Bronchus-associated lymphoid tissue(BALT)develops at unpredictable locations around lung bronchi following pulmonary *** formation and composition of BALT have primarily been investigated by immunohistology that,due to the size of the invested organ,is usually restricted to a limited number of histological *** assess the entire BALT of the lung,other approaches are urgently ***,we introduce a novel light sheet microscopy-based approach for assessing lymphoid tissue in the *** antibody staining of whole lung lobes and optical clearing by organic solvents,we present a method that allows in-depth visualization of the entire bronchial tree,the lymphatic vasculature and the immune cell composition of the induced ***,three-dimensional analysis of the entire lung allows the qualitative and quantitative enumeration of the induced *** this approach,we show that a single intranasal application of the replication-deficient poxvirus MVA induces BALT that constitutes up to 8%of the entire lung volume in mice deficient in CCR7,in contrast to wild type mice(WT).furthermore,BALT induced by heat-inactivated *** is dominated by a pronounced T cell infiltration in Cxcr5-deficient mice,in contrast to WT mice.

Liver expression of steroid hormones and Apolipoprotein D receptors in hepatocellular carcinoma

AIM: To evaluate the tissular expression of Androgen (A), Estrogen (E) and Progesterone (Pg) receptors, and Apolipoprotein D (ApoD), in liver tumors from resected hepatocellular carcinoma (HCC) cases in order to assess their possible relationship to prognosis. METHODS: We performed an immunohistochemical study using tissue microarrays (containing more than 260 cancer specimens, from 31 HCC patients and controls) to determine the presence of specif ic antibodies against AR, ER, PgR and ApoD, correlating their findings with several clinico-pathological and biological variables. The staining results were categorized using a semi-quantitive score based on their intensity, and the percentage of immunostained cells was measured. RESULTS: A total of 21 liver tumors (67.7%) were positive for AR; 16 (51.6%) for ER; 26 (83.9%) for PgR and 12 (38.7%) stained for ApoD. We have found a wide variability in the immunostaining score values for each protein, with a median (range) of 11.5 (11.5-229.5) for AR; 11.1 (8.5-65) for ER; 14.2 (4-61) for PgR; and 37.7 (13.8-81.1) for ApoD. A history of heavy ethanol consumption, correlated positively with AR and PgR and negatively with ER status. HCV chronic infection also correlated positively with AR and PgR status. However, the presence of ApoD immunostaining did not correlate with any of these variables. Tumors with a positive immuno-staining for PgR showed a better prognosis. CONCLUSION: Our results indicate a moderate clinical value of the steroid receptor status in HCC, emphasizing the need to perform further studies in order to evaluate the possible role of new hormonal-based therapies.

The constant emergence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)variants indicates the evolution and adaptation of the *** innate immune evasion through increased expression of viral antagonist proteins,including ORf9b,contributes to the improved transmission of the Alpha variant;hence,more attention should be paid to these viral ***9b is an accessory protein that suppresses innate immunity via a monomer conformation by binding to ***,we solved the dimeric structure of SARS-CoV-2 ORf9b with a long hydrophobic tunnel containing a lipid molecule that is crucial for the dimeric conformation and determined the specific lipid ligands as monoglycerides by conducting a liquid chromatography with tandem mass spectrometry analysis,suggesting an important role in the viral life ***,a long intertwined loop accessible for host factor binding was observed in the *** phosphorylated residues in ORf9b were identified,and residues S50 and S53 were found to contribute to the stabilization of dimeric ***,we proposed a model of multifunctional ORf9b with a distinct conformation,suggesting that ORf9b is a fold-switching protein,while both lipids and phosphorylation contribute to the ***,the ORf9b monomer interacts with Tom70 to suppress the innate immune response,whereas the ORf9b dimer binds to the membrane involving mature virion *** results provide a better understanding of the multiple functions of ORf9b.

Plant communities and reproductive phenology in mountainous regions of northern Libya

Within the semi-desert landscape of northern Libya, two sub-humid escarpments occur: Al-Akhdar in the east and Nafusa (Jabal Al-Gharbi) in the west. This study compares plant communities in the two regions, which are along an elevation gradient, in terms of species composition and diversity, frequency of different Raunkiaer life forms, and reproductive phenology. The two regions differed in species composition and life-form frequency between regions and between elevation zones within each region. Patterns were associated with the lower rainfall and lower moisture-holding capacity of soils at Nafusa, resulting in more xeric conditions. Only 13% of species were shared between the two regional landscapes. Species diversity, life-form frequency, and duration of the flowering-fruiting phenophase were all affected by elevation above sea level. The duration of flowering and fruiting in spring and fall was associated with environmental conditions, although there were different thresholds in the two regions. There was both a spring and fall episode of flowering at Nafusa, but only spring flowering at Al-Akhdar. It is anticipated that there will be a gradual shift of plant communities to higher elevations and loss of certain sensitive species in response to ongoing climate change.

