Hippo-YAP/TAZ signaling in breast cancer:Reciprocal regulation of.microRNAs and implications in precision medicine

Breast cancer is a molecularly heterogeneous disease and the most common f.male *** recent years,therapy approaches have evolved to accommodate molecular diversity,with a f.cus on more biologically based therapies to minimize negative *** regulate cell f.te in human breast cells,the Hippo signaling pathway has been associated with the alpha subtype of.estrogen *** pathway regulates tissue size,regeneration,and healing,as well as the survival of.tissue-specif.c stem cells,prolif.ration,and apoptosis in a variety of.organs,allowing f.r cell *** signaling is mediated by the kinases MST1,MST2,LATS1,and LATS2,as well as the adaptor proteins SAV1 and *** kinases phosphorylate the downstream ef.ectors of.the Hippo pathway,yes-associated protein(YAP),and transcriptional coactivator with PDZ-binding motif.TAZ),suppressing the expression of.their downstream target *** Hippo signaling pathway kinase cascade plays a signif.cant role in all *** the principles of.this kinase cascade would prevent the occurrence of.breast *** recent years,small noncoding RNAs,or microRNAs,have been implicated in the development of.several malignancies,including breast *** interconnections between miRNAs and Hippo signaling pathway core proteins in the breast,on the other hand,remain poorly *** this review,we f.cused on highlighting the Hippo signaling system,its key parts,its importance in breast cancer,and its regulation by miRNAs and other related pathways.

Towards implementation of.alloy-specif.c thermo-f.uid modelling f.r laser powder-bed f.sion of.Mg alloys

Multi-physics thermo-f.uid modeling has been extensively used as an approach to understand melt pool dynamics and def.ct f.rmation as well as optimizing the process-related parameters of.laser powder-bed f.sion(L-PBf..However,its capabilities f.r being implemented as a reliable tool f.r material design,where minor changes in material-related parameters must be accurately captured,is still in *** the present research,f.rst,a thermo-f.uid computational f.uid dynamics(Cf.)model is developed and validated against experimental *** the predicted material properties of.the pure Mg and commercial ZK60 and WE43 Mg alloys,parametric studies are done attempting to elucidate how the dif.erence in some of.the material properties,i.e.,saturated vapor pressure,viscosity,and solidif.cation range,can inf.uence the melt pool *** is f.und that a higher saturated vapor pressure,associated with the ZK60 alloy,leads to a deeper unstable keyhole,increasing the keyhole-induced porosity and evaporation mass *** viscosity and wider solidif.cation range can increase the non-unif.rmity of.temperature and velocity distribution on the keyhole walls,resulting in increased keyhole instability and f.rmation of.***,the WE43 alloy showed the best behavior in terms of.def.ct f.rmation and evaporation mass loss,providing theoretical support to the extensive use of.this alloy in *** summary,this study suggests an approach to investigate the ef.ect of.materials-related parameters on L-PBf.melting and solidif.cation,which can be extremely helpf.l f.r f.ture design of.new alloys suitable f.r L-PBf.

Tauopathies are a group of.neurological disorders,including Alzheimer’s disease and f.ontotemporal dementia,which involve progressive neurodegeneration,cognitive def.cits,and aberrant tau protein *** development of.tauopathies cannot currently be stopped or slowed down by treatment *** the signif.cant contribution of.tau burden in primary tauopathies and the strong association between pathogenic tau accumulation and cognitive def.cits,there has been a lot of.interest in creating therapies that can alleviate tau pathology and render neuroprotective ***,small molecules,immunotherapies,and gene therapy have been used to reduce the pathological tau burden and prevent neurodegeneration in animal models of.***,the major pitf.ll of.the current therapeutic approach is the dif.iculty of.drugs and gene-targeting modalities to cross the blood-brain barrier and their unintended side *** this review,the current therapeutic strategies used f.r tauopathies including the use of.oligonucleotide-based gene therapy approaches that have shown a promising result f.r the treatment of.tauopathies and Alzheimer’s disease in preclinical animal models,have been discussed.

Ef.ects of.grain's shape-and size-polydispersities,orientation,and area f.action on tortuosity and permeability of.2D granular media

The microstructure of.granular media, including grain's shape- and size-polydispersities, orientation, and area f.action can potentially af.ect its permeability. However, f.w studies consider the coupling ef.ects of.these f.atures. This work employs geometrical probability and stereology to establish quantitative relationships between the above microstructural f.atures and the geometric tortuosity of.the two-dimensional granular media containing superellipse, superoval, and polygon grains. Then the lattice Boltzmann method (LBM) is used to determine the permeabilities of.these granular media. By combining the tortuosity model and the LBM-derived permeabilities, modif.ed K–C equations are f.rmulated to predict the permeability and the shape f.ctor, considering the grain's shape- and size-polydispersities, orientation, and area f.action. The reliability of.these methods can be verif.ed by comparing them with both our simulations and available experimental, theoretical, and numerical data reported in the literature. The f.ndings implicate that the tortuosity and permeability of.the granular media are strongly correlated with the grain's shape, orientation, and area f.action but unaf.ected by the size polydispersity and spatial arrangement of.grains. Only circularity is not enough to derive a unif.ed f.rmula f.r considering the impact of.grain shape on tortuosity and permeability, other shape parameters need to be explored in the f.ture.

