dose of interferon alfa-2a used before (3 to 6 million units three times per week) and ribavirin (10 mg/ kg daily) for 6 months. In complete responders, interferon alfa-2a was administered for further 6 months at the same dose used before as *** Seven (20.6%) of 34 non-responders stopped the combined therapy due to adverse events, including two patients with histological and clinical Child A cirrhosis. In 17/27 (63%)non-responders, the combined therapy was stopped after three months because of non-response. Ten of the 27 non-responders completed the 1;2-month treatment course. At a mean follow up of 28 months (16- 37 months)after the treatment, 4/10 (15%) previous non-responders still remained complete responders,All 13 previous relapsers completed the 12-month treatment course. At a mean follow up of 22months (9 - 36 months) after treatment, 6/13(46%) the previous relapsers were stillsustained complete *** Our treatment schedule of the combined therapy for 6 months of interferon alfa2a with a low dose of ribavirin (10 mg/kg/day)followed by 6 months of interferon alfa-2amonotherapy is able to induce a sustainedcomplete response rate in 15% of non-responders and 46% of relapsers with chronic hepatitis C virus-related liver diseases comparable to those obtained with the standarddoses of ribavirin 1000 - 1200 mg/*** prospective controlled trials using lower total amounts of ribavirin in combination with interferon should be p

Automated ex-situ detection of pitting corrosion and its effect on the mechanical integrity of rare earth magnesium alloy-WE43
