Wedelolactone attenuates sepsis-associated acute liver injury by regulating the macrophage M1/M2 polarization balance through the PI3K/AKT/NF-κB signalling pathway

Background:Liver injury caused by sepsis seriously impairs the normal physiology of the ***(WED)has an obvious anti-inflammatory effect against liver damage caused by various ***,further research is needed to determine if WED might mitigate acute liver damage linked to sepsis by influencing macrophage ***:We first assessed the effect of WED on lipopolysaccharides-triggered liver injury by biochemistry assay and tissue *** factors were assessed using the ELISA *** expression of Cluster of Differentiation 86(CD86)and Cluster of Differentiation 206(CD206)was measured by immunofluorescence *** protein levels of inducible nitric oxide sythase(iNOS),Arginase 1(Arg-1),phosphatidylinositol 3-kinase(PI3K),protein kinase B(AKT),PI3K phosphorylation(p-PI3K),AKT phosphorylation(p-AKT),inhibitor of kappa B kinase(IKK),inhibitor of kappa B(IκB),and nuclear factor kappa-B(NF-κB)p65 were quantified by western blot ***:WED decreased the level of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP)and malondialdehyde,and increased the activity of superoxide dismutase(SOD)and glutathione peroxidase(GSH-PX).Moreover,WED exerted effective anti-inflammatory effects by decreasing the level of Tumor necrosis factor-α(TNF-α)and Interleukin 6(IL-6)and increasing the level of Interleukin 10(IL-10)in serum and *** not only decreased CD86 and iNOS expression but also increased CD206 and Arg-1 *** also downregulated the increased expression of PI3K,AKT,p-PI3K,p-AKT,IKK,and NF-κB p65 induced by lipopolysaccharides,while up-regulated the decreased expression of Iκ***,LY294002 with WED decreased the expression of protein PI3K,AKT,p-PI3K,p-AKT,IKK and NF-κB p65,and raised the expression of IκBα.Conclusion:Wedelolactone could attenuate sepsis-associated acute liver injury,and its mechanism may be associated with balancing pro-inflammatory and anti-inflamm

An Improved Golden Jackal Optimization Algorithm Based on Multi-strategy Mixing for Solving Engineering Optimization Problems

Maternal weight and its association with risk of overweight in offspring:a trajectory analysis from a birth cohort in China

Background Most studies on the association of maternal pregnancy weight with offspring weight trajectory have a short follow-up *** study aimed to explore the associations of maternal prepregnancy body mass index(BMI)and gestational weight gain(GWG)with childhood weight trajectories in a 7-year birth *** A total of 946 mother–child pairs(467 boys and 479 girls)from a longitudinal birth cohort in Tianjin City,China,were included in this study,ranging from pregnancy to offspring at 7 *** outcome variable was defined as overweight or not overweight in offspring at the last round.A group-based trajectory model was applied to identify childhood BMI trajectory *** Five discrete BMI trajectory groups were identified and characterized as constant underweight(25.2%),constant normal weight(42.8%),and high or increasing trajectory[at risk of overweight(16.9%),progressive overweight(11.0%)and progressive obesity(4.1%)].Maternal prepregnancy overweight was associated with 1.72(95%CI 1.14–2.60,P=0.01)to 4.02(95%CI 1.94–8.36,P<0.001)times the risk of all high or increasing trajectory groups,and excessive GWG was related to groups at risk of overweight[relative risk ratio(RRR)2.09,95%CI 1.27–3.46,P=0.004]and progressive obesity(RRR 3.33,95%CI 1.13–9.79,P=0.029).Children in all high or increasing trajectory groups were associated with greater overweight risk at the last round[risk ratios(RRs)ranged from 3.54(95%CI 2.53–4.95,P<0.001)to 6.18(95%CI 4.05–9.42,P<0.001)].Conclusion Maternal prepregnancy overweight and excessive gestational weight gain were associated with increasing or high-level childhood body mass index trajectories as well as a greater risk of overweight at 7 years.

This study investigates the unsteady natural convection and entropy generation under the effects of magnetic field and baffles inside a nanofluid filled E-shaped *** nanofluid flow is driven by time-varying sidewall temperature and is partitioned by *** factors are discussed,including the enclosure aspect ratio(0.2≤AR≤0.7),nanofluid volume fractions(0≤φ≤0.1),Hartmann numbers(0≤Ha≤80),frequency of time-varying side wall temperature(0.01≤ω≤0.1),baffle locations(0≤d≤0.4)and length(0≤l≤0.4).An economic analysis is conducted to show the nanofluid cost of enhancing thermal transfer and reducing entropy *** modelling results show that increasing aspect ratio and nanofluid volume fraction enhance the thermal transfer behavior,while the magnetic field suppresses the nanofluid natural *** entropy generation monotonically decreases with the increasing nanofluid volume fraction and Hartmann *** baffles into horizontal walls can boost the thermal transfer behavior and decrease the total entropy generation.

