Interferon gamma boosts the nucleotide oligomerization domain 2-mediated signaling pathway in human dendritic cells in an X-linked inhibitor of apoptosis protein and mammalian target of rapamycin-dependent manner

The cytoplasmic nucleotide oligomerization domain 2 （NOD2） receptor recognizes the bacterial cell wall component muramyl dipeptide （MDP）. NOD2 ligation initiates the nuclear factor kappa B and the mitogen-activated protein kinase cascades. However, administering MDP alone is insufficient to elicit strong cytokine responses in various immune cells, including dendritic cells （DCs）. Because the simultaneous presence of various microbial products and cytokines in inflamed tissues modulates DC function, we initiated this study to examine how interferon gamma （IFNy）, a central modulator of inflammation, affects the NOD2-mediated signaling pathway in human conventional DCs （cDCs）. Synergistic stimulation of DCs with MDP and IFNy increased the expression of CD40, CD80, CD83, CD86, and human leukocyte antigen DQ proteins and significantly elevated the production of pro-inflammatory cytokines IL- 1β, IL-6, IL-12, and tumour necrosis factor （TNF）, as well as anti-inflammatory cytokine IL-10. Furthermore, the simultaneous presence of MDP and I FNy was necessary to decrease IkBe protein levels. By investigating various mechanisms implicated in MDP- and IFNγ-mediated signaling pathways, we revealed that the increased production of pro-inflammatory cytokines is highly dependent on the X-linked inhibitor of apoptosis protein （XIAP） but not on cellular lAP1 and lAP2. We also found that the NOD2 signaling pathway is regulated by the mammalian target of rapamycin （mTOR） but is not affected by phosphatidylinositol-3 kinase or signal transducer and activator of transcription 1 inhibition. Our results demonstrate, for the first time, that IFNy positively affects NOD2-mediated signaling in human cDCs, in a manner considerably dependent on XIAP and partially dependent on mTOR.

Photosystem Ⅱ （PSII） complexes drive the water-splitting reaction necessary to transform sunlight into chemical energy. However, too much light can damage and disrupt PSlh In cyanobacteria, the flv4-2 operon encodes three proteins （FIv2, FIv4, and S110218）, which safeguard PSll activity under air-level CO2 and in high light conditions. However, the exact mechanism of action of these proteins has not been clarified yet. We demonstrate that the PSII electron transfer properties are influenced by the flv4-2 operon-encoded pro- teins. Accelerated secondary charge separation kinetics was observed upon expression/overexpression of the flv4-2 operon. This is likely induced by docking of the FIv2/FIv4 heterodimer in the vicinity of the QB pocket of PSlI, which, in turn, increases the QB redox potential and consequently stabilizes forward electron transfer. The alternative electron transfer route constituted by FIv2/FIv4 sequesters electrons from QB- guaranteeing the dissipation of excess excitation energy in PSII under stressful conditions. In addition, we demonstrate that in the absence of the flv4-2 operon-encoded proteins, about 20% of the phycobilisome antenna becomes detached from the reaction centers, thus decreasing light harvesting. Phycobilisome detachment is a consequence of a decreased relative content of PSII dimers, a feature observed in the absence of the SII0218 protein.

Metabolic activity of Streptococcus mutans biofilms and gene expression during exposure to xylitol and sucrose

The objective of the study was to analyse Streptococcus mutans biofilms grown under different dietary conditions by using multifaceted methodological approaches to gain deeper insight into the cariogenic impact of carbohydrates. S. mutans biofilms were generated during a period of 24 h in the following media： Schaedler broth as a control medium containing endogenous glucose, Schaedler broth with an additional 5% sucrose, and Schaedler broth supplemented with 1% xylitol. The confocal laser scanning microscopy（CLSM）-based analyses of the microbial vitality, respiratory activity（5-cyano-2,3-ditolyl tetrazolium chloride, CTC） and production of extracellular polysaccharides（EPS） were performed separately in the inner, middle and outer biofilm layers. In addition to the microbiological sample testing, the glucose/sucrose consumption of the biofilm bacteria was quantified, and the expression of glucosyltransferases and other biofilm-associated genes was investigated. Xylitol exposure did not inhibit the viability of S. mutans biofilms, as monitored by the following experimental parameters： culture growth, vitality, CTC activity and EPS production. However,xylitol exposure caused a difference in gene expression compared to the control. Gtf C was upregulated only in the presence of *** xylitol exposure, gtf B was upregulated by a factor of 6, while under sucrose exposure, it was upregulated by a factor of *** with glucose and xylitol, sucrose increased cell vitality in all biofilm layers. In all nutrient media, the intrinsic glucose was almost completely consumed by the cells of the S. mutans biofilm within 24 h. After 24 h of biofilm formation, the multiparametric measurements showed that xylitol in the presence of glucose caused predominantly genotypic differences but did not induce metabolic differences compared to the control. Effect and feasibility of wearable physical activity trackers and pedometers for increasing physical activity and improving health outcomes in cancer survivors:A systematic review and meta-analysis

