Objective:To examine the potent of water as a solvent agent in the preparation of traditional herbal ***:Water extracts of 18 plants were prepared through reflux and examined(25μg/mL) to evaluate their possibility for inhibiting protein tyrosine phosphatase 1B(PTP1B).The determination of IC50 values was performed for the samples possessing more than 80%***,those exhibiting IC50 values more than 7.0 μg/mL were further profiled for their chemical constituents through nuclear magnetic resonance(NMR) ***:About 44%(8) of the examined samples showed more than 80%inhibition against PTP1 *** water extracts of Elephantopus scaber,Helicteres isora aerial parts,Elaeocarpus grandiflorus(***) fruits,Melaleuca leucadendron leaves,and Quercus infectoria gum had IC50 values ranging from 2.05 to 6.90 μg/***,Andropogon nardus and Centella asiatica were at the area of δ 3.0-4.0 ***,the13 C NMR observation of samples possessing the most intensive signals in their proton NMR Cinnamomum burmannii and *** showed the peaks at the area of δ 60-90 ppm as the supportive evidence for sugar group ***,a disaccharide from *** could be an active inhibitor towards PTB1 ***:In contrast to the mainstream solvents currently used in modern herbal manufactures especially Jamu medicine in Indonesia,pure-water-extracted materials should be reconsidered and could be reemerged for future studies and for the manufacture of herbal *** addition,the activity of Jamu components should be confirmed that their antidiabetes and antiobesity activities could be through the inhibition of PTP1 B.

Accuracy of two optical chlorophyll meters in predicting chemical composition and in vitro ruminal organic matter degradability of Brachiaria hybrid,Megathyrsus maximus, and Paspalum atratum

The objective of this study was to determine the accuracy and reliability of 2 optical chlorophyll meters:fieldScout CM 1,000 NDVI and Yara N-Tester, in predicting neutral detergent fibre(NDf), acid detergent fibre(ADf), acid detergent lignin(ADL), acid detergent insoluble nitrogen(ADIN) and in vitro ruminal organic matter degradability(IVOMD) of 3 tropical grasses. Optical chlorophyll measurements were taken at 3 stages(4, 8 and 12 weeks) of regrowth in Brachiaria hybrid, and Megathyrsus maximus and at 6 and 12 weeks of regrowth in Paspalum atratum(cv. Ubon). Optical chlorophyll measurements showed the highest correlation(r = 0.57 to 0.85) with NDf concentration. The fieldScout CM 1,000 NDVI was better than the Yara N-Tester in predicting NDf(R^2 = 0.70) and ADf(R^2 = 0.79) concentrations in Brachiaria hybrid and NDf(R^2 = 0.79) in M. maximus. Similarly, fieldScout CM 1,000 NDVI produced better estimates of 24 h IVOMD(IVOMD_(24 h)) in Brachiaria hybrid(R^2 = 0.81) and IVOMD_(48 h) in Brachiaria hybrid(R^2 = 0.65) and M. maximus(R^2 = 0.75). However, these prediction models had relatively low concordance correlation coefficients, i.e., CCC >0.90, but random errors were the main source of bias. It was,therefore, concluded that both optical chlorophyll meters were poor and unreliable predictors of ADIN and ADL concentrations. Overall, the fieldScout CM 1,000 NDVI shows potential to produce useful estimates of IVOMD_(24h) and ADf in Brachiaria hybrid and IVOMD_(48 h) and NDf concentrations in M. maximus.

The Relationship between Salinity and Bacterioplankton in Three Relic Coastal Ponds (Macchiatonda Wetland, Italy)

