Molecular Phylogeny of the Genus Gloydius(Serpentes: Crotalinae)

Based on two mitochondrial genes (cyt b, ND4) and one nuclear gene (c-mos), we explored the relationships within the Asian pit viper genus Gloydius. In total, 23 samples representing 10 species were analyzed. All phylogenetic analyses support a monophyletic Gloydius with two major clades, one comprising G. brevicaudus, G. blomhoffii, and G. ussuriensis with the sister clade consisting of G. intermedius, G. saxatilis, G. halys and G. shedaoensis. The relationships among the three montane species G. strauchi, G. qinlingensis and G. liupanensis, as well as the two monophyletic groups, are unstable, and discussed. Divergence date estimation indicates that Gloydius lineage formed 15 Ma and diversification of the genus occurred at 9.89 Ma. Issues regarding the taxonomy of this genus are discussed where necessary.

Spatial Interpolation of Tidal Data Using a Multiple-Order Harmonic Equation for Unstructured Grids

A general spatial interpolation method for tidal properties has been developed by solving a partial differential equation with a combination of different orders of harmonic operators using a mixed finite element method. Numerically, the equation is solved implicitly without iteration on an unstructured triangular mesh grid. The paper demonstrates the performance of the method for tidal property fields with different characteristics, boundary complexity, number of input data points, and data point distribution. The method has been successfully applied under several different tidal environments, including an idealized distribution in a square basin, coamplitude and cophase lines in the Taylor semi-infiite rotating channel, and tide coamplitude and cophase lines in the Bohai Sea and Chesapeake Bay. Compared to Laplace’s equation that NOAA/NOS currently uses for interpolation in hydrographic and oceanographic applications, the multiple-order harmonic equation method eliminates the problem of singularities at data points, and produces interpolation results with better accuracy and precision.

【目的】探讨切割修复交叉互补基因 2 (excisionrepaircross complementingrodentrepairdeficiencygene 2 ,ERCC2 )表达与人肿瘤对抗癌药耐药的关系。【方法】采用Westernblot检测美国国家癌症研究所 (NationalCancerInstitute,NCI)用于抗癌药筛选的 6 0株人肿瘤细胞的ERCC2表达 ,并与 170种抗癌药物的细胞毒试验结果进行相关性分析。【结果】肿瘤细胞ERCC2的蛋白表达水平高低不一 ,其中 6株细胞的ERCC2蛋白水平在可测定范围以下。 170种抗癌药物中有 2 8种化疗药物的耐药性与肿瘤细胞的ERCC2蛋白水平相关性显著或非常显著。根据药物作用机理分析 ,肿瘤细胞ERCC2表达与其对烷化剂和抗有丝分裂剂类抗癌药耐药相关性显著。【结论】肿瘤细胞具有DNA修复能力 ,如核苷酸切割修复。

Perianal Crohn’s disease remains a challenging condition to treat and can have a substantial negative impact on quality of *** often requires combined surgical and medical ***-tumor necrosis factor(anti-TNF)therapy,including infliximab and adalimumab,remain preferred medical therapies for perianal Crohn’s *** has been shown to be efficacious in improving fistula closure rates in randomized controlled *** can be faced with a number of questions relating to the optimal use of anti-TNF therapy in perianal Crohn’s *** issues include evaluation for the presence of perianal sepsis,the treatment target of therapy,the ideal time to commence treatment,whether additional medical therapy should be used in conjunction with anti-TNF therapy,and the duration of *** article will discuss key studies which can assist clinicians in addressing these matters when they are considering or have already commenced anti-TNF therapy for the treatment of perianal Crohn’s *** will also discuss current evidence regarding the use of vedolizumab and ustekinumab in patients who are failing to achieve a response to anti-TNF therapy for perianal Crohn’s ***,new therapies such as local injection of mesenchymal stem cell therapy will be discussed.

