How Polydopamine Modulates Biological Responses to PTFE Prostheses

Diaphragm repair after congenital diaphragmatic hernia is associated with hernia recurrence due to prosthesis failure. Expanded polytetrafluoroethylene (e-PTFE), a synthetic non-degradable biomaterial, is currently used for those diaphragmatic defect repairs. The drawback of e-PTFE is its poor wettability that leads to coating difficulties, bonding that could favor implant integration. However, polydopamine (PDA) can be deposited as well on organic as on inorganic substrates. Therefore, we assessed the biological responses of a clinically used e-PTFE biomaterial treated with PDA in two different manners: one impregnated with PDA and the other coated with a one side PDA film. Mechanical properties of the raw e-PTFE, the PDA soaked biomaterial and the PDA coated surface were characterized by colloidal probe atomic force microscopy. Behaviors of primary human fibroblasts and Wharton’s jelly stem cells were investigated by electron microscopy. Findings reveal that the mechanical properties at the microscopic scale are not modified by the PDA treatments. Cells spread onto both PDA functionalized substrates. In addition, microscopic observations disclose numerous focal cell contacts, evidencing cell attachment, and cytoplasmic projections particularly with the nanoscale PDA coating. Results clearly suggest that PDA in general but above all the PDA coating enhance cellular colonization of the implant material.

In this paper we perform a numerical study of the spectra,eigenstates,and Lyapunov exponents of the skew-shift counterpart to Harper’s *** study is motivated by various conjectures on the spectral theory of these’pseudo-random’models,which are reviewed in detail in the initial sections of the *** numerics carried out at different scales are within agreement with the conjectures and show a striking difference compared with the spectral features of the Almost mathieu *** particular our numerics establish a small upper bound on the gaps in the spectrum(conjectured to be absent).

Potential Compressive Sites of the Anterior Interosseous Nerve in the Proximal Forearm: An Anatomic Study

Background: The etiology and treatment of spontaneous paralysis variants of anterior interosseous nerve (AIN) syndrome remains controversial. Variation and multiple sites of potential compression complicate the successful performance of neurolysis. This anatomic study of the AIN and sites of potential compression in the proximal forearm facilitates critical steps involved in neurolytic procedures and management. Methods: Upper extremities of twelve cadavers were examined to evaluate potential sites of AIN compression in the proximal forearm. Potential sites of musculoaponeurotic compression were evaluated, including: lacertus fibrosus;inferior fibrous arch of the humeral head of the pronator teres (PT) muscle;inferior fibrous arch of the ulnar head of the PT muscle;fibrous arch in the flexor digitorum superficialis (FDS) muscle;Gantzer’s muscle;and vascular structures near the AIN and median nerve. Results: The AIN arose at a mean distance of 54.5 mm distal to the elbow from the posterior (n = 9) or ulnar side (n = 3) of the median nerve. Relative positions of AIN branches were variable. A fibrous arch was found between the lacertus fibrosus and the PT in two cases. Nine cadavers had two fibrous arches in the PT and FDS, and three cadavers had one arch. An accessory head in the FDS was found to be a risk of AIN compression. Gantzer’s muscle was present in six cases, crossing the AIN superficially. Two potentially compressive vascular arches were identified. Conclusions: Our observations confirm that multiple musculoaponeurotic and/or vascular structures can contribute to AIN compression in the proximal forearm. Understanding the complex anatomic relationships of this nerve is crucial to improving outcomes of neurolysis in cases of non-regressive AIN paralysis.

Topological Modelling of Deep Ulcerations in Patients with Ulcerative Colitis

Aims: Steadily the clinicians of our team in inflammatory bowel disease encounter ulcerative colitis patients that develop deep ulcers during their treatment. Currently, these practitioners are only equipped with their grade of expertise in inflammatory domains to decide what new therapy maybe use in such cases. Encouraged by the limited knowledge of this frequent pathology, we seek to determine the molecular conditions underlying the recurrent formation of deep ulcerations in certain group of patients. Method: The goal of this strategy is to expose differences between groups of patients based on similarities computed by random walk graph kernels and performing functional inference on those differences. Results: We apply the methodology to a cohort of eleven miRNA microarrays of ulcerative colitis patients. Our results showed how the group of ulcerative colitis patients with presence of deep ulcers is topologically more similar (0.35) than ulcerative colitis patients (0.18) to control. Such topological constraint drove functional inference to complete the information that clinicians need. Conclusions: Our analyses reveal highly interpretable in the guidance of practitioners to eventually correct initial therapies of ulcerative colitis patients that develop deep ulcers. The methodology can provide them with useful molecular hypotheses necessaries prior to make any decision on the newest course of the treatment.