Purpose:This systematic review and meta-analysis aimed to evaluate the effect of wearable devices for improving physical activity and healthrelated outcomes in cancer ***:CINAHL,Cochrane,Ebscohost,MEDLINE,Pubmed,ProQuest Health and Medical Complete,ProQuest Nursing and Allied Health Source,ScienceDirect,and SPORTDiscus databases were searched for randomized controlled trials published before September 1,2020,that evaluated interventions involving wearable devices in cancer *** mean differences(SMDs)were calculated to assess effects on physical activity and health-related *** analyses were conducted to assess whether the effects differed by interventions and cancer *** of bias was assessed using the Cochrane risk of bias ***:Thirty-five trials were included(breast cancer,n=15,43%).Intervention durations ranged between 4 weeks and 1 *** trials(n=25,71%)involved pedometer-based physical activity ***(20%)involved Fitbit-based interventions,and 3(9%)involved other wearable physical activity trackers(e.g.,Polar,Garmin).Compared to usual care,wearable devices had moderate-to-large effects(SMD range 0.54-0.87,p<0.001)on moderate-intensity physical activity,moderate-to-vigorous-intensity physical activity,total physical activity,and daily *** to usual care,those in the intervention had higher quality of life,aerobic fitness,physical function,and reduced fatigue(SMD range=0.18-0.66,all p<0.05).Conclusion:Wearable physical activity trackers and pedometers are effective tools that increase physical activity and improve health-related outcomes in individuals with *** how these devices can be implemented for longer-term use with other intervention components remains an area for future research.

From variome to phenome:Pathogenesis,diagnosis and management of ectopic mineralization disorders

Ectopic mineralization- inappropriate biomineralization in soft tissues- is a frequent finding in physiological aging processes and several common disorders, which can be associated with significant morbidity and mortality. Further, pathologic mineralization is seen in several rare genetic disorders, which often present life-threatening phenotypes. These disorders are classified based on the mechanisms through which the mineralization occurs: metastatic or dystrophic calcification or ectopic ossification. Underlying mechanisms have been extensively studied, which resulted in several hypotheses regarding the etiology of mineralization in the extracellular matrix of soft tissue. These hypotheses include intracellular and extracellular mechanisms, such as the formation of matrix vesicles, aberrant osteogenic and chondrogenic signaling, apoptosis and oxidative stress. Though coherence between the different findings is not always clear, current insights have led to improvement of the diagnosis and management of ectopic mineralization patients, thus translating pathogenetic knowledge(variome) to the phenotype(phenome). In this review, we will focus on the clinical presentation, pathogenesis and management of primary genetic soft tissue mineralization disorders. As examples of dystrophic calcification disorders Pseudoxanthoma elasticum, Generalized arterial calcification of infancy, Keutel syndrome, Idiopathic basal ganglia calcification and Arterial calcification due to CD73(NT5E) deficiency will be discussed. Hyperphosphatemic familial tumoral calcinosis will be reviewed as an example of mineralization disorders caused by metastatic calcification.

CXC family of chemokines as prognostic or predictive biomarkers and possible drug targets in colorectal cancer

