Resistance to cancer therapy continues to be a major limitation for the successful treatment of cancer. There are many published studies on therapy resistance in breast and prostate cancers; however, there are currently no data on molecular markers associated with resistance. The conflicting data were reported regarding the AKT/m-TOR signaling pathway components as markers predicting resistance. The AKT/m-TOR signaling pathway is involved in the development of many human cancers; its activation is related to cell proliferation, angiogenesis, apoptosis, as well as to therapy resistance. molecular alterations in the AKT/m-TOR signaling pathway provide a platform to identify universal markers associated with the development of resistance to cancer therapy.

Purpose: The aim of the study is to reveal associations between NF-κB, HIF-1alpha, VEGF expres-sions, proteasome and calpain activities with tumor progression in patients with kidney cancers and to find molecular parameters, associated with the effective pazopanib therapy. 93 patients with clear cell kidney cancers are included in investigation. 26 patients with disseminated kidney cancer have the pazopanib therapy. methods: Transcription factors, VEGF, VEGFR2 and p-m-TOR expression are measured by ELISA kits. Proteasome and calpain activity are determined using specific fluorogenic substrate. Results: It is found the increase of NF-κB, HIF-1 expression in cancer tissues followed the hematogenic metastasis development. Coefficient NF-κB р65/р50 and VEGF expression are increased in cancer tissues with single metastasis and are decreased in cancer tissues with multiple ones. It is observed in the low proteasome activity in metastatic cancer tissues. The partial cancer regression is revealed in 29.6% of patients treated with pazopanib, cancer stabilization—in 61.5% of patients and cancer progression—in 11.5% of patients. The increased level of transcription factors NF-κB, HIF-1, growth factor VEGF and high proteasome activity in cancer tissues before targeted therapy are associated with the effective treatment. It is obtained the significant decrease of investigated markers after pazopanib application in metastatic kidney cancer patients. Conclusion: Coefficient NF-κB р65/р50, VEGF expression and proteases activities are the potential prognostic molecular markers of hematogenic metastasis development in kidney cancers. NF-κB, HIF-1 and VEGF levels can be considered as additional molecular markers predicting the effective pazopanib therapy.

Astrocytes,the main population of glial cells in the central nervous system(CNS),exert essential tasks for the control of brain tissue homeostasis,supporting neuron and other glial cell activity from the developmental stage to adult *** maintain the optimal functionality of the brain,astroglial cells are particularly committed to reacting to every change in tissue homeostatic conditions,from mild modifications of the physiological environment,a process called astrocyte activation,to the more severe alterations occurring in pathological situations causing astrocyte reactivity or reactive astrogliosis(Escartin et al.,2021).During these reactive states,astrocytes mount an active,progressive response encompassing morphological,molecular,and interactional remodeling,leading to the acquisition of new functions and the loss of others,whose intensity,duration,and reversibility are dependent on the nature of the stimulus and regulated in a context-specific manner.

The formation of C–C bonds is a crucial aspect of organic synthesis. Synthesists' long-term objective is to identify novel catalysts to optimize reaction systems based on traditional transition metal catalysis. This study explores the synthesis and catalytic potential of palladium-installed covalent metal-organic frameworks(CmOFs) based on copper cyclic trinuclear units(Cu-CTUs). By incorporating palladium ions into the imine-based framework JNm-7 through a straightforward immersion method, we achieved a notable enhancement in catalytic activity for critical C–C coupling reactions, including C–H arylation and Sonogashira cross-coupling. The resulting JNm-7-Pd demonstrated remarkable stability and recyclability, maintaining its efficiency over at least three cycles without any decline in efficiency.

The initial-boundary value problem for a general balance law in a bounded domain is proved to be well posed. Indeed, we show the existence of an entropy solution, its uniqueness and its Lipschitz continuity as a function of time, of the initial datum and of the boundary datum. The proof follows the general lines in [4], striving to provide a rigorous treatment and detailed references.

