Effect of Daxx on cholesterol accumulation in hepatic cells

AIM： To study the effect of Daxx on cholesterol accumulation in hepatic cells. METHODS： Sprague Dawley （SD） rats were fed a normal or high fat diet for 6 wk, and serum lipids and Daxx expression of hepatic tissues were measured by immunoblot assays. HepG2 cells were transfected with the pEGFP-C1/Daxx or pEGFP-C1 plasmid. Cells stably transfected with Daxx were identified by RTPCR analysis. Total cholesterol levels were determined by high performance liquid chromatography. Activated- SREBP and caveolin-1 were assayed by western blotting. RESULTS： Hepatic Daxx protein was higher in normal rats than in high fat diet-fed rats. Noticeable negative correlations were seen between Daxx and LDL-C （γ=-7.56, ρ=0.018）, and between Daxx and TC （γ=-9.07, ρ= 0.01）, respectively. The total cholesterol of HepG2/GFP-Daxx cells was lower than that of control cells or HepG2/GFP cells （9.28±0.19 vs 14.36± 4.45 or 13.94±2.62, both P 〈 0.05）. Furthermore, in HepG2/GFP cells, the expression of activated SREBP was lower than that of control cells, whereas caveolin-1 expression was higher. CONCLUSION： Overexpression of Daxx in HepG2 cells decreased intracellular cholesterol accumulation, which might be associated with inhibition of SREBP activity and an increase in caveolin-1 expression.

Background We have used suppression subtractive hybridization to construct a subtracted cDNA library of hepatocellular carcinoma （HCC） and isolated a panel of differential expression sequence tag （ESTs）. By using bioinformatics and rapid amplification of cDNA ends （RACE）, we found a novel HCC-associated gene IDD01. To further investigate its function, a recombinant eukaryotic vector pEGFP/ORF was constructed and transfected into the HepG2 cell line.Methods The open reading frame （ORF） of IDD01 was amplified by RT-PCR, digested with Bamh Ⅰ and Hind Ⅲ, and subcloned into the pEGFP-C1 vector. The ligation reaction was conducted with T4 DNA ligase, and the recombinant vector was named pEGFP/ORF. Untransfer control （control group）, pEGFP-C1 （HepG2/C1 group） and pEGFP/ORF （HepG2/ORF group） transfer groups were designed. Gene transfer was conducted with lipofectamine. To obtain stable transfection in HepG2 cells, selection was initiated with 500 μg/ml G418. Cellular IDD01 mRNA levels were assayed by semi-quantitative RT-PCR. The MTT colorimetric method and flow cytometry were used to determine the cell proliferation. The tumorigenic potential of transformed cells was determined from their ability to grow as anchorage-independent colonies on soft agar. Transient transfections were performed to observe subcellular location of GFP-IDD01 fusion protein.Results A 778 bp specific band of ORF was obtained by RT-PCR, and the positive clone of recombinant plasmid pEGFP/ORF （5.5 Kb） was identified by restriction endonuclease cleavage and sequence. The brightness ratio of IDD01 mRNA was not obvious between control and pEGFP/C1 groups, whereas the ratio of pEGFP/ORF was higher than that in the other two groups. After culture for 24-72 hours, the A490 values in pEGFP/ORF were higher than those in the other two groups （P〈0.01）. 目的探讨二甲双胍联合胰岛素对妊娠糖尿病(GDM)患者妊娠结局及血清中网膜素1(Omentin-1)、促代谢因子(Betatrophin)水平的影响。方法前瞻性纳入2019年1月至2021年12月沧州市人民医院收治的GDM患者400例作为研究对象,按随机数字表法将其分为观察组和对照组,每组各200例。两组均接受知识宣讲、饮食摄入控制、运动训练。对照组给予胰岛素注射液皮下注射,于餐前15~30 min注射,初次剂量0.2~0.3 U/(kg·d),3次/d。观察组在对照组基础上联合二甲双胍(1次/d,0.5 g/次)治疗。两组均治疗3周。比较两组血糖指标[空腹血糖、餐后2 h血糖(2 hPG)、糖化血红蛋白(GHb)],另外评估其氧化应激指标[晚期糖基化终末产物(AGEs)、血管细胞黏附分子-1(VCAM-1)、谷胱甘肽过氧化物酶(CSH-PX)]以及血清Omentin-1、Betatrophin水平,并比较两组不良妊娠结局。结果治疗后,两组空腹血糖、2 hPG、GHb水平均较治疗前降低,且观察组空腹血糖、2 hPG、GHb水平分别为(3.85±1.01)mmol/L、(6.12±0.86)mmol/L、(5.02±0.59)%,均低于对照组[(4.65±1.26)mmol/L、(7.85±1.22)mmol/L、(6.52±0.56)%],差异均有统计学意义(P<0.05)。治疗后,两组VCAM-1、AGEs水平均较治疗前降低,GSH-PX水平均较治疗前升高,且观察组VCAM-1、AGEs水平分别为(39.64±3.51)、(37.59±4.15)ng/L,均低于对照组[(52.15±4.02)、(42.59±5.06)ng/L],GSH-PX水平为(29.85±4.03)μmol/L,高于对照组[(21.86±3.46)μmol/L],差异均有统计学意义(P<0.05)。治疗后,两组血清Omentin-1、Betatrophin水平均较治疗前降低,观察组血清Omentin-1水平为(174.56±34.59)ng/mL,高于对照组[(151.58±36.45)ng/mL],Betatrophin水平为(572.56±85.49)pg/mL,低于对照组[(701.23±98.56)pg/mL],差异均有统计学意义(P<0.05)。观察组不良妊娠结局发生率为12.00%,低于对照组(19.50%),差异有统计学意义(P<0.05)。结论二甲双胍联合胰岛素治疗GDM不仅能有效改善患者血糖指标以及氧化应激指标,还能改善其血清Omentin-1、Betatrophin水平,具备安全性。

