Asian dust comprises a large portion of the northern hemisphere atmospheric dust load,thereby exerting substantial influence on the Earth’s climate,global biogeochemistry and hydrological cycle through accelerated snow and ice *** deposited on alpine glaciers encodes information on broad scale atmospheric-environmental ***(^(234)u/^(238)u)values of dust fine particulates can reflect the comminuting time and intermediate processes;thus,it provides a new method for the provenance of aeolian dust in the glacial snowpack/*** we present results from a comprehensive survey of uranium isotopic concentrations in dust collected from cryoconites on the glaciers of the northern Tibetan Plateau(TP).These results indicate significant spatial heterogeneity in the(^(234)u/^(238)u)values associated with snowpack/cryoconite dust over a broad range of glaciers in the northern *** values of the(^(234)u/^(238)u)ratio in the glaciers of western Qilian Mountains(Qiyi Glacier,Shiyi Glacier,Laohugou Glacier No.12)were the highest,followed by the Tanggula(Dunkemadi Glacier)and Kunlun Mountains(Yuzhufeng Glacier),whereas these values were the lowest in the eastern Qilian Mountains(Jingyangling Snowpack,Dabanshan Snowpack,Lenglongling Glacier).By including the analysis of Sr-Nd isotopic compositions,we find the spatial isotopic distribution reflects a combination of local dust,which is associated with short comminuting times,and dust transported over long *** data indicate that the dust production in the west and north TP,Alxa arid lands and Gobi Deserts,may have a significant impact on the TP ***,u-Sr-Nd isotopic composition and end-member mixing models(EMMA)were used in our study to find out the relative contribution of distinct Asian dust sources to the dust budget in the TP *** results reveal that snowpack/cryoconite dust is derived from both local sources(low comminution signatures)as well as other dust sources in th

Objective: The DNA repair capacity (DRC) of tumor cells is an important contributor to resistance to radiation and platinum-based drugs. Because DRC may be affected by tumor cell metabolism, we measured DRC in lymphocytes from patients with non-small-cell lung cancer (NSCLC) and compared the findings with the maximum standardized uptake value (SuVmax) on 18 F-fluorodeoxyglucose positron emission tomography (FDG PET) after (chemo)radiation therapy. Methods: This study included 151 patients with stage IA-IV NSCLC who had FDG PET at a single institution and donated blood samples before chemotherapy. We assessed the correlation of DRC, measured in peripheral T lymphocytes by a host-cell reac-tivation assay with SuVmax and their associations with overall survival (OS) time by hazards ratios calculated with a Cox pro-portional hazards regression model. Results: SuVmax of the primary tumor at diagnosis was inversely associated with lymphocyte DRC (r＝－0.175, P＝0.032), particularly among patients with advanced disease (r ＝－0.218, P ＝ 0.015). However, △SuVmax of primary tumor was not significantly associated with DRC (r＝0.005, P＝0.968). SuVmax of regional lymph nodes at diagnosis (r＝－0.307, P＝0.0008) and after (chemo)radiation treatment (r＝－0.329, P＝0.034) and SuVmax of the primary tumor after (chemo)radiation treatment (r＝－0.253, P＝0.045) were also inversely associated with OS time. Conclusion: DRC was inversely associated with primary tumor SuVmax before treatment but not with △SuVmax after (chemo)radiation.

Phytopathogen infections are frequently influenced by both biotic and abiotic factors in a crop field. The effect of brown stink bug, Euschistus servus （Hemiptera： Pentatomidae）, feeding and planting date and sampling time on common smut （ustilago maydis） infection percentage of maize plants was examined in 2005 and 2006, and 2010 and 2011, respectively. Brown stink bug adult feeding on maize hybrid ＂DKC6971＂ at flowering in 2005 and 2006 did not influence smut infection percentage when examined using 3 treatments （i.e., 0 adult, 5 adults, and 5 adults mixed with the smut spores）. The smut infection percentages were 〈 3% （n =12） in the 3 treatments. The smut infection percentage among the 4 weekly samplings was the same, so was natural aflatoxin contamination at harvest among the treatments. The 2nd experiment showed that planting date did not affect the smut infection percentage in either 2010 or 2011. But, the smut infection percentage from the postflowering sampling was greater than preflowering sampling in both years. The smut infection percentage varied among the germplasm lines in 2010, but not in 2011. This study demonstrated that brown stink bug feeding at flowering had no effect on smut infection in maize, and the best time for smut evaluation would be after flowering. The temperature and precipitation might have also influenced the percentage of smut-infected maize plants during the 4 years when the experiments were conducted. The similarity between kernel-colonizing u. maydis and Aspergillus flavus infections and genotype × environment interaction were also discussed.

