Combined Treatment with Bojungikgi-tang(Buzhong Yiqi Decoction)and Riluzole Attenuates Cell Death in TDP-43-Expressing Cells

Objective To examine the effect of combined treatment with Bojungikgi-tang(BJIGT,Buzhong Yiqi Decoction)and riluzole(RZ)in transactive response DNA-binding protein 43(TDP-43)stress granule(SG)cells,a amyotrophic lateral sclerosis(ALS)cell line using transcriptomic and molecular *** TDP-43 SG cells were pretreated with BJIGT(100µg/mL),RZ(50µmol/L),and combined BJIGT(100µg/mL)/RZ(50µmol/L)for 6 h before treatment with lipopolysaccharide(LPS,200µmol/L).Cell viability assay was performed to elucidate cell toxicity in TDP-43 SC cells using a cell-counting kit-8(CCK8)assay *** expression levels of cell death-related proteins,including Bax,caspase 1,cleaved caspase 3 and DJ1 in TDP-43 SG cells were examined by Western blot *** autophagy-related proteins,including pmTOR/mTOR,LC3b,P62,ATG7 and Bcl-2-associated athanogene 3(Bag3)were investigated using immunofluorescence and immunoblotting *** Cell viability assay and Western blot analysis showed that combined treatment with BJIGT and RZ suppressed LPS-induced cell death and expression of cell death-related proteins,including Bax,caspase 1,and DJ1(P<0.05 or P<0.01).Immunofluorescence and immunoblotting assays showed that combined treatment with BJIGT and RZ reduced LPS-induced formation of TDP-43 aggregates and regulated autophagy-related protein levels,including p62,light chain 3b,Bag3,and ATG7,in TDP-43-expressing cells(P<0.05 or P<0.01).Conclusion The combined treatment of BJIGT and RZ might reduce inflammation and regulate autophagy dysfunction in TDP-43-induced ALS.

Effects of ocean acidifi cation driven by elevated CO_2 on larval shell growth and abnormal rates of the venerid clam, Mactra veneriformis

A bstract The venerid clam(M actra veneriformis Reeve 1854) is one of the main cultured bivalve species in intertidal and shallow subtidal ecosystems along the west coast of Korea. To understand the effects of ocean acidifi cation on the early life stages of Korean clams, we investigated shell growth and abnormality rates and types in the D-shaped, umbonate veliger, and pediveliger stages of the venerid clam M. veneriformis during exposure to elevated seawater pCO_2. In particular, we examined abnormal types of larval shell morphology categorized as shell deformations, shell distortions, and shell fi ssures. Specimens were incubated in seawater equilibrated with bubbled CO_2-enriched air at(400±25)×10^(-6)(ambient control),(800±25)×10^(-6)(high pCO_2), or(1 200±28)×10^(-6)(extremely high pCO_2), the atmospheric CO_2 concentrations predicted for the years 2014, 2084, and 2154(70-year intervals; two human generations), respectively, in the Representative Concentration Pathway(RCP) 8.5 scenario. The mean shell lengths of larvae were signifi cantly decreased in the high and extremely high pCO_2 groups compared with the ambient control groups. Furthermore, under high and extremely high pCO_2 conditions, the cultures exhibited signifi cantly increased abundances of abnormal larvae and increased severity of abnormalities compared with the ambient control. In the umbonate veliger stage of the experimental larvae, the most common abnormalities were shell deformations, distortions, and fi ssures; on the other hand, convex hinges and mantle protuberances were absent. These results suggest that elevated CO_2 exerts an additional burden on the health of M. veneriformis larvae by impairing early development.

Inhibitory effect of Jeju endemic seaweeds on the production of pro-inflammatory mediators in mouse macrophage cell line RAW 264.7

Seaweed has been used in traditional cosmetics and as a herbal medicine in treatments for cough,boils,goiters,stomach ailments,and urinary diseases,and for reducing the incidence of tumors,ulcers,and *** the fact that seaweeds are frequently used in the practice of human health,little is known about the role of seaweed in the context of *** study aimed to investigate the influence of Jeju endemic seaweed on a mouse macrophage cell line(RAW 264.7) under the stimulation of lipopolysaccharide(LPS).Ethyl acetate extracts obtained from 14 different kinds of Jeju seaweeds were screened for inhibitory effects on pro-inflammatory *** results revealed that extracts from five seaweeds,Laurencia okamurae,Grateloupia elliptica,Sargassum thun-bergii,Gloiopeltis furcata,and Hizikia fusiformis,were potent inhibitors of the production of pro-inflammatory mediators such as nitric oxide(NO),prostaglandin E2(PGE2),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α).Based on these results,the anti-inflammatory effects and low cell toxicity of these seaweed extracts suggest potential thera-peutic applications in the regulation of the inflammatory response.

