Liver expression of steroid hormones and Apolipoprotein D receptors in hepatocellular carcinoma

AIm: To evaluate the tissular expression of Androgen (A), Estrogen (E) and Progesterone (Pg) receptors, and Apolipoprotein D (ApoD), in liver tumors from resected hepatocellular carcinoma (HCC) cases in order to assess their possible relationship to prognosis. mETHODS: We performed an immunohistochemical study using tissue microarrays (containing more than 260 cancer specimens, from 31 HCC patients and controls) to determine the presence of specif ic antibodies against AR, ER, PgR and ApoD, correlating their findings with several clinico-pathological and biological variables. The staining results were categorized using a semi-quantitive score based on their intensity, and the percentage of immunostained cells was measured. RESULTS: A total of 21 liver tumors (67.7%) were positive for AR; 16 (51.6%) for ER; 26 (83.9%) for PgR and 12 (38.7%) stained for ApoD. We have found a wide variability in the immunostaining score values for each protein, with a median (range) of 11.5 (11.5-229.5) for AR; 11.1 (8.5-65) for ER; 14.2 (4-61) for PgR; and 37.7 (13.8-81.1) for ApoD. A history of heavy ethanol consumption, correlated positively with AR and PgR and negatively with ER status. HCV chronic infection also correlated positively with AR and PgR status. However, the presence of ApoD immunostaining did not correlate with any of these variables. Tumors with a positive immuno-staining for PgR showed a better prognosis. CONCLUSION: Our results indicate a moderate clinical value of the steroid receptor status in HCC, emphasizing the need to perform further studies in order to evaluate the possible role of new hormonal-based therapies.

Combining material characterization with single and multi-oxyanion adsorption for mechanistic study of chromate removal by cationic hydrogel

Cationic hydrogel with magnetic property was synthesized via radical polymerization and its removal capacity of chromate from contaminated water was found to be 200 mg/*** fourier transform infrared spectroscopy（fT-IR） study,the mechanism of chromate removal by hydrogel was found to be non-specific adsorption,mainly due to ion exchange,as evidenced by the positively charged functional group,trimethyl ammonium-N＋（CH3）3,in the *** were accordingly determined by testing di？erent oxyanion adsorption onto the *** results of the chromate adsorption experiments illustrated that the amount of chromate adsorbed was nearly equal to that of the chloride released from the hydrogel,which is part of the evidence for ion *** and multi-oxyanion adsorption experiments were also performed,and it was demonstrated that ion removal was species independent,but charge dependent,another characteristic of the ion exchange *** was found that the same Langmuir model can be applied to best fit the findings of single and multi-oxyanion adsorption,which further indicates the mechanism of chromate removal is attributed to ion *** view of the above,the background anions compete for adsorption sites with chromate,evidenced by inhibitive chromate removal in the presence of background electrolytes in the batch studies,further echoing the ion exchange mechanism.

A biosynthetic gene cluster for the bioactive fungal sesterterpenoids variecolin(1)and variecolactone(2)was identified in Aspergillus aculeatus ATCC *** production of 1 and 2 was achieved in Aspergillus oryzae by expressing the sesterterpene synthase VrcA and the cytochrome P450 ***,the replacement of VrcB with hom.logous P450s from other fungal terpenoid pathways yielded three new variecolin analogues(5-7).Analysis of the compounds’anticancer activity in vitro and in vivo revealed that although 5 and 1 had comparable activities,5 was associated with significantly reduced toxic side effects in cancer-bearing mice,indicating its potentially broader therapeutic *** study describes the first tests of variecolin and its analogues in animals and demonstrates the utility of synthetic biology for creating molecules with improved biological activities.

Seminal biomarkers for the evaluation of male infertility

for men struggling to conceive with their partners, diagnostic tools are limited and often consist of only a standard semen analysis. This baseline test serves as a crude estimation of male fertility, leaving patients and clinicians in need of additional diagnostic biomarkers. Seminal fluid contains the highest concentration of molecules from the male reproductive glands, therefore, this review focuses on current and novel seminal biomarkers in certain male infertility scenarios, including natural fertility, differentiating azoospermia etiologies, and predicting assisted reproductive technique success. Currently available tests include antisperm antibody assays, DNA fragmentation index, sperm fluorescence in situ hybridization, and other historical sperm functional tests. The poor diagnostic ability of current assays has led to continued efforts to find more predictive biomarkers. Emerging research in the fields of genomics, epigenetics, proteomics, transcriptomics, and metabolomics holds promise for the development of novel male infertility biomarkers. Seminal protein-based assays of TEXI01, ECm 1, and ACRV1 are already available or under final development for clinical use. Additional panels of DNA, RNA, proteins, or metabolites are being explored as we attempt to understand the pathophysiologic processes of male infertility. future ventures will need to continue data integration and validation for the development of clinically useful infertility biomarkers to aid in male infertility diagnosis, treatment, and counseling.