The great environmental importance of wetlands is linked to the high biodiversity of flora and fauna they support, so that the international Ramsar Convention focused on these areas and highlighted the need to preserve them. The bacterial communities that thrive in these ecosystems play a key role in regulating the local biogeochemical processes and yet their distribution, abundance and dynamics are poorly known. This work is aimed to study the bacterial assemblages over a year long, to contribute to the understanding of the natural processes occurring in wetlands at variable salinity. The knowledge of bacterial groups, species or assemblages can provide a useful bioindicator for conservation and restoration efforts. Macchiatonda Natural Reserve (Santa Severa, Rome, Italy) is a relic ecosystem, once found along the entire Tyrrhenian coast. This wetland encompasses three coastal ponds with different salinity, where both peculiar vegetation and highly diverse migratory and resident avifauna can be found. This ancient system has been scarcely investigated and nothing is known about its microbial community. The molecular metagenomic analyses performed to investigate the salinity/bacterioplankton relationship, highlighted differences in the bacterial structure, between ponds and seasons. Analogous trends in SSCP profiles, Shannon Index, and bacterial composition (16S) were observed in the two saltier ponds, whereas the entire set of results was different for the less salty one. The species diversity in the three ponds varied according the salinity gradient, with the maximum diversity corresponding to a salt concentration range between 20 and 30. At higher and lower salinity, the microbial diversity lowers, according to the “Intermediate Disturbance Hypothesis”.

Polyethylene glycol (PEG), a phenol binding agent has been used extensively to measure the biological activity of tannins in forage species. The optimum inclusion rate of PEG, per unit weight of sample varies from species to species. Determining optimum inclusion levels can prevent wastage and reduce the cost of diagnosing the biological activity of tannins, especially in developing countries. This study was designed to determine the optimum PEG inclusion levels required to completely ameliorate In vitro ruminal bioactivity of tannins in leaves from Leucaena leucocephala, Gliricidia sepium and Trichanthera gigantea using the Reading Pressure Technique. fermentation parameters were generated by fitting gas production data to the Orskov and McDonald (1979) non-linear equation: . An asymptotic response to incremental levels of PEG was observed with cumulative gas production at 48 h post inoculation. The minimum level of PEG required to maximize In vitro ruminal fermentation of tree leaves was found to be 200 mg PEG/g DM for all tree species. Gas production rate constant for the insoluble fraction (c) showed an increase (P In vitro organic matter degradability (iOMD) declined (P In vitro ruminal tannin biological activity in leaves of the three tree species.

In Vitro and In Silico Studies of Lunacridine from Lunasia Amara Blanco as Anticancer

Lunasia amara Blanco is a famous plant in South Sulawesi. It was used largely by local people as antibacteria and aphrodisiac. Quinoline alkaloid lunacridine was known as the active principle from Lunasia amara Blanco. Its activity was reported as a DNA Intercalating Topoisomerase II inhibitor. In this study, we have isolated and assayed the cytotoxic activity of lunacridine on P388 murine leukemia cells by MTT colorimetric assay (in vitro). Lunacridine showed the less cytotoxic activity with the IC50 of 39.52 μg/mL. With the aim to explore the structural determinants responsible for this activity, molecular docking study have been carried out with DNA model using AutoDock 4.0 software with various total of energy evaluations (in silico). The best docking reached at the total energy evaluations of 2.5 × 107 with the binding free energy of -6.63 kcal/mol. Analysis of the docking results was in accordance with the ability of lunacridine to intercalate between base pairs of DNA. Cytotoxic activity of lunacridine is less probably due to low affinity and molecular interaction. Therefore this study suggests to design and to develop lunacridine as a lead compound for anticancer drug.

背景I-gel喉罩是一种不带充气套囊的声门上通气道，其设计允许气管导管无障碍通过，且既往研究已表明其能与声门进行良好对位。在本前瞻性随机临床试验中，我们比较了经i-gel喉罩和LMA fastrach喉罩辅助进行气管插管盲插的成功率。方法将160例需行全身麻醉和气道管理的患者随机分为2组，每组分别进行经i-gd喉罩或LMAfastrach喉罩辅助的气管插管。全麻诱导后，每组患者置入相应喉罩并确认有效肺通气，随后进行气管导管盲插。观测气管插管次数并计算首次插管成功率和最终插管成功率。结果每组各入选80例患者。i-gel喉罩辅助插管组首次插管成功率为69％，而LMA fastrach喉罩辅助插管组为74％（差异的95％可信区间CI为-9％-19％，P=0．60）。l-gel喉罩组的最终插管成功率低于LMAfastrach喉罩组（分别为73％和91％，差异的95％CI为7％。31％，P〈0．0001）。结论经i-gd喉罩或LMA fastrach喉罩辅助气管导管盲插首次成功率相近，但首次插管失败后，i-gel喉罩并不提高后续插管成功率，且其最终插管成功率低于LMAfastrach喉罩。