Engineered cardiac constructs(ECC)aid in the progression of.regenerative medicine,disease modeling and targeted drug delivery to adjust and aim the release of.remedial combination as well as decrease the side ef.ects of.*** this research,polycaprolactone/gold nanoparticles(PCL/GNPs)three-dimensional(3D)composite scaf.olds were manuf.ctured by 3D printing using the f.sed deposition modeling(f.M)method and then coated with gelatin/spironolactone(GEL/SPL).Scanning electron microscopy(SEM)and f.urier transf.rm-inf.ared spectroscopy(f.IR–ATR)were applied to characterize the ***,drug release,biodegradation,behavior of.the myoblasts(H9C2)cell line,and cytotoxicity of.the 3D scaf.olds were *** microstructural observation of.the scaf.olds reported interconnected pores with 150–300µm in *** 3D scaf.olds were degraded signif.cantly af.er 28 days of.immersion in stimulated body f.uid(SBf.,with the maximum rate of.GEL-coated 3D *** release f.om cross-linked GEL coating demonstrated the excess of.drug release over time,and according to the control release systems,the drug delivery systems(DDS)went into balance af.er the 14th *** addition,cell culture study showed that with the addition of.GNPs,the prolif.ration of.H9C2)was enhanced,and with GEL/SPL coating the cell attachment and viability were improved *** f.ndings suggested that PCL/GNPs 3D scaf.olds coated with GEL/SPL can be an appropriate choice f.r myocardial tissue engineering.

In-depth analysis of.VARTM-based solid-state supercapacitors utilizing CNT-dispersed cobalt-bismuth-samarium ternary hydroxide on woven carbon f.ber f.r enhanced energy storage

Multi-metal hydroxides possess unique physical and chemical properties,making them promising candidates f.r supercapacitor working *** their electrochemical perf.rmance can be achieved through a combination with carbon *** this study,we synthesized a composite material by hydrothermally dispersed 4,6,and 10 wt% carbon nanotubes(CNT) into ternary cobaltbismuth-samarium hydroxide(CoBiSm-TOH).These nanocomposites were employed as the material f.r the working electrode in a *** f.ndings reveal that at 1.5 A/g,the specif.c capacitance of.CNT3@CoBiSm-TOH,using a three-electrode system,was f.und to be 852.91 f.g,higher than that of.CoBi-BOH,CoBiSm-TOH,CNT1@CoBiSm-TOH and CNT5@CoBiSm-TOH-measuring 699.69,750.34,789.54 and 817.79 f.g,***,CNT3@CoBiSm-TOH electrodes exhibited a capacitance retention of.around 88% over 10,000 *** demonstrate practical applicability,CNT3@CoBiSm-TOH was grown on woven carbon f.ber(WCf.,and a solid-state supercapacitor device was developed using the VARTM(vacuum-assisted resin transf.r molding).This device displayed a specif.c capacitance of.272.67 f.g at 2.25 A/***,it achieved a maximum energy density of.53.01 Wh/kg at a power density of.750 W/kg and sustained excellent cycle stability over 50,000 cycles,maintaining 70% of.its initial *** results underscore the importance of.interf.cial nanoengineering and provide crucial insights f.r the development of.f.ture energy storage devices.

Precise taxonomy and the chronostratigraphic calibration of.the Middle Eocene Alveolina f.om Central Iran is here undertaken f.om the Chah-Talkh section of.the southern Sabzevar region(Central Iran). We have identif.ed Alveolina kieli, Alveolina stercusmuris and Alveolina nuttalli along with the new species Alveolina ozcani n. sp. that we include into the Alveolina elliptica group. We have also f.und Nummulites uroniensis, Nummulites obesus and Nummulites cf. verneuili and associated calcareous nannof.ssils that look reliable to make thoughtf.l correlations with the Shallow Benthic Zones(SBZ). The f.raminif.ral biostratigraphy suggests an assignment to the upper part of.the lower Lutetian-lower part of.the middle Lutetian,SBZ13(Middle Eocene), f.rther strengthened through the identif.cation of.the calcareous nannof.ssil NP14b-NP15b or CNE8-CNE10 biozones, providing a solid correlation with the global stratigraphic standards.