Optimal Assimilation of Microwave Upper-Level Sounding Data in CMA-GFS

Various approaches have been proposed to minimize the upper-level systematic biases in global numerical weather prediction(NWP)models by using satellite upper-air sounding channels as ***,since the China Meteorological Administration Global Forecast System(CMA-GFS)has a model top near 0.1 hPa(60 km),the upper-level temperature bias may exceed 4 K near 1 hPa and further extend to 5 *** this study,channels 12–14 of the Advanced Microwave Sounding Unit A(AMSU-A)onboard five satellites of NOAA and METOP,whose weighting function peaks range from 10 to 2 hPa are all used as anchor observations in *** is shown that the new“Anchor”approach can effectively reduce the biases near the model top and their downward propagation in three-month assimilation *** bias growth rate of simulated upper-level channel observations is reduced to±0.001 K d^(–1),compared to–0.03 K d^(–1)derived from the current dynamic correction *** relatively stable bias significantly improves the upper-level analysis field and leads to better global medium-range forecasts up to 10 days with significant reductions in the temperature and geopotential forecast error above 10 hPa.

Relationship between the Stratospheric Arctic Vortex and Surface Air Temperature in the Midlatitudes of the Northern Hemisphere

It is known that different relationships exist between the strength and displacement of the stratospheric polar vortex(SPV),and the surface air temperature(SAT)patterns in Eurasia and North America,but the mechanisms behind these relationships remain unclear,especially on an interannual *** on empirical orthogonal function(EOF)analysis using NCEP reanalysis data over 1958–2018,this study attempts to ascertain the relationship between the SPV intensity and displacement over the Arctic and the SATs in the midlatitudes of the Northern *** results indicate that a strengthened SPV corresponds to an SAT increase in Eurasia and a decrease in eastern North America and *** the SPV is shifted towards Eurasia,however,a corresponding SAT increase occurs in both North America and Eurasia,with a larger increase in North America than in ***,a strengthened SPV tends to correspond to a positive North Atlantic Oscillation-like circulation in the troposphere with negative geopotential height(GH)anomalies in Greenland and eastern North American continent and positive GH anomalies to the north of 45°N in Eurasia,which corresponds to lower SATs in North America than in ***,when the SPV shifted towards Eurasia,it was accompanied by a positive Pacific/North American-like pattern with a deepened Aleutian low,which corresponds to the increasing SATs in North *** tropospheric circulation changes are related to the response of tropospheric planetary wave activity to the SPV.A strengthened SPV corresponds to the weakening of tropospheric planetary wave-1 waves,which is accompanied by a negative GH in North America but a positive GH in *** the SPV shifted towards Eurasia,the tropospheric planetary wave-1(-2)waves strengthened(weakened),and the combined effects of the planetary wave-1 and wave-2 waves would cause positive GH anomalies in both Eurasia and North America.

A Weakly-Supervised Crowd Density Estimation Method Based on Two-Stage Linear Feature Calibration

In a crowd density estimation dataset,the annotation of crowd locations is an extremely laborious task,and they are not taken into the evaluation *** this paper,we aim to reduce the annotation cost of crowd datasets,and propose a crowd density estimation method based on weakly-supervised learning,in the absence of crowd position supervision information,which directly reduces the number of crowds by using the number of pedestrians in the image as the supervised *** this purpose,we design a new training method,which exploits the correlation between global and local image features by incremental learning to train the ***,we design a parent-child network(PC-Net)focusing on the global and local image respectively,and propose a linear feature calibration structure to train the PC-Net simultaneously,and the child network learns feature transfer factors and feature bias weights,and uses the transfer factors and bias weights to linearly feature calibrate the features extracted from the Parent network,to improve the convergence of the network by using local features hidden in the crowd *** addition,we use the pyramid vision transformer as the backbone of the PC-Net to extract crowd features at different levels,and design a global-local feature loss function(L2).We combine it with a crowd counting loss(LC)to enhance the sensitivity of the network to crowd features during the training process,which effectively improves the accuracy of crowd density *** experimental results show that the PC-Net significantly reduces the gap between fullysupervised and weakly-supervised crowd density estimation,and outperforms the comparison methods on five datasets of Shanghai Tech Part A,ShanghaiTech Part B,UCF_CC_50,UCF_QNRF and Jhu-CROWD++.
