Microbial diversity in Huguangyan Maar Lake of China revealed by high-throughput sequencing

Huguangyan Maar Lake is a typical maar lake in the southeast of China. It is well preserved and not disturbed by anthropogenic activities. In this study, microbial community structures in sediment and water samples from Huguangyan Maar Lake were investigated using a high-throughput sequencing method. We found significant differences between the microbial community compositions of the water and the sediment. The sediment samples contained more diverse Bacteria and Archaea than did the water samples. Actinobacteria, Betaproteobacteria, Cyanobacteria, and Deltaproteobacteria predominated in the water samples while Deltaproteobacteria, Anaerolineae, Nitrospira, and Dehalococcoidia were the major bacterial groups in the sediment. As for Archaea, Woesearchaeota (DHVEG-6), unclassified Archaea, and Deep Sea Euryarchaeotic Group were detected at higher abundances in the water, whereas the Miscellaneous Crenarchaeotic Group, Thermoplasmata, and Methanomicrobia were significantly more abundant in the sediment. Interactions between Bacteria and Archaea were common in both the water column and the sediment. The concentrations of major nutrients (NO^3-, PO4^3-, SiO3^2- and NH4^+) shaped the microbial population structures in the water. At the higher phylogenetic levels including phylum and class, many of the dominant groups were those that were also abundant in other lakes;however, novel microbial populations (unclassified) were often seen at the lower phylogenetic levels. Our study lays a foundation for examining microbial biogeochemical cycling in sequestered lakes or reservoirs.

Effects of Temperature and Hydraulic Residence Time (HRT) on Treatment of Dilute Wastewater in a Carrier Anaerobic Baffled Reactor

Objective To examine the effect of hydraulic residence time （HRT） on the performance and stability, to treat dilute wastewater at different operational temperatures in a carrier anaerobic baffled reactor （CABR）, and hence to gain a deeper insight into microbial responses to hydraulic shocks on the base of the relationships among macroscopic performance, catabolic intermediate, and microcosmic alternation. Methods COD, VFAs, and microbial activity were detected with constant feed strength （300 mg/L） at different HRTs （9-18 h） and temperatures （10℃-28℃） in a CABR. Results The removal efficiencies declined with the decreases of HRTs and temperatures. However, the COD removal load was still higher at short HRT than at long HRT. Devastating reactor performance happened at temperature of 10℃ and at HRT of 9 h. HRTs had effect on the VFAs in the reactor slightly both at high and low temperatures, but the reasons differed from each other. Microbial activity was sensitive to indicate changes of environmental and operational parameters in the reactor. Conclusion The CABR offers to certain extent an application to treat dilute wastewater under a hydraulic-shock at temperatures from 10℃to 28℃.

Impacts of water surface area of watershed on design flood

In order to analyze the impact of the water surface area of a watershed on the design flood, the watershed was classified into a land watershed and a water surface watershed for flood flow calculation at the same time interval. Then, the design flood of the whole watershed was obtained by adding the two flood flows together. Using this method, we calculated design floods with different water surface areas of three reservoirs and analyzed the impact of water surface area on the flood volume and peak flow. The results indicate that larger water surface areas lead to greater impacts on the flood volume and peak flow. For the same watershed area, the impact of water surface area on the flood volume and peak flow is positively proportional to the flood frequency, i.e., the higher the frequency, the greater the impact becomes.

Mpox is an infectious and contagious zoonotic disease caused by the mpox virus(MPXV),which belongs to the genus *** 2022,MPXV has posed a signiﬁcant threat to global public *** emergence of thousands of cases across the Western Hemisphere prompted the World Health Organization to declare an *** extensive coevolutionary history of poxviruses with humans has enabled these viruses to develop sophisticated mechanisms to counter the human immune ***ﬁcally,MPXV employs unique immune evasion strategies against a wide range of immunological elements,presenting a considerable challenge for treatment,especially following the discontinuation of routine smallpox vaccination among the general *** this review,we start by discussing the entry of the mpox virus and the onset of early infection,followed by an introduction to the mechanisms by which the mpox virus can evade the innate and adaptive immune *** caspase-1 inhibitory proteins and a PKR escape-related protein have been identiﬁed as phylogenomic hubs involved in modulating the immune environment during the MPXV *** respect to adaptive immunity,mpox viruses exhibit unique and exceptional T-cell inhibition capabilities,thereby comprehensively remodeling the host immune *** viral envelope also poses challenges for the neutralizing effects of antibodies and the complement *** unique immune evasion mechanisms employed by MPXV make novel multi-epitope and nucleic acid-based vaccines highly promising research directions worth ***,we brieﬂy discuss the impact of MPXV infection on immunosuppressed patients and the current status of MPXV vaccine *** review may provide valuable information for the development of new immunological treatments for mpox.