Jeju seaweeds suppress lipopolysaccharide-stimulated proinflammatory response in RAW 264.7 murine macrophages

Objective:To investigate the anti-inflammatory effects of Jeju seaweeds on macrophage RAW264.7 cells under lipopolysaccharide(LPS)***:Ethyl acetate fractions were prepared from five different types of Jeju seaweeds,Dictyopteris divaricata(***),Dictyopteris prolifera(***),Prioutis cornea(***,Grateloupia laceolata(G,lanceolate,and Cralcloupia filicina(***)They were screened for inhibitory effects on proinflammatory mediators and cytokines such as nitric oxide(NO),prostaglandin E,,tumor necrosis factor-a(TNF-a),and interleukin-6(11.-6).Results:Our results revealed that ***,***,***,***,and *** potently inhibited ***-stimulaled NO production(IC_(50),values were 18.0,38.36,38.43,32.81 and 37.14μg/mL,respectively).Consistent with these findings,***,***,***,and *** also reduced the IPS-induced and prostaglandin E,production in a concentration-dependent ***,they suppressed the expression of inducible NO synthase and cyclooxygenase-2 at the protein level in a dose-dependent manner in the RAW264.7 cells,as detennined by western *** addition,the levels of TNF-a and IL-6,released into the medium,were also reduced by ***,***,***,G,lanceolata,and *** in a dose-dependent manner(IC_(50)values for TNF-a were 16.11,28.21,84.27,45.52 and74.75μg/mL,respectively;IC_(50),values for IL-6 were 37.35,80.08,103.28,62.53 and 84.28μg/mL,respectively).The total phlorotannin content was measured by the Folin-Ciocalteu method and expressed as phloroglucinol *** content was 92.0μg/mg for ***,151.8μg/mg for ***,57.2μg/mg for ***,53.0 pg/mg for ***,and 40.2μg/mg for ***:Thus,these findings suggest that Jeju seaweed extracts have potential therapeutic applications for inflammatory responses.

In this study,the tribological characteristics of TiN,AlTiN,and AlTiCrN coatings sliding against a SUS420J1 stainless steel pin were investigated in atmospheric and vacuum *** coatings were deposited on SUS440C substrates using the arc-physical vapor deposition *** friction and wear behavior of the coatings were evaluated based on the systematic analyses of the friction coefficient data as well as the physical and chemical state of the wear *** results revealed that the friction coefficients of the SUS440C specimen and AlTiCrN coatings increased,whereas those of the TiN and AlTiN coatings decreased when the environment was changed from atmospheric to *** was confirmed that the formation of an oxide layer and adsorption of oxides on the surface were dominant factors that influenced the tribological behavior in the atmospheric *** the other hand,the compatibility,oxidation inhibition,and droplets of the surface mainly affected the frictional characteristics in the vacuum *** results of this work are expected to aid in the selection of proper coating materials for tribological systems operating in a vacuum.

Protective effects of electroacupuncture on acetylsalicylic acid-induced acute gastritis in rats

AIM: To investigate the protective effects of electroacupuncture (EA) pretreatment on acetylsalicylic acid (ASA)-induced ulceration in rats. METHODS: We randomly divided 72 rats into three groups including control (administered with distilled water), ASA group (administered 100 mg/kg ASA) and EA group (administered EA + 100 mg/kg ASA). Each rat was fasted for 18 to 24 h before experimentation, and lesion scores, gastric acidity, cyclooxygenase (COX)-1 and -2 mRNA levels, and total nitric oxide (NO) concentration were measured. RESULTS: The lesion scores of the EA group were significantly lower than those of the ASA group. Gastric acidity of the ASA and EA groups was reduced compared to the control group. COX-1 and -2 mRNA levels were significantly increased in the EA group as compared to the control and ASA groups, and NO levels were also significantly increased in the EA group as compared to the ASA group. CONCLUSION: These results suggest that EA-mediated protection against ASA-induced ulceration in rats may occur via gastric defense components.

Long-term outcomes of very low birth weight infants with intraventricular hemorrhage:a nationwide population study from 2011 to 2019

Background Advancements in neonatal care have increased preterm infant survival but paradoxically raised intraventricular hemorrhage(IVH)*** study explores IVH prevalence and long-term outcomes of very low birth weight(VLBW)infants in Korea over a *** Using Korean National Health Insurance data(NHIS,2010-2019),we identified 3372 VLBW infants with IVH among 4,129,808 live ***-related claims data,encompassing diagnostic codes,diagnostic test costs,and administered procedures were sourced from the NHIS *** results of the developmental assessments are categorized into four groups based on standard deviation(SD)*** characteristics and complications were compared among the *** regression models were employed to identify significant changes in the incidence of complications and to calculate odds ratios with corresponding 95%confidence intervals for each risk factor associated with mortality and morbidity in ***-term growth and development were compared between the two groups(years 2010-2013 and 2014-2017).Results IVH prevalence was 12%in VLBW and 16%in extremely low birth weight(ELBW)*** the past decade,IVH rates increased significantly in ELBW infants(P=0.0113),while mortality decreased(P=0.0225).Major improvements in certain neurodevelopmental outcomes and reductions in early morbidities have been observed among VLBW infants with *** percent of the population received surgical treatments such as external ventricular drainage(EVD)or a ventriculoperitoneal(VP)shunt,with the choice of treatment methods remaining consistent over *** IVH with surgical intervention group exhibited higher incidences of delayed development,cerebral palsy,seizure disorder,and growth failure(height,weight,and head circumference)up to 72 months of age(P<0.0001).Surgical treatments were also significantly associated with abnormal developmental screening test *** The neurodevelopmental outcomes of infa