Colorectal cancer(CRC) is the third most common cancer in men and the second most common cancer in women,worldwide. In the early stages of the disease, biomarkers predicting early relapse would improve survival *** metastatic patients, the use of predictive biomarkers could potentially result in more personalized treatments and better outcomes. The CXC family of chemokines(CXCL1 to 17) are small(8 to 10 kDa) secreted proteins that attract neutrophils and lymphocytes. These chemokines signal through chemokine receptors(CXCR) 1 to *** studies have reported that these chemokines and receptors have a role in either the promotion or inhibition of cancer, depending on their capacity to suppress or stimulate the action of the immune system, *** general terms, activation of the CXCR1/CXCR2 pathway or the CXCR4/CXCR7 pathway is associated with tumor aggressiveness and poor prognosis; therefore,the specific inhibition of these receptors is a possible therapeutic strategy. On the other hand, the lesser known CXCR3 and CXCR5 axes are generally considered to be tumor suppressor signaling pathways, and their stimulation has been suggested as a way to fight *** pathways have been studied in tumor tissues(using immunohistochemistry or measuring mRNA levels)or serum [using enzyme-linked immuno sorbent assay(ELISA) or multiplexing techniques], among other sample types. Common variants in genes encoding for the CXC chemokines have also been investigated as possible biomarkers of the disease. This review summarizes themost recent findings on the role of CXC chemokines and their receptors in CRC and discusses their possible value as prognostic or predictive biomarkers as well as the possibility of targeting them as a therapeutic strategy.

Confounding response of macrofauna from a confluence of impacts: brine and sewage pollution

Throughout the shores of the world, a confluence of different sources of pollution is common, e.g., through outlets releasing brine, industrial, and domestic sewage. In this study, we assessed whether a combined disposal of domestic sewage and brine altered the patterns of abundance and assemblage structure of sub- tidal macrofauna inhabiting sandy seabeds on the south coast of Gran Canaria （Canary Islands, NE Atlantic Ocean）. Samples were collected in two surveys （May 2008 and January 2009） at three distances （0, 15, and 30 m） away from the discharge point. Macrofaunal abundances showed different patterns with varying proximity from the disposal point in the two surveys. In May 2008, lower abundances were observed at 0 m （53.89±46.82 ind.） than at 15 m （120±91.7 ind.） and 30 m （283.89±189.33 ind.） away from the dis- posal point. In January 2009, however, higher abundances were observed at 0 m （131.33±58.69 ind.） than at 30 m （100±24.44 ind.） and 15 m （84.78±58.39 ind.） away from the disposal point. Significant differ- ences in macrofaunal assemblage structure were found with varying proximity from the discharge point in both surveys. The effect of proximity from the disposal point was confounded by changes in sedimen- tary composition between surveys, which contributed to explain differences in macrofaunal abundances and assemblage structure with varying proximity from the discharge point. As a result, confounding driver- s of macrofaunal assemblage structure, here, changes in sediment composition, accounted for changes in macrofaunal abundances and assemblage structure with varying proximity from a combined disposal point of brine and sewage.

Wheat streak mosaic virus： incidence in field crops, potential reservoir within grass species and uptake in winter wheat cultivars

Wheat streak mosaic virus（WSMV）has become a re-emerging pathogen in cereal crops in the Czech *** was first reported in the former Czechoslovakia in the early 1980s,and then no record of the virus was documented until *** incidence of the virus was recorded in recent years in several winter wheat fields and many grass ***,we surveyed the incidence of WSMV in cereal *** results demonstrated the existence of the virus in winter wheat and volunteer wheat during each year of the monitoring period,which spanned from 2013–*** the range of infected samples was low（6.4%of the total tested samples）,a high incidence of well-distributed virus was *** at least six fields,the virus reached severe and potentially epidemic *** accordance with our previous report detailing WSMV infection of native grasses,we tested several grass species commonly grown in the Czech *** found that some grass species acted as experimental hosts and possible reservoirs of the virus;these included Anthoxanthum odoratum（sweet vernal grass）,Arrhenatherum elatius（false oat-grass）,Lolium multiflorum（Italian rye-grass）,Bromus japonicus（Japanese chess）,Echinochloa crus-galli（barnyard grass）,Holcus lanatus（meadow soft grass）and Holcus mollis（creeping soft grass）.Some of these grass species are also important weeds of cereals,which may be the potential source of WSMV infection in cereal *** widely used winter wheat cultivars were tested in the field after artificial inoculation with WSMV to evaluate virus titre by ***,the tested cultivars had a low virus titre,which is associated with mild disease symptoms and may provide a good level of crop resistance to WSMV.

Magnetically responsive composite materials have been used in interesting applications in various areas of bioscience, biotechnology, and environmental technology. In this work, a simple method to determine the amount of magnetic iron oxide nano- and microparticles attached to magnetically-modified partic- ulate diamagnetic materials has been developed using a commercially available magnetic permeability meter, The procedure is fast and enables dry particulate magnetically modified materials to be analysed without any modification or pretreatment. We show that the magnetic permeability can be measured for materials containing up to 20% magnetic iron oxide, The magnetic permeability measurements are highly reproducible.