Concordance of non-invasive mechanical and serum tests for liver fibrosis evaluation in chronic hepatitis C

AIM To determine the sensitivity and specificity of liver stiffness measurement(LSM) and serum markers(SM) for liver fibrosis evaluation in chronic hepatitis *** Between 2012 and 2014,81 consecutive hepatitis C virus(HCV) patients had METAVIR score from liver biopsy compared with concurrent results from LSM [transient elastography(TE) [fibroS can~/ARfI technology(Virtual Touch~)] and SM [fIB-4/aspartate aminotransferase-toplatelet ratio index(APRI)].The diagnostic performance of these tests was assessed using receiver operating characteristic *** optimal cut-off levels of each test were chosen to define fibrosis stages f ≥ 2,f ≥ 3 and f = *** Kappa index set the concordance *** fifty point six percent were female and the median age was 51 years(30-78).fifty-six patients(70%) weretreatment-na?*** optimal cut-off values for predicting f ≥ 2 stage fibrosis assessed by TE were 6.6 kP a,for acoustic radiation force impulse(ARfI) 1.22 m/s,for APRI 0.75 and for fIB-4 *** f ≥ 3 TE was 8.9 kP a,ARfI was 1.48 m/s,APRI was 0.75,and fIB-4 was *** f = 4,TE was 12.2 kP a,ARfI was 1.77 m/s,APRI was 1.46,and fIB-4 was *** APRI could not distinguish between f2 and f3,P = *** negative predictive value for f = 4 for TE and ARfI was 100%.Kappa index values for f ≥ 3 METAVIR score for TE,ARfI and fIB-4 were 0.687,0.606 and 0.654,*** demonstrates strong concordance between all three screening methods,and moderate to strong concordance between them and APRI(Kappa index = 0.507).CONCLUSION Given the costs and accessibility of LSM methods,and the similarity with the outcomes of SM,we suggest that fIB-4 as well as TE and ARfI may be useful indicators of the degree of liver *** is of particular importance to developing countries.

Voltammetric sensor based on cobalt-poly(methionine)-modified glassy carbon electrode for determination of estriol hormone in pharmaceuticals and urine

A voltammetric sensor based on the electropolymerization of cobalt-poly(methionine)(Co-poly(Met)) on a glassy carbon electrode (GCE) was developed and applied for the determination of estriol by differential pulse voltammetry (DPV) for the first time. The electrochemical properties of the Co-poly(Met)/GCE were analysed by cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS) were used to characterize the polymers on the GCE surface. The deposition of the Co-poly(Met) film on the GCE surface enhanced the sensor electronic transfer. CV studies revealed that estriol exhibits an irreversible oxidation peak at t0.58 V for the Co-poly(Met)/GCE (vs. Ag/AgCl reference electrode) in 0.10 mol/L Britton-Robinson buffer solution (pH=7.0). Different voltammetric scan rates (10-200 mV/s) suggested that the estriol oxidation on the Co-poly(Met)/GCE surface is controlled by adsorption and diffusion processes. Based on the optimized DPV conditions, the linear responses for estriol quantification were from 0.596 μmol/L to 4.76 μmol/L (R2 =0.996) and from 5.66 μmol/L to 9.90 μmol/L (R2 =0.994) with a limit of detection (LOD) of 0.0340 μmol/L and a limit of quantification (LOQ) of 0.113 μmol/L. The DPV-Co-poly(Met)/GCE method provided good intra-day and inter-day repeatability with RSD values lower than 5%. Also, no interference of real sample matrices was observed on the estriol voltammetric response, making the DPV-Copoly( Met)/GCE highly selective for estriol. The accuracy test showed that the estriol recovery was in the ranges 96.7%-103% and 98.7%-102% for pharmaceutical tablets and human urine, respectively. The estriol quantification in pharmaceutical tablets performed by the Co-poly(Met)/GCE-assisted DPV method was comparable to the official analytical protocols.

Phylogenetically Distinct Cellulose Synthase Genes Support Secondary Wall Thickening in Arabidopsis Shoot Trichomes and Cotton fiber

Through exploring potential analogies between cotton seed trichomes （or cotton fiber） and arabidopsis shoot trichomes we discovered that CesAs from either the primary or secondary wall phylogenetic clades can support secondary wall thickening. CesA genes that typically support primary wall synthesis, AtCesA 1,2,3,5, and 6, underpin expansion and secondary wall thickening of arabidopsis shoot trichomes. In contrast, apparent orthologs of CesA genes that support secondary wall synthesis in arabidopsis xylem, AtCesA4,7, and 8, are up-regulated for cotton fiber secondary wall deposition. These conclusions arose from：（a） analyzing the expression of CesA genes in arabidopsis shoot trichomes; （b） observing birefringent secondary walls in arabidopsis shoot trichomes with mutations in AtCesA4, 7, or 8; （c） assaying up-regulated genes during different stages of cotton fiber development; and （d） comparing genes that were *** with primary or secondary wall CesAs in arabidopsis with genes up- regulated in arabidopsis trichomes, arabidopsis secondary xylem, or cotton fiber during primary or secondary wall deposition. Cumulatively, the data show that：（a） the xylem of arabidopsis provides the best model for secondary wall cellulose synthesis in cotton fiber; and （b） CesA genes within a ＂cell wall toolbox＂ are used in diverse ways for the construction of particular specialized cell walls.

Effects of Basella alba and Hibiscus macranthus extracts on testosterone production of adult rat and bull Leydig cells

Aim： To determine the androgenic effects of Basella alba and Hibiscus macranthus extracts in the rat and the bull, and to develop a novel in vitro test system using Leydig cells from bull testes. Methods： The effect of methanol extracts from both plants on testosterone production in isolated Leydig cells from the rat and the bull was analyzed using ^125I-radioimmunoassay （^125I-RIA）. Rat Leydig cells were obtained by common methods, whereas a novel technique was used to purify Leydig cells from bull testes. Results： Bull testes from the slaughter house were a cheap source of pure Leydig cells. In culture, these cells produced testosterone for 5-6 days, which can be stimulated by human chorionic gonadotrophin （hCG）. Basella alba extracts significantly enhanced testosterone production in bull and rat Leydig cells in a concentration-dependent manner. Hibiscus macranthus showed no androgenic effect but was shown to inhibit testosterone production at higher concentrations. Conclusion： Leydig cells purified from bull testes can be used as an alternative tool in experimental animal research. Certain fractions of Basella alba extract demonstrated androgenic potential whereas Hibiscus macranthus extracts did not.