Gregarious pupation act as a defensive mechanism against cannibalism and intraguild predation

Coccinellid pupae use an array of defensive strategies against their natural enemies. This study aims to assess the efficiency of gregarious pupation as a defensive mechanism against intraguild predators and cannibals in coccinellid. The study was designed specifically （i） to determine the natural occurrence of gregarious pupation in the field for different coccinellid species, and （ii） to evaluate the adaptive value of gregarious pupation as a defensive mechanism against 2 types of predators （i.e., cannibals and intraguild predators）. In the field, gregarious pupation consisted of a group of 2-5 pupae. The proportion of gregarious pupation observed varied according to species, the highest rate being observed with Harmonia axyridis Pallas （Coccinellidae; 14.17%）. Gregarious pupation had no impact on the probability that intraguild predators and cannibals locate pupae. Intraguild predation occurred more often in site with gregarious pupation, while cannibalism occurred as often in site with gregarious pupation as in site with isolated pupa. However, for a specific pupa, the mortality rate was higher for isolated pupae than for pupae located in a gregarious pupation site both in the presence of intraguild predators and in the presence of cannibals. The spatial location of pupae within the group had no impact on mortality rate. Since it reduces the risk of predation, it is proposed that gregarious pupation act as a defensive mechanism for 11. axyridis pupae.

The advent of sentinel lymph-node technique has led to a shift in lymph-node staging,due to the emergence of new entities namely micrometastases(p N1mi) and isolated tumor cells [p N0(i+)].The prognostic significance of this low positivity in axillary lymph nodes is currently debated,as is,therefore its *** article provides updates evidence-based medicine data to take into account for treatment decision-making in this setting,discussing the locoregional treatment in p N0(i+) and p N1 mi patients(completion axillary dissection,axillary irradiation with or without regional nodes irradiation,or observation),according to systemic treatment,with the goal to help physicians in their daily practice.

Eating Habits among Lithiasic Patients in Kinshasa, the Democratic Republic of Congo

Introduction: Worldwide, the incidence of kidney stones has been progressively increasing. Various factors can influence the risk of stone formation, including lifestyle and eating habits. The dietary investigation is a standard of care in patients with urolithiasis. The objectives of this study were to determine the dietary habits of lithiasis patients in the city of Kinshasa and to investigate the association between dietary habits and the composition of the stones. Material and Methods: From January 2017 to September 2019, 85 patients attending 8 hospitals participated in the nutrition survey. Various foods commonly consumed in the Democratic Republic of Congo were categorized based on their composition: foods rich in animal proteins, foods rich in calcium, foods high in sugar, foods high in oxalate, and various vegetables and fruits. We also investigated daily water intake. The composition of the collected stones was analyzed by infrared spectrophotometry. Results: The mean (SD) age of patients was 47.1 (14.0) years, 63.5% of patients were males, 75.3% of the patients had stones located in the upper urinary tract, and 7.1% were undernourished. Most of patients consumed vegetables (77.6%), animal proteins (62.4%), and foods rich in oxalate (58.8%). The daily water intake was less than 1500 mL in more than half of the patients (68.2%). Major anhydrous uric acid stones were associated with a high body mass index (p = 0.025). Male patients with a high oxalate diet had more calcium oxalate stones (64.3%) compared to other types of stones (p = 0.041). Conclusion: High consumptions of vegetables and low water intake were linked to the formation of calcium oxalate monohydrate stones. More data are needed to confirm these findings.

目的在16p13-12．1位点20 cm的区域内明确致病基因,了解基因型和临床表型的关系。方法采用双向DNA直接测序法筛选候选基因;运用免疫组化和蛋白免疫印迹法等了解突变基因的表达情况。运用质粒重组及体外表达方法获得野生型和突变型蛋白,使用免疫共沉淀法了解这两者的结合程度。结果在两个家系中发现编码平滑肌肌球蛋白的MYH11基因突变,所有TAAD和PDA患者均为突变基因携带者。在法国家系中发现两个突变。前者为剪切点供体杂合子突变,导致编码71个氨基酸的第32外显子完全缺失:第2个突变为第37外显子上的1个点突变,导致精氨酸变为谷氨酸。在美国家系中发现1个杂合子突变,为第28外显子上编码24个氨基酸的72个碱基缺失。在法国患者的病变动脉标本上运用蛋白免疫印迹法,发现野生型和突变型肌球蛋白蛋白共存,但有异常免疫组化表现。体外实验显示重组野生型和突变型蛋白不能正常结合。结论MYH11基因突变导致家族性TAAD合并PDA,野生型和突变型平滑肌肌球蛋白不能正常结合,推论杂合子突变导致“Dominant-negative”作用。