Modular protein engineering is suited to recruit complex and multiple functionalities in single-chain polypeptides. Although still unexplored in a systematic way, it is anticipated that the positioning of functional domains would impact and refine these activities, including the ability to organize as supramolecular entities and to generate multifunctional protein materials. To explore this concept, we have repositioned functional segments in the modular protein T22-GFP-H6 and characterized the resulting alternative fusions. In T22-GFP-H6, the combination of T22 and H6 promotes selfassembling as regular nanoparticles and selective binding and internalization of this material in CXCR4-overexpressing tumor cells, making them appealing as vehicles for selective drug delivery. The results show that the pleiotropic activities are dramatically affected in module-swapped constructs, proving the need of a carboxy terminal positioning of H6 for protein self-assembling, and the accommodation of T22 at the amino terminus as a requisite for CXCR4^+ cell binding and internalization. Furthermore, the failure of self-assembling as regular oligomers reduces cellular penetrability of the fusions while keeping the specificity of the T22-CXCR4 *** these data instruct how multifunctional nanoscale protein carriers can be designed for smart, protein-driven drug delivery, not only for the treatment of CXCR4^+ human neoplasias, but also for the development of anti-HIV drugs and other pathologies in which CXCR4 is a relevant homing marker.

The development of a new generation of nonantibody protein drug delivery systems requires site-directed conjugation strategies to produce homogeneous,reproducible and scalable *** that,the genetic addition of cysteine residues into solvent-exposed positions allows the thiol-mediated cysteine coupling of therapeutic drugs into protein-based ***,the high reactivity of unpaired cysteine residues usually reduces protein stability,consequently imposing the use of more methodologically demanding purification *** is especially relevant for disulfide-containing nanocarriers,as previously observed in ***,although many protein scaffolds and targeting ligands are also rich in disulfide bridges,the use of these methodologies over emerging non-antibody carrier proteins has been completely ***,we report the development of a simple and straightforward procedure for a onestep production and site-directed cysteine conjugation of disulfide-containing non-antibody thiolated carrier proteins(THIOCAPs).This method is validated in a fluorescent C-X-C chemokine receptor 4(CXCR4)-targeted multivalent nanocarrier containing two intramolecular disulfide bridges and one reactive cysteine residue strategically placed into a solvent-exposed position(THIO-T22-GFP-H6)for drug conjugation and in a humanized alternative intended for clinical applications(T22-HSNBT-H6).Thus,we produce very stable,homogeneous and fully functional antitumoral nanoconjugates(THIO-T22-GFP-H6-MMAE and T22-HSNBT-H6-MMAE)that selectively eliminate target cancer cells via ***,the developed methodology appears as a powerful tool for the rational engineering of emerging non-antibody,cell-targeted protein nanocarriers that contain disulfide bridges together with a solvent-exposed reactive cysteine(THIOCAP).This should pave the way for the development of a new generation of stable,homogeneous and efficient nanomedicines.

A soil coring protocol was developed to cooptimize the estimation of root length distribution(RLD)by depth and detection of functionally important variation in root system architecture(RSA)of maize and *** functional-structural model OpenSimRoot was used to perform in silico soil coring at six locations on three different maize and bean RSA *** were compared to two seasons of field soil coring and one *** one-sided T-test(TOST)analysis of in silico data suggests a between-row location 5 cm from plant base(location 3),best estimates whole-plot RLD/D of deep,intermediate,and shallow RSA phenotypes,for both maize and *** discriminant analysis indicates location 3 has~70%categorization accuracy for bean,while an in-row location next to the plant base(location 6)has~85%categorization accuracy in *** of field data suggests the more representative sampling locations vary by year and *** silico and field studies suggest location 3 is most robust,although variation is significant among seasons,among replications within a field season,and among field soil coring,trench,and *** propose that the characterization of the RLD profile as a dynamic rhizo canopy effectively describes how the RLD profile arises from interactions among an individual plant,its neighbors,and the pedosphere.

Background In children, surveys on Staphylococcus aureus have focused on specific infections, situations or strains but no study has so far given an overview on S. aureus isolation without any selection. Here, we describe the overall bacteriologi-cal and clinical characteristics of S. aureus isolation in children, with a special focus on isolates harbouringtst,sea, and/or luk-PV genes, respectively, encoding the three clinically relevant toxins: toxic shock syndrome toxin-1, enterotoxin A and Panton–Valentine leukocidin. Methods Data associated with S. aureus isolation were reviewed: isolation site, infection status,tst,sea andluk-PV genes, antimicrobial susceptibility pattern,agr typing. Results Three hundred and seventy-seven isolates retrieved from 328 children during S. aureus infection (55.2%) or colo-nisation (44.8%) were ***,sea and luk-PV genes were amplified in 14.3, 9.5 and 5.8% of the isolates, respectively. These isolates were significantly more frequently retrieved during infection (69.1%) than colonisation but differences were observed according to isolation site. Methicillin-resistance was found in 7.2% of the isolates, 78% of which harboured ≥ 1 of the targeted toxin-encoding genes. Conclusions This first comprehensive study of S. aureus in children showed S. aureus to be mainly retrieved during infection and a high rate of colonisation, not limited to the nasopharynx. Predominant infections were skin and soft tissue infections wheretst was most frequently ***-PV was most commonly detected during bone and joint infections. Isolates harbouring targeted toxin-encoding genes were significantly associated with infections but a quarter of children were asymp-tomatic carriers representing a reservoir for dissemination of isolates with virulence potency.
