Negative impact of bone-marrow-derived mesenchymal stem cells on dextran sulfate sodium-induced colitis

AIM:To investigate the effects of mesenchymal stem cells(MSCs)on dextran sulfate sodium-induced inflammatory bowel disease(IBD).METHODS:C57BL/6 mice were fed 3.5%(g/L)dextran sulfate *** day seven,the mice received intraperitoneal injections of 1×106 *** survival rate,disease activity index values,and body weight,were monitored *** day ten,colon lengths and histopathologic changes were *** addition,immunoregulatory changes following MSC administration were evaluated by determining the levels of effector T cell responses in the spleen and mesenteric lymph nodes,and the expression levels of inflammatory cytokines in homogenized ***:Intraperitoneal administration of MSCs did not prevent development of colitis and did not reduce the clinicopathologic severity of *** significant difference was evident in either survival rate or disease activity index score between the control and MSCtreated *** ten-sacrificed mice exhibited no significant difference in either colon length or histopathologic ***,the MSC-treated group exhibited elevated levels of interleukin(IL)-6 and transforming growth factor-β,and a reduced level of IL-10,in spleens,mesenteric lymph nodes,and homogenized *** IL-17 level was lower in the mesenteric lymph nodes of the MSC-treated group(P=0.0126).In homogenized colons,the IL-17 and tumor necrosis factor-α(P=0.0092)expression levels were also lower in the treated ***:MSC infusion provided no significanthistopathologic or clinical improvement,thus representing a limited therapeutic approach for *** enhancement of MSCs is needed in further study.

Single-cell and spatial omics:exploring hypothalamic heterogeneity

Elucidating the complex dynamic cellular organization in the hypothalamus is critical for understanding its role in coordinating fundamental body functions. Over the past decade, single-cell and spatial omics technologies have significantly evolved, overcoming initial technical challenges in capturing and analyzing individual cells. These high-throughput omics technologies now offer a remarkable opportunity to comprehend the complex spatiotemporal patterns of transcriptional diversity and cell-type characteristics across the entire hypothalamus. Current single-cell and single-nucleus RNA sequencing methods comprehensively quantify gene expression by exploring distinct phenotypes across various subregions of the hypothalamus. However, single-cell/single-nucleus RNA sequencing requires isolating the cell/nuclei from the tissue, potentially resulting in the loss of spatial information concerning neuronal networks. Spatial transcriptomics methods, by bypassing the cell dissociation, can elucidate the intricate spatial organization of neural networks through their imaging and sequencing technologies. In this review, we highlight the applicative value of single-cell and spatial transcriptomics in exploring the complex molecular-genetic diversity of hypothalamic cell types, driven by recent high-throughput achievements.

Suboptimal Feature Selection Techniques for Effective Malicious Traffic Detection on Lightweight Devices

With the advancement of wireless network technology,vast amounts of traffic have been generated,and malicious traffic attacks that threaten the network environment are becoming increasingly *** signature-based detection methods,static analysis,and dynamic analysis techniques have been previously explored for malicious traffic detection,they have limitations in identifying diversified malware traffic *** research has been focused on the application of machine learning to detect these ***,applying machine learning to lightweight devices like IoT devices is challenging because of the high computational demands and complexity involved in the learning *** this study,we examined methods for effectively utilizing machine learning-based malicious traffic detection approaches for lightweight *** introduced the suboptimal feature selection model(SFSM),a feature selection technique designed to reduce complexity while maintaining the effectiveness of malicious traffic *** performance was evaluated on various malicious traffic,benign,exploits,and generic,using the UNSW-NB15 dataset and SFSM sub-optimized hyperparameters for feature selection and narrowed the search scope to encompass all *** improved learning performance while minimizing complexity by considering feature selection and exhaustive search as two steps,a problem not considered in conventional *** experimental results showed that the detection accuracy was improved by approximately 20%compared to the random model,and the reduction in accuracy compared to the greedy model,which performs an exhaustive search on all features,was kept within 6%.Additionally,latency and complexity were reduced by approximately 96%and 99.78%,respectively,compared to the greedy *** study demonstrates that malicious traffic can be effectively detected even in lightweight device *** verified the possibility of detecting various attack

Calculation of the intake retention fraction and dose coefficients in ^99mTc-labelled compound for internal exposure for medical workers

