The interaction of calf thymus DNA（ctDNA） with berberine（Ber） was systematically investigated via UV-Vis,fluorescence and phosphorescence spectroscopies in aqueous solution and on solid *** very weak fluorescence intensity of Ber at 525 nm in aqueous solution can be greatly enhanced by *** UV-Vis spectrum shows that Ber could bind with DNA by *** addition,strong fluorescence of Ber alone could be observed on filter paper substrate and can be quenched by ctDNA effectively,and the increase of fluorescence polarization of Ber on the filter paper implies the intercalation *** emits phosphorescence at 619 nm in the presence of thallium（I） *** lifetime of Ber increased from 38.4 ms to 43.4 ms with the increase of *** increase of lifetime is possibly attributed to the increase of rigidity of Ber after its intercalating into the ctDNA base *** explanation is also augmented by the anionic quenching *** behaves as a typical intercalating agent into *** intrinsic binding constant between Ber and ctDNA is（1.84±0.12）×10^4 L/mol and n=0.16.

AIM: To evaluate double balloon enteroscopy (DBE) in post-surgical patients to perform endoscopic retrograde cholangiopancreatography (ERCP) and interventions. METHODS: In 37 post-surgical patients, a stepwise approach was performed to reach normal papilla or enteral anastomoses of the biliary tract/pancreas. When conventional endoscopy failed, DBE-based ERCP was performed and standard parameters for DBE, ERCP and interventions were recorded. RESULTS: Push-enteroscopy (overall, 16 procedures) reached enteral anastomoses only in six out of 37 post-surgical patients (16.2%). DBE achieved a high rate of luminal access to the biliary tract in 23 of the remaining 31 patients (74.1%) and to the pancreatic duct (three patients). Among all DBE-based ERCPs (86 procedures), 21/23 patients (91.3%) were successfully treated. Interventions included ostium incision or papillotomy in 6/23 (26%) and 7/23 patients (30.4%), respectively. Biliary endoprosthesis insertion and regular exchange was achieved in 17/23 (73.9%) and 7/23 patients (30.4%), respectively. furthermore, bile duct stone extraction as well as ostium and papillary dilation were performed in 5/23 (21.7%) and 3/23 patients (13.0%), respectively. Complications during DBE-based procedures were bleeding (1.1%), perforation (2.3%) and pancreatitis (2.3%), and minor complications occurred in up to 19.1%. CONCLUSION: The appropriate use of DBE yields a high rate of luminal access to papilla or enteral anastomoses in more than two-thirds of post-surgical patients, allowing important successful endoscopic therapeutic interventions.

Integrated futures for Europe’s Mountain Regions: Reconciling Biodiversity Conservation and Human Livelihoods

Europe’s mountains cover nearly half of the continent’s area and are home to one fifth of the European population. Mountain areas are hotspots of biodiversity and agriculture has played a multifunc- tional role in defining and sustaining mountain biodiversity. Ongoing trends of agricultural decline are having negative impacts on mountain biodiversity. This paper presents results from an interdisciplinary European research project, BioScene, which investi- gated the relationship between agriculture and biodi- versity in six mountain study areas across Europe to provide recommendations for reconciling biodiversity conservation with social and economic activities through an integrated rural development strategy. BioScene used scenario analysis and stakeholder participation as tools for structuring the analysis of alternative mountain futures. Three main BioScene scenarios were evaluated: Business as Usual (BaU), Agricultural Liberalisation (Lib), Managed Change forBiodiversity (MCB). BioScene brought together ecologists, economists, sociologists and rural geogra- phers, to carry out interdisciplinary analysis of the scenarios: identifying key drivers of change, assessing the biodiversity consequences and evaluating cost- effectiveness. BioScene used a sustainability assess- ment to integrate the research outputs across natural and social science disciplines to assess the broader sustainability of the scenarios in terms of biodiversity, natural resources, rural development, social devel- opment, economic development and institutional capacity. The sustainability assessment showed that the MCB scenario was potentially the most sustain- able of the three BioScene scenarios. Through the reconciliation of potentially conflicting objectives, such as conservation, economic development and human livelihoods, and with a strong participatory planning approach, the MCB scenario could represent an alternative approach to BaU for sustainable rural development in Europe’s mountains.

The imbalance between pathogenic and protective T cell subsets is a cardinal feature of autoimmune disorders such as multiple sclerosis(MS).Emerging evidence indicates that endogenous and dietary-induced changes in fatty acid metabolism have a major impact on both T cell fate and *** date,however,the molecular mechanisms that underlie the impact of fatty acid metabolism on T cell physiology and autoimmunity remain poorly ***,we report that stearoyl-CoA desaturase-1(SCD1),an enzyme essential for the desaturation of fatty acids and highly regulated by dietary factors,acts as an endogenous brake on regulatory T-cell(Treg)differentiation and augments autoimmunity in an animal model of MS in a T cell-dependent *** by RNA sequencing and lipidomics analysis,we found that the absence of Scd1 in T cells promotes the hydrolysis of triglycerides and phosphatidylcholine through adipose triglyceride lipase(ATGL).ATGL-dependent release of docosahexaenoic acid enhanced Treg differentiation by activating the nuclear receptor peroxisome proliferator-activated receptor *** findings identify fatty acid desaturation by SCD1 as an essential determinant of Treg differentiation and autoimmunity,with potentially broad implications for the development of novel therapeutic strategies and dietary interventions for autoimmune disorders such as MS.

Comparative Study of the Physicochemical and Microbiological Quality of Water from Wells, Boreholes and Springs Consumed in the Massissia District in Brazzaville, Republic of the Congo

The Republic of Congo in the course of the last two decades has been marked by a strong demographic explosion. The increase of its population has been accompanied by a significant demand for drinking water. The Congolese populations in order to meet their water needs, have recourse to rainwater, surface water and groundwater, because it is easy to access. The city of Brazzaville, political capital of the Republic of Congo, is also affected by this problem. The study carried out here aimed to assess the physicochemical and microbiological parameters of groundwater (wells, boreholes and springs) consumed by the populations of the Massissia district. In this work the physicochemical parameters below were evaluated: the pH, the conductivity, the hardness, the major ions, the undesirable substances as well as some toxic elements (Lead, Cadmium, Iron and Zinc) thanks to techniques of colorimetry, potentiometry and spectrophotometry. The main microbiological parameters determined are E. Coli, Staphylococcus, Total Germs, Total Coliforms, Yeasts and Molds. The physicochemical analyzes revealed that these waters are acidic with pH values between 5 and 6.5, the nitrate concentration evaluated between 50 mg/L and 60 mg/L, and a presence of the element Cadmium at values variants between 0.01 and 0.5 mg/L, values greater than the standard required by the WHO. Microbiological analyzes revealed bacteriological contamination by presence of E. coli in wells and boreholes whose origin is faecal contamination, the presence of yeasts and molds, staphylococcus, Total germs and Total coliforms and Ps Aeuginosis in all water points, making these waters a real danger for consumers.
