Tissue factor in predicted severe acute pancreatitis

AIM: To study tissue factor (TF) in acute pancreatitis and evaluate the role of TF as a predictive marker of severity. METhODS: Forty-nine consecutive patients admitted to Lund University hospital, fulfilling the criteria of predicted severe acute pancreatitis (AP), were recruited prospectively between 2002 and 2004. Blood samples for TF analyses were drawn at inclusion in the study and 12 h, 1 d and 3 d later. RESULTS: Twenty-seven patients developed mild AP, and 22 patients severe AP. At inclusion in the study, the groups were comparable with respect to gender, aetiology, Acute Physiology and Chronic health Evaluation Ⅱ score, and duration of pain. At inclusion in the study and at 12 h, TF was higher in the severe AP group (P = 0.035 and P = 0.049, respectively). After 1 and 3 d, no differences in TF levels were noted. Interleukin (IL)-6 was significantly higher in the severe AP group at all of the studied time points. C-reactive protein (CRP) was significantly higher in the AP group at 1 and 3 d. In receiver operating characteristic-curves, the area under the curve (AUC) for TF was 0.679 (P = 0.035) at inclusion in the study, and a cut off level for TF of 40 pg/mL showed a sensitivity of 71% and a specificity of 67%, whereas corresponding AUC for IL-6 was 0.775, P = 0.001, and for CRP was 0.653. IL-6 showed better AUC-values than TF at all time points studied. CONCLUSION: TF-levels are raised early in severe AP. TF as an early predictive marker of severe AP is superior to CRP, but inferior to IL-6.

Guar [Cyamopsis tetragonoloba （L.） Taub.] is one of the most important industrial crops due to its richness in gum. Understanding the inheritance of gum content is a key to its successful genetic improvement. Gum content expression is reported to be controlled by additive, dominant, and epistatic effects, and modified by the environment, fg%, a relative gum content calculated by comparing with the gum content of Kinman was used as gum content （%） in this experiment. Reciprocal crosses of two lines of guar, PI 217923 and Lewis, were made to study the heritability of gum content （fg%）. fg% of four plant introductions and four commercial varieties were studied in Lubbock in 1999-2002. Estimates of broadsense heritability （h^2 b,s.） of fg% in Lewis x PI 217923 and PI 217923 x Lewis were 75.53 and 52.74%, respectively. Estimates of narrow-sense heritability （h2 n.s.） of fg% were 40.00 and 29.00% thr Lewis x PI 217923 and PI 217923 x Lewis, respectively. At least one pair of genes were estimated to control the fg% expression in these two crosses, Significant differences of fg% were found among these eight entries. PI 217923 was found to have the highest fg% among the eight entries.

Tomato production is influenced by shoot branching,which is controlled by different *** we produced tomato plants overexpressing the cytokinin-deactivating gene CYTOKININ OXYDASE 2(CKX2).CKX2-overexpressing(CKX2-OE)plants showed an excessive growth of axillary shoots,the opposite phenotype expected for plants with reduced cytokinin content,as evidenced by LC-MS analysis and ARR5-GUS *** TCP transcription factor SlBRC1b was downregulated in the axillary buds of CKX2-OE and its excessive branching was dependent on a functional version of the GRAS-family gene LATERAL SUPPRESSOR(LS).Grafting experiments indicated that increased branching in CKX2-OE plants is unlikely to be mediated by root-derived *** CKX2-OE plants with transgenic antisense plants for the strigolactone biosynthesis gene CAROTENOID CLEAVAGE DIOXYGENASE(CCD7-AS)produced an additive phenotype,indicating independent effects of cytokinin and strigolactones on increased *** the other hand,CKX2-OE plants showed reduced polar auxin transport and their bud outgrowth was reduced when combined with auxin ***,CKX2-OE basal buds did not respond to auxin applied in the decapitated *** results suggest that tomato shoot branching depends on a fine-tuning of different hormonal balances and that perturbations in the auxin status could compensate for the reduced cytokinin levels in CKX2-OE plants.

Leukocyte activation has been linked to atherogenesis, but there is little in vivo evidence for its role in the progression of atherosclerosis. We evaluated the predictive value for progression of coronary artery disease （CAD） of leukocyte activation markers in the coronary circulation. Monocyte and neutrophil CDllb, neutrophil CD66b expression and intracellular neutrophil myeloperoxidase （MPO） in the coronary arteries were determined by flow cytometry in patients undergoing coronary angiography. The primary outcome included fatal and nonfatal myocardial infarction or arterial vascular intervention due to unstable angina pectoris. In total 99 subjects who were included, 70 had CAD at inclusion （26 patients had single-vessel disease, 18 patients had two- vessel disease and 26 patients had three-vessel disease）. The median follow-up duration was 2242 days （interquartile range： 2142-2358）. During follow-up, 13 patients （13%） developed progression of CAD. Monoeyte CDllb, neutrophil CDllb and CD66b expression and intraceilular MPO measured in blood obtained from the coronary arteries were not associated with the progression of CAD. These data indicate that coronary monocyte CDllb, neutrophil CDllb and CD66b expression and intracellular MPO do not predict the risk of progression of CAD.

To obtain a global overview of how mitochondria respond to stress, we aimed to define the plant mitochondrial stress response （MSR）. By combining a set of 1196 Arabidopsis thaliana genes that putatively encode mitochondrial proteins with 16 microarray experiments on stress-related conditions, 45 nuclear encoded genes were defined as widely stress-responsive. Using green fluorescent protein （GFP） fusion assays, the mitochondrial targeting of a large number of these proteins was tested, confirming in total 26 proteins as mitochondrially targeted. Several of these proteins were observed to be dual targeted to mitochondria and plastids, including the small heat shock proteins shSP23.5 and shSP23.6. In addition to the well defined stress components of mitochondria, such as alternative oxidases, nicotinamide adenine dinudeotide （NAD（P）h） dehydrogenases, and heat shock proteins, a variety of other proteins, many with unknown function, were identified. The mitochondrial carrier protein family was over-represented in the stress-responsive genes, suggesting that stress induces altered needs for metabolite transport across the mitochondrial inner membrane. Although the genes encoding many of these proteins contain common cis-acting regulatory elements, it was apparent that a number of distinct regulatory processes or signals likely triggered the MSR. Therefore, these genes provide new model systems to study mitochondrial retrograde regulation, in addition to the widely used alternative oxidase model. Additionally, as changes in proteins responsive to stress did not correlate well with changes at a transcript level, it suggests that post-transcriptional mechanisms also play an important role in defining the MSR.