In search of effective and stable bifunctional electrocatalyst for electrocatalytic water splitting is still a major challenge for the highly efficient H_(2) ***,we reported a facile strategy to design high-indexed Cu_(3)Pd_(13)_S_(7) nanoparticles(NPs)in situ synthesized on the three-dimensional(3D)carbon nanofibers(CNFs)by combining electrospinning and chemical vapor deposition(CVD)*** high-index facets with abundant active sites,the 3D architecture CNFs with high specific surface area and synergistic effect of Cu-Pd-S bonds with strong electron couplings together promote the elec-trocatalytic *** Cu_(3)Pd_(13)_S_(7)/CNFs shows excellent electrocatalytic activity with low overpotentials of 52 mV(10 mA cm^(−2))for hydrogen evolution reaction(HER)and 240 mV(10 mA cm^(−2))for oxygen evolution reaction(OER).The excellent protection of Cu_(3)Pd_(13)_S_(7) by CNFs from aggregation and electrolyte corrosion lead to the high stability of Cu_(3)Pd_(13)_S_(7)/CNFs under acidic and alkaline conditions.

Optimization of Uncertain Structures with Interval Parameters Considering Objective and Feasibility Robustness

For the purpose of improving the mechanical performance indices of uncertain structures with interval parameters and ensure their robustness when fluctuating under interval parameters, a constrained interval robust optimization model is constructed with both the center and halfwidth of the most important mechanical performance index described as objective functions and the other requirements on the mechanical performance indices described as constraint functions. To locate the optimal solution of objective and feasibility robustness, a new concept of interval violation vector and its calculation formulae corresponding to different constraint functions are proposed. The math?ematical formulae for calculating the feasibility and objective robustness indices and the robustness?based preferential guidelines are proposed for directly ranking various design vectors, which is realized by an algorithm integrating Kriging and nested genetic algorithm. The validity of the proposed method and its superiority to present interval optimization approaches are demonstrated by a numerical example. The robust optimization of the upper beam in a high?speed press with interval material properties demonstrated the applicability and effectiveness of the proposed method in engineering.