Hereditary non-polyposis colorectal cancer(HNPCC) was previously synonymous with Lynch syndrome; however,identification of the role of germline mutations in the DNA mismatch repair(mmR) genes has made it possible to differentiate Lynch syndrome from other conditions associated with familial colorectal cancer(CRC). Broadly,HNPCC may be dichotomized into conditions that demonstrate defective DNA mmR and microsatellite instability(mSI) vs those conditions that demonstrate intact DNA mmR. Conditions characterized by mmR deficient CRCs include Lynch syndrome(germline mmR mutation),Lynch-like syndrome(biallelic somatic mmR mutations),constitutional mmR deficiency syndrome(biallelic germline mmR mutations),and sporadic mSI CRC(somatic biallelic methylation of mLH1). HNPCC conditions with intact DNA mmR associated with familial CRC include polymerase proofreading associated polyposis and familial colorectal cancer type X. Although next generation sequencing technologies have elucidated the genetic cause for some HNPCC conditions,others remain genetically undefined. Differentiating between Lynch syndrome and the other HNPCC disorders has profound implications for cancer risk assessment and surveillance of affected patients and their at-risk relatives. Clinical suspicion coupled with molecular tumor analysis and testing for germline mutations can help differentiate the clinical mimicry within HNPCC and facilitate diagnosis and management.

Effect of storage conditions on long-term stability of Ag nanoparticles formed via green synthesis

Spherical Ag nanoparticles(AgNPs) with a diameter of 20 nm or smaller were biologically synthesized using algae Parachlorella kessleri. The effect of storage conditions on the long-term stability of AgNPs was investigated. UV/Vis spectrophotometry, transmission electron microscopy, and dynamic light scattering measurements revealed that the long-term stability of AgNPs was influenced by light and temperature conditions. The most significant loss of stability was observed for the AgNPs stored in daylight at room temperature. The AgNPs stored under these conditions began to lose their stability after approximately 30 d; after 100 d, a substantial amount of agglomerated particles settled to the bottom of the Erlenmeyer flask. The AgNPs stored in the dark at room temperature exhibited better long-term stability. Weak particle agglomeration began at approximately the 100 th day. The AgNPs stored in the dark at about 5℃ exhibited the best long-term stability; the AgNPs stored under such conditions remained spherical, with a narrow size distribution, and stable(no agglomeration) even after 6 months. Zeta-potential measurements confirmed better dispersity and stability of AgNPs stored under these conditions.

Grain production in the countries of the former USSR sharply declined during the past two decades and has only recently started to recover. In the context of the current economic and food-price crisis, Russia, Ukraine, and Kazakhstan might be presented with a window of opportunity to reemerge on the global agricultural market, if they succeed in increasing their productivity. The future of their agriculture, however, is highly sensitive to a combination of internal and external factors, such as institutional changes, land-use changes, climate variability and change, and global economic trends. The future of this region＇s grain production is likely to have a significant impact on the global and regional food security over the next decades.

BACKGROUND Early identification of patients at risk of congestive heart failure(HF)may alter their poor *** aim was therefore to test whether simple electrocardiographic variables,the P-wave and PR-interval,could predict incident *** The PIVUS(Prospective Investigation of the Vasculature in Uppsala Seniors)study(1016 individuals all aged 70 years,50%women)was used to identify predictors of *** with prevalent HF,QRS duration≥130 ms,atrial tachyar-rhythmias,implanted pacemaker/defibrillator,second-and third-degree atrioventricular block or delta waves at baseline were *** proportional hazard analysis was used to relate the PR interval,P-wave duration(Pdur)and amplitude(Pamp),measured in lead V1,to incident *** was performed for gender,RR-interval,beta-blocking agents,systolic blood pressure,body mass index and *** Out of 836 subjects at risk,107 subjects were diagnosed with HF during a follow-up of 15 *** the multivariate analysis,there was a strong U-shaped correlation between Pdur in lead V1 and incident HF(P=0.0001)which was significant for a Pdurms[HR=2.75;95%CI:1.87-4.06,at Pdur 40 ms]but not for prolonged *** was no significant relationship between incident HF and the PR-interval or the Pamp.A Pdurms improved discrimination by 3.7%when added to the tradi-tional risk factors including sex,RR-interval,beta-blocking agents,systolic blood pressure,BmI and smoking(P=0.048).CONCLUSIONS A short Pdur,an easily measured parameter on the ECG,may potentially be a useful marker of future HF,enabling its early detection and prevention,thus improving outcomes.
