Purpose: Bleomycin is an antibiotic medication that inhibits the synthesis of DNA, RNA, and proteins and is now used in a variety of medical conditions including vascular anomalies. The aim of this study was to evaluate the clinical efficacy of transmucosal intralesional injection of bleomycin in the management of tongue lymphatic malformations. Method: A single institutional case series was presented on patients with recalcitrant lymphatic malformations of the tongue who were treated with bleomycin. Age at the time of injection, gender, number of treatments, amount of bleomycin injected per session, post-injection complications, pre- and post-injection symptoms, and anatomic extent of the lymphatic malformation were all recorded and analyzed. Results: Five patients received transmucosal bleomycin and were followed over a 10-month period. The patients included 4 females and 1 male, aged from 3.25 to 36 years (average 13.52 years). Four patients had one treatment while 1 required two treatments. A total of 1 to 6 units were injected per session. Overall reduction in size of the lymphatic malformation and improvement in all symptoms were observed in the patients by day 14. Average follow-up was 9 to 12 months. Conclusion: Intralesional injection of bleomycin is an effective treatment modality in patients with lymphatic malformations of the tongue.

Background:Patients with von Hippel-Lindau(VHL)disease have a high incidence of renal cell carcinoma owing to VHL gene inactivation and constitutive activation of the transcription factor hypoxia-inducible factor 2α(HIF-2α).Methods:In this phase 2,open-label,single-group trial,we investigated the efficacy and safety of the HIF-2αinhibitor belzutifan(MK-6482,previously called PT2977),administered orally at a dose of 120 mg daily,in patients with renal cell carcinoma associated with VHL *** primary end point was objective response(complete or partial response)as measured according to the Response Evaluation Criteria in Solid Tumors,version 1.1,by an independent central radiology review *** also assessed responses to belzutifan in patients with non-renal cell carcinoma neoplasms and the safety of belzutifan.

To determine cancer pathway activities in nine types of primary tumors and NCI60 cell lines, we applied an in silico approach by examining gene signatures reflective of consequent pathway activation using gene expression data. Supervised learning approaches predicted that the Ras pathway is active in -70% of lung adenocarcinomas but inactive in most squamous cell carcinomas, pulmonary carcinoids, and small cell lung carcinomas. In contrast, the TGF-β, TNF-α, Src, Myc, E2F3, and β-catenin pathways are inactive in lung adenocarcinomas. we predicted an active Ras, Myc, Src, and/or E2F3 pathway in significant percentages of breast cancer, colorectal carcinoma, and gliomas. Our results also suggest that Ras may be the most prevailing oncogenic pathway. Additionally, many NCI60 cell lines exhibited a gene signature indicative of an active Ras, Myc, and/or Src, but not E2F3, β-catenin, TNF-α, or TGF-β pathway. To our knowledge, this is the first comprehensive survey of cancer pathway activities in nine major tumor types and the most widely used NCI60 cell lines. The ＂gene expression pathway signatures＂ we have defined could facilitate the understanding of molecular mechanisms in cancer development and provide guidance to the selection of appropriate cell lines for cancer research and pharmaceutical compound screening.

Pestalotiopsis is a taxonomically confused,pathogenic and chemically creative genus requiring a critical reexamination using a multi-gene phylogeny based on ex-type and ex-epitype *** this study 40 isolates of Pestalotiopsis,comprised of 28 strains collected from living and dead plant material of various host plants from China were studied by means of morphology and analysis of ITS,β–tubulin and tef1 gene sequence *** on molecular and morphological data we describe 14 new species(Pestalotiopsis asiatica,***,***,***,***,***,***,***,***,***,***,***,*** and ***)and three species are epitypified(***,*** and ***).Of the 10 gene regions(ACT,β-tubulin,CAL,GPDH,GS,ITS,LSU,RPB 1,SSU and tef1)utilized to resolve cryptic Pestalotiopsis species,ITS,β–tubulin and tef1 proved to be the better *** other gene regions were less useful due to poor success in PCR amplification and/or in their ability to resolve species *** a single gene tef1 met the requirements for an ideal candidate and functions well for species delimitation due to its better species resolution and PCR ***β-tubulin showed fairly good differences among species,a combination of ITS,β-tubulin and tef1 gene data gave the best resolution as compared to single gene *** work provides a backbone tree for 22 ex-type/epitypified species of Pestalotiopsis and can be used in future studies of the genus.

Gene amplification followed by functional diversification is a major force in evolution. A typical example of this is seen in the wUSCHEL-RELATED HOMEOBOX （wOX） gene family, named after the Arabidopsis stem cell regulator wUSCHEL. Here we analyze functional divergence in the wOX gene family. Members of the wUS clade, except the cambium stem cell regulator wOX4, can substitute for wUS function in shoot and floral stem cell maintenance to different degrees. Stem cell function of wUS requires a canonical wUS-box, essential for interaction with TPL/TPR co-repressors, whereas the repressive EAR domain is dispensable and the acidic domain seems only to be required for female fertility. In contrast to the wUS clade, members of the ancient wOX13 and the wOX9 clades cannot support stem cell maintenance. Although the homeodomains are interchangeable between wUS and wOX9 clade members, a wUS- compatible homeodomain together with canonical wUS-box is not sufficient for stem cell maintenance. Our results suggest that wOX function in shoot and floral meristems of Arabidopsis is restricted to the modern wUS clade, suggesting that stem cell control is a derived function. Yet undiscovered functional domains in addition to the homeodomain and the wUS-box are necessary for this function.