AIM: To investigate the associations between the polymorphisms of cell cycle pathway genes and the risk of hepatocellular carcinoma(HCC). METHODS: We enrolled 1127 cases newly diagnosed with HCC from the Tumor Hospital of Guangxi Medical University and 1200 non-tumor patients from the First Affiliated Hospital of Guangxi Medical University. General demographic characteristics, behavioral information, and hematological indices were collected by unified questionnaires. Genomic DNA was isolatedfrom peripheral venous blood using Phenol-Chloroform. The genotyping was performed using the Sequenom Mass ARRAY i PLEX genotyping method. The association between genetic polymorphisms and risk of HCC was shown by P-value and the odd ratio(OR) with 95% confidence interval(CI) using the unconditional logistic regression after adjusting for age, sex, nationality, smoking, drinking, family history of HCC, and hepatitis B virus(HBV) infection. Moreover, stratified analysis was conducted on the basis of the status of HBV infection, smoking, and alcohol ***: The HCC risk was lower in patients with the MCM4 rs2305952 CC(OR = 0.22, 95%CI: 0.08-0.63, P = 0.01) and with the CHEK1 rs515255 TC, TT, TC/TT(OR = 0.73, 95%CI: 0.56-0.96, P = 0.02; OR = 0.67, 95%CI: 0.46-0.97, P = 0.04; OR = 0.72, 95%CI: 0.56-0.92, P = 0.01, respectively). Conversely, the HCC risk was higher in patients with the KAT2 B rs17006625 GG(OR = 1.64, 95%CI: 1.01-2.64, P = 0.04). In addition, the risk was markedly lower for those who were carriers of MCM4 rs2305952 CC and were also HBs Ag-positive and non-drinking and nonsmoking(P < 0.05, respectively) and for those who were carriers of CHEK1 rs515255 TC, TT, TC/TT and were also HBs Ag-negative and non-drinking(P < 0.05, respectively). The elastic scattering angular distributions of ^(13)C at 340 MeV and ^(14)C at 294 MeV and 342 MeV on a ^(208)Pb target,which correspond to approximately five times the Coulomb barriers,were measured at the Radioactive Ion Beam Line in *** data were analyzed within the optical model and continuum-discretized coupled-channels(CDCC)framework,and the results of both calculations could effectively account for the experimental *** differential cross sections of elastic scattering revealed no particular suppression at the Coulomb nuclear interference peak angles,suggesting that the breakup coupling effects on the elastic scattering angular distributions were negligibly small in this incident energy *** contributions from the couplings with inelastic states to the elastic cross sections were of minor importance within the angular range covered by these experiments.

To investigate the inhibitory effect of the Bcl-XL small interfering RNA(siRNA)on BcI-XLgene expression in the human gastric cancer cell line MGC-803,green fluorescent protein(GFP)siRNAwas constructed and transfected into MGC-803 ceils,together with GFP expression vector pTrace *** expression levels were observed using fluorescence ***-XL siRNA and negative siRNAwere then constructed and stably transfected into MGC-803 ***-PCR and immunofluorescence wereused to detect the expression of *** apoptosis was detected by acridine orange(AO)andflow *** were as follows:(1)48 h after GFP expression vector and GFP siRNA co-transfection,the expression level of GFP in the GFP siRNA group was much lower than the negative siRNA group,according to fluorescence microscopy *** mRNA and protein levels of Bcl-XL in Bcl-XL siRNAstable transfectants were reduced to almost background level compared with negative siRNA transfectantsor untreated cells.(2)Changes in nucleus morphology was observed by AO staining nucleic and flowcytometry analysis,which showed that stable Bcl-XL siRNA transfectants have an increased spontaneousapoptosis (21.17%+1.26% vs.1.19%+0.18% and 1.56%+0.15% respectively,P<0.05 *** siRNAor untreated control),siRNA targeting GFP or Bcl-XL genes can specifically suppress GFP or BcI-XLexpression in MGC-803 cells,and Bcl-XL siRNA can increase spontaneous ***-XL siRNA maybe a beneficial agent against human gastric adenocarcinoma.