Regional difference and dynamic mechanism of locality of the Chinese farming-pastoral ecotone based on geotagged photos from Panoramio

Cross-regional locality research reflects the influences of natural environment and the human activities due to the abundant land types and the multiple landscape combinations in related regions. The Chinese farming-pastoral ecotone is a typical large-scale region but few studies were conducted. This research contributed to the understanding of cross-regional locality of the Chinese farming-pastoral ecotone from different scales, including national, sectional, and provincial administrative units by utilizing geotagged photos（GTPs） obtained from the Panoramio website. The major results were as follows：（1） the locality elements of the Chinese farming-pastoral ecotone included 52 free nodes classified into 8 types of scene attributes;（2） there were huge differences between locality elements of different regions, and there was a negative correlation between the similarity degree of elements of different provinces and their spatial distances;（3） the Chinese farming-pastoral ecotone could be divided into the northern, central and southern sections, whose localities had differences in element constitution, association structure and the strength of elements, system stability and the anti-interference capability; and（4） the evolution of the localities of the northern and central sections was mainly influenced by human activities, while the locality of southern section retained more natural features. On a theoretical level, this research aimed to establish the research methodology of locality from the perspective of open data on the web with strong operability and replicability. On a practical level, this research could enrich the structuring recognition of the locality of the Chinese farming-pastoral ecotone and the comprehension of its dynamic mechanism. The results provide a reference for locality differentiation protection and the development of a cross-regional scale.

Effects of Phenylacetate on Cell Proliferation and Homeobox Genes Expression in the HCT-8 Colorectal Carcinoma Cell Line

To study the effects of phenylacetate （PA） on cell proliferation and homeobox （HOX） genes expression in the colorectal carcinoma HCT-8 cell line, HCT-8 cells were grown in the presence or absence of PA. The cellular proliferation inhibition was evaluated by the MTT assay. Twenty-two HOX genes were divided into three groups （ P1, P2, P3） according to their primer sequences, and the samples of cells were analyzed for the HOX genes＇ mRNA expression by means of the semi-quantitative reverse transcription-polymerase chain reaction （RT-PCR）. The level of the HOX genes＇ expression was expressed as the ratio expression rate of HOX gene to the β-actin. HCT-8 cells were treated with 1.0-5.0 mmol/L PA for 24-72 h. With the increase of the PA concentration or the prolongation of the treating time, the cell proliferation is inhibited in a dose- and time-dependent manner. The P1 group mRNA＊ expression（0. 5781 ±0. 0836） is significantly lower than that of the untreated group （0. 7701 ± 0. 0883 ） in HCT-8 cells （p 〈 0. 001 ）. Both the mRNA expressions of groups P2 （0. 3941 ± 0. 0819） and P3 （0. 5601 ± 0. 0736） in the PA treated group are significantly higher than those of the untreated groups P2（0. 1221±0. 0782） and P3 （0. 1806 ± 0. 0811 ） in HCT-8 cells（p 〈 0. 001）. PA could effectively inhibit cell proliferation by regulating the HOX genes expression and the mechanisms of the PA action are correlated with the transcription process in HCT-8 cells.

Implications for Cultural Landscape in a Chinese Context: Geo-analysis of Spatial Distribution of Historic Sites

The protection of historic sites, especially their relationship with urban development, has become a worldwide issue, both in developed and developing countries. In the context of rapid urbanization in China, the realistic compatibility between urban construction and the protection of historic sites is always a key research topic. In this study, first, to comprehend their spatial distribution patterns, 828 historic sites throughout the country are selected based on certain criteria. Then, we conduct quantitative research using GIS software, adopting indicators that include Nearest Neighbor Index, Gini Coefficient, and Geographic Concentration Index to analyze the spatial characteristics of historic sites on the three levels of city, province and nation. The results indicate that the spatial distribution of the different types of historic sites is an agglomeration on the nationwide scale, most of which is located in the regions of the Pearl(Zhujiang) River Delta, Yangtze(Changjiang) River Delta and Beijing-Tianjin Region. Because the majority of historic sites are located within approximately 10 km of the downtown area, a certain pattern has emerged, showing that the larger cities own more historic areas, which are in a more incomplete state of preservation, indicating the fragmentation of heritage spaces. The formation mechanism of the historic sites' distribution pattern is based on the conditions of the cities/towns as well as the bid-rent theory.

Through-Wall Tracking of Human Movers Using Continuous-Wave Radar

An intelligent frequency fitting algorithm is presented for continuous-wave radar to track human movers through walls. With the proper fitting dimension,which is usually determined by the practical detection requirements, this technique can improve the localization accuracy and the tracking performance without introducing too much calculation burden, and more precisely identify different targets even in the frequency ambiguous *** technology can also help to match the estimation results to their corresponding targets automatically, which is of great importance to certain urban sensing applications,such as specific target surveillance and tracking. To further improve the detection performance and better identify the weak targets in the presence of strong noise, CLEAN algorithm and adaptive filter technology are also *** results are provided to illustrate the algorithm performance.