For recent decades, a considerable amount of 99mTc has been stimulated for diagnosis radiopharmaceuticals because of its physical advantages. The increase in the use of 99mTc makes naturally more chances of internal exposure for not only patients but also medical workers. The patient internal exposure by an intravenous injection has been assessed relatively well with the reports of International Commission on Radiological Protection (ICRP) or Medical Internal Radiation Dose(MIRD). However, there are few studies which can support its accurate assessment for medical worker who treats 99mTc. In spite of the absence of information, the physiological information of each 99mTc-labelled compound for patient, provided by ICRP, can be used optionally for worker exposure because the behaviors after uptake to blood are similar for patient and worker. Using the data, in this study, the data for bioassay were given as the intake whole-body retention and urinary excretion function. We selected the two most frequently used 99mTc-labelled compounds based on statistical data;99mTc-phosphonate, pertechnetate. The data of the Human Alimentary Tract Model (HATM, publication 100) and the revised Human Respiratory Tract Model (revised HRTM, OIR) were used for compartment models which depict the physiological behavior in body after intake. In case of 99mTc-phosphonate, we adopted the systemic model for patient intake described in ICRP publication 53 based on the assumption that the behaviors after uptake to blood are similar for patient and worker. On the other hand, recent updated systemic model in OIR report could be directly adopted for pertechnetate. We used Birchall's algorithm for calculation and developed the module which could calculate the retention amounts in each compartment at given time by MATLAB. Analysis of expanded criteria to select candidates for active surveillance of low-risk prostate cancer

We aimed to analyze the value of each criterion for clinically insignificant prostate cancer （PCa） in the selection of men for active surveillance （AS） of low-risk PCa. We identified 532 men who were treated with radical prostatectomy from 2006 to 2013 who met 4 or all 5 of the criteria for clinically insignificant PCa （clinical stage 〈 T1, prostate specific antigen [PSA] density 0.15, biopsy Gleason score 〈 6, number of positive biopsy cores 〈 2, and no core with 〉 50% involvement） and analyzed their pathologic and biochemical outcomes. Patients who met all 5 criteria for clinically insignificant PCa were designated as group A （n -- 172）, and those who met 4 of 5 criteria were designated as group B （n = 360）. The association of each criterion with adverse pathologic features was assessed via logistic regression analyses. Comparison of group A and B and also logistic regression analyses showed that PSA density 0.15 ng ml-1 and high （〉7） biopsy Gleason score were associated with adverse pathologic features. Higher （〉 Tlc） clinical stage was not associated with any adverse pathologic features. Although 〈 3 positive cores were not associated with any adverse pathology, 〉4 positive cores were associated with higher risk of extracapsular extension. Among potential candidates for AS, PSA density ~ 0.15 ng ml-~ and biopsy Gleason score ~ 6 pose significantly higher risks of harboring more aggressive disease. The eligibility criteria for AS may be expanded to include men with clinical stage T2 tumor and 3 positive cores.

Tailoring Light–Matter Interactions in Overcoupled Resonator for Biomolecule Recognition and Detection

Plasmonic nanoantennas provide unique opportunities for precise control of light–matter coupling in surface-enhanced infrared absorption(SEIRA)spectroscopy,but most of the resonant systems realized so far suffer from the obstacles of low sensitivity,narrow bandwidth,and asymmetric Fano resonance ***,we demonstrated an overcoupled resonator with a high plasmon-molecule coupling coefficient(μ)(OC-Hμresonator)by precisely controlling the radiation loss channel,the resonator-oscillator coupling channel,and the frequency detuning *** observed a strong dependence of the sensing performance on the coupling state,and demonstrated that OC-Hμresonator has excellent sensing properties of ultra-sensitive(7.25%nm^(−1)),ultra-broadband(3–10μm),and immune asymmetric Fano *** characteristics represent a breakthrough in SEIRA technology and lay the foundation for specific recognition of biomolecules,trace detection,and protein secondary structure analysis using a single array(array size is 100×100μm^(2)).In addition,with the assistance of machine learning,mixture classification,concentration prediction and spectral reconstruction were achieved with the highest accuracy of 100%.Finally,we demonstrated the potential of OC-Hμresonator for SARS-CoV-2 *** findings will promote the wider application of SEIRA technology,while providing new ideas for other enhanced spectroscopy technologies,quantum photonics and studying light–matter interactions.