Numerical analysis and centrifugal modeling of LNAPLs transport in subsurface system

The transport of non-aqueous phase liquids (NAPLs) in unsaturated soils and groundwater is an important research topic in geo-environmental engineering. In this paper, the mechanism of light NAPLs (LNAPLs) transport in subsurface system is briefly introduced, and the mass transport equations, fluid flow equations, and the constitutive model of relative permeability - saturation - capillary pressure are discussed. Then the numerical method is introduced to simulate the multiphase flow problems in porous media, and the tempospatial distribution of LNAPLs is obtained. moreover, different boundary conditions are em.loyed in numerical simulation to investigate its effect on transport behavior. To verify the numerical data, centrifugal tests are conducted to model the LNAPLs migration in unsaturated soils and groundwater. The calculation results are agreeable with the experimental findings of centrifugal modeling, which indicates that LNAPLs from leaking point move downwards due to gravity force, and form a high concentration zone above the capillary fringe, and then spread out laterally along the groundwater table. Some LNAPL enters groundwater system to further migrate. The combination of numerical simulation and centrifuge modeling can be a useful means to study the transport behavior of LNAPLs in subsurface system.

Joostella strains are emerging candidates for biosurfactant production. Here such ability was analyzed for Joostella strain A8 in comparison with Alcanivorax strain A53 and Pseudomonas strain A6, all previously isolated from hydrocarbon enrichment cultures made of polychaete homogenates. In pure cultures Joostella sp. A8 showed the highest stable emulsion percentage（78.33%）, hydrophobicity rate（62.67%）, and an optimal surface tension reduction during growth in mineral medium supplemented with diesel oil（reduction of about 12 mN/m）, thus proving to be highly competitive with Alcanivorax and Pseudomonas strains. During growth in pure culture different level of biodegradation were detected for Alcanivorax strain A53（52.7%）,Pseudomonas strain A6（38.2%） and Joostella strain A8（26.8%）. When growing in consortia,isolates achieved similar abundance values, with the best efficiency that was observed for the Joostella-Pseudomonas co-culture. Gas-chromatographic analysis revealed an increase in the biodegradation efficiency in co-cultures（about 90%）, suggesting that the contemporary action of different bacterial species could improve the process. Results were useful to compare the efficiencies of well-known biosurfactant producers（i.e. Pseudomonas and Alcanivorax representatives） with a still unknown biosurfactant producer, i.e. Joostella, and to confirm them as optimal biosurfactant-producing candidates.

CO oxidation under lean and stoichiometric conditions over ceriazirconia with very low metal contents(Cu,Co,Ag and Pt)

In this work,several ceria-zirconia based catalysts with very lo w(and equimolar) metal contents were prepared,characterised and tested for the CO oxidatio n reaction(under lean and stoichiometric conditions),trying to emulate those conditions found in a diesel oxidation catalyst(DOC) system from a diesel engine and those encountered under gasoline exhaust(λ=1).The metals chosen are Cu,Co,Ag and Pt(as a reliable benchmark).The results reveal enormous differences among reducibility and catalytic activity despite quite similar structural and textural properties of the catalysts,showing differences among dispersion(Ag-catalyst seems to present a low level of dispersion).This catalyst seems to be characterised,as well,by a strong electronic interaction between Ce and Ag centres which is suggested to yield an improved reducibility under H_(2)-TPR ***,the order in catalytic activity(Cu>Ag>Co≈Pt>>support) seems not to follow the order found in reducibility and the Cu-catalyst seems to be the most active independently on the reaction conditions,yielding nearly overlapped CO oxidation catalytic *** restingly,a stro ng correlation between the catalytic activity under the two conditions tested and the OSC values of the Ce_(0.8)Zr_(0.2)O_(2)-supported metal catalysts is ***,OSC parameter measured at 150℃ can be used as a relevant descriptor to evaluate the CO oxidation activity at low and medium conversions for the investigated catalysts,much better than the H_(2)-TPR measurements.

Simultaneous removal of chromium and arsenate from contaminated groundwater by ferrous sulfate:Batch uptake behavior

Chromium and/or arsenate removal by fe（Ⅱ） as a function of pH, fe（Ⅱ） dosage and initial Cr（Ⅵ）/As（Ⅴ） ratio were examined in batch tests. The presence of arsenate reduced the removal efficiency of chromium by fe（Ⅱ）, while the presence of chromate significantly increased the removal efficiency of arsenate by fe（Ⅱ） at pH 6-8, In the absence of arsenate, chromium removal by fe（Ⅱ） increased to a maximum with increasing pH from 4 to 7 and then decreased with a further increase in pH. The increment in fe（Ⅱ） dosage resulted in an improvement in chromium removal and the improvement was more remarkable under alkaline conditions than that under acidic conditions. Chromium removal by fe（Ⅱ） was reduced to a larger extent under neutral and alkaline conditions than that under acidic conditions due to the presence of 10 μtmol/L arsenate. The presence of 20 μmol/L arsenate slightly improved chromium removal by fe（Ⅱ） at pH 3.9-5,8, but had detrimental effects at pH 6.7-9.8. Arsenate removal was improved significantly at pH 4-9 due to the presence of 10μmol/L chromate at fe（Ⅱ） dosages of 20-60 μmol/L. Elevating the chromate concentration from 10 to 20μmol/L resulted in a further improvement in arsenate removal at pH 4.0-4.6 when fe（Ⅱ） was dosed at 30-60 μmol/L.