Background:This study examined real-world treatment and management of bacillus Calmette-Guérin(BCG)-unresponsive patients across 3 continents,including patients unable or unwilling to undergo *** and methods:Physicians actively involved in managing patients with nonmuscle invasive bladder cancer completed online case report forms for their 5 consecutive patients from the broad BCG-unresponsive population and a further 5 consecutive BCG-unresponsive patients who did not undergo cystectomy(in Japan,physicians provided a total of 5 patients across both cohorts).Results:Most patients had received 1(37%)or 2(24%)maintenance courses of *** or more maintenance BCG courses were received by patients in Japan(59%)and China(31%),while in Germany 76%of patients received only 1 *** patients became BCG-unresponsive during their first(44%)or second(22%)treatment course;in Germany,77%became BCG-unresponsive during their first treatment *** countries did not provide another course of BCG after a patient first became unresponsive,whereas unresponsive patients in Japan and China were most likely to be retreated with BCG."Untreated-on watch and wait"was the main treatment/management approach received post-BCG treatment for 42%or more of patients in most countries except China(39%)and the United States(36%)."Following treatment guidelines"was consistently the top reason for post-BCG treatment selection across all treatment ***:This study confirmed the global unmet need for patients with nonmuscle invasive bladder cancer,and found that many patients experienced periods of no treatment after not responding to BCG therapy.

目的:探讨核苷酸切除修复（NER,nucleotide excision repair）系统的重要因子切除修复交叉互补基因2（excision repair cross-com- plementing rodent repair deficiency gene 2,ERCC2）表达与人肿瘤对抗癌药耐药的关系。方法:采用 western blot 检测美国国家癌症研究所（National Cancer Institute,NCI）用于抗癌药筛选的60株人肿瘤细胞的 ERCC2表达,并与170种抗癌药物的细胞毒试验结果进行相关性分析。结果:肿瘤细胞 ERCC2的蛋白表达水平高低不一,其中6株细胞的 ERCC2蛋白水平在可测定范围以下。ERCC2蛋白水平与肿瘤细胞对化疗药物耐药的相关性在170种抗癌药物中有28种非常显著（p ＜0.05）。根据药物作用机理分析,肿瘤细胞 ERCC2表达与其对烷化剂和抗有丝分裂剂类抗癌药耐药相关性显著。结论:肿瘤细胞具有 DNA 修复能力,如核苷酸切除修复,更具体是具有 ERCC2基因表达将表现为对主要是烷化剂和抗有丝分裂剂类抗癌药的耐药。

Osteomyelitis is a devastating disease caused by microbial infection of bone. While the frequency of infection following elective orthopedic surgery is low, rates of reinfection are disturbingly high. Staphylococcus aureus is responsible for the majority of chronic osteomyelitis cases and is often considered to be incurable due to bacterial persistence deep within bone. Unfortunately, there is no consensus on clinical classifications of osteomyelitis and the ensuing treatment algorithm. Given the high patient morbidity,mortality, and economic burden caused by osteomyelitis, it is important to elucidate mechanisms of bone infection to inform novel strategies for prevention and curative treatment. Recent discoveries in this field have identified three distinct reservoirs of bacterial biofilm including: Staphylococcal abscess communities in the local soft tissue and bone marrow, glycocalyx formation on implant hardware and necrotic tissue, and colonization of the osteocyte-lacuno canalicular network(OLCN) of cortical bone. In contrast, *** intracellular persistence in bone cells has not been substantiated in vivo, which challenges this mode of chronic osteomyelitis. There have also been major advances in our understanding of the immune proteome against S. aureus, from clinical studies of serum antibodies and media enriched for newly synthesized antibodies(MENSA), which may provide new opportunities for osteomyelitis diagnosis, prognosis, and vaccine development. Finally, novel therapies such as antimicrobial implant coatings and antibiotic impregnated 3D-printed scaffolds represent promising strategies for preventing and managing this devastating disease. Here, we review these recent advances and highlight translational opportunities towards a cure.
