Climate,female traits and population features as drivers of breeding timing in Mediterranean red deer populations

Understanding the factors that lead to variation in the timing of breeding in widespread species such as red deer(Cervus elaphus)is crucial to predict possible responses of wild populations to different climate ***,we sought to further understand the causes of inter-annual variation in the reproduction timing of female deer in Mediterranean *** integrative approach was used to identify the relative importance of individual,population and climate traits in the date of conception of free-ranging deer,based on a dataset of 829 hinds culled during 12 *** found that a population trait,density,was the most important factor explaining the variation in conception dates,with greater densities causing later conception *** mass was the second in importance,with heavier females conceiving earlier than lighter *** equally important was the spring real bioclimatic index,a measure of plant productivity,causing later conception dates in the least productive springs(drier and hotter).Another climatic component,the end of summer drought,showed that the sooner the autumn arrives(greater rainfalls and cooler temperatures)the earlier the conception ***,age class was found to be a minor factor in determining conception *** older females(≥10 years old)conceived significantly later,suggesting reproductive *** study highlights not only the importance of population and individual traits but also the influence of climatic parameters on the deer reproductive cycle in Mediterranean environments,giving valuable insight into how reproductive phenology may respond to seasonality and global climate changes.

Overview of the New OMAH Technique for Scaling OMA Mode Shapes

Methods for scaling mode shapes determined by operational modal analysis(OMA)have been extensively investigated in the last years.A recent addition to the range of methods for scaling OMA mode shapes is the so-called OMAH technique,which is based on exciting the structure by harmonic forces applied by an *** applying harmonic forces in at least one degree-of-freedom(DOF),and measuring the response in at least one response DOF,while using at least as many frequencies as the number of mode shapes to be scaled,the mode shape scaling(modal mass)of all modes of interest may be *** previous publications on the method the authors have proven that the technique is easy and robust to apply to both small scale and large scale ***,it has been shown that the technique is capable of scaling highly coupled modes by using an extended multiple reference *** present paper summarizes the theory of the OMAH method and gives recommendations of how to implement the method for best *** is pointed out,as has been shown in previous papers,that the accuracy of the mode scaling is increased by using more than one response DOF,and by selecting DOFs with high mode shape *** determine the harmonic force and responses,it is recommended to use the three-parameter sine fit *** is shown that by using this method,the measurement time can be kept short by using high sampling frequency and bandpass filtering whereas spectrum based methods require long measurement *** means that even for structures with low natural frequencies,the extra measurement time for scaling the mode shapes can be kept relatively short.

Type 1 diabetes(T1D),also referred to as insulin-dependent diabetes mellitus,is a debilitating disease that follows the destruction of pancreatic insulin-producingβcells by autologous T-cells.T1D primarily affects children and has a strong genetic component,with>50 susceptibility loci identified,including HLA-DQβchains.1 However,the striking differences between European regions with similar genetic backgrounds and a sharp rise in T1D incidence in developed countries over the last several decades suggest that environmental factor(s)also play a relevant etiopathogenic role.2 Given the accumulating evidence linking the gut microbiota to protection against metabolic diseases,3 Mariño et al.4 recently tested the hypothesis that short-chain fatty acids(SCFAs),which are the end products of fermentation of dietary fibers by anaerobic intestinal microbiota,can protect genetically susceptible mice from developing ***,the authors found that diets enriched in acetate or butyrate(two of the main SCFAs)can protect animals from developing diabetes through different and complementary cellular *** findings significantly enhance our understanding of the role of diet and gut microbiota in the development of autoimmune diseases and indicate that the use of medicinal foods may be a cost-effective treatment against T1D and other autoimmune diseases with a cellular *** that current anti-T1D approaches(which focus on prevention or modulation of the adaptive specific immune response against autoantigens)have been generally disappointing,5 such a new and refreshing strategy has attracted a great deal of attention(reviewed in Ref.2).

Planetary boundaries delimit a‘safe operating space for humanity’that should not be overstepped to maintain stable Holocene-like conditions on *** chemical pollutants have the potential to pose a planetary boundary threat to the functioning of vital Earth system processes as so-called‘novel entities’.Recently,an exposure-based prioritization scheme was developed that uses model-estimated data on persistence and degree of mobility in air and water to identify and prioritise chemical substances that may be planetary boundary *** a case study,chemicals of emerging Arctic concern identified by the Arctic Monitoring and Assessment Programme(AMAP)were used to develop the *** exposure-based prioritization scheme cannot address all scenarios for a chemical to pose an unknown planetary boundary threat,but it does allow for prioritization of chemicals according to environmental fate and exposure profiles.A key limitation of the scheme is that the quantity of chemical potentially released to the environment was not considered in the ***,we use data on chemical production and use reported under the European chemicals regulation REACH,and an emission scoring system developed by the European Food Safety Authority(EFSA)to attempt to add information about estimated emissions to the exposure-based prioritization *** REACH data and the EFSA scoring system,we ranked the AMAP substances according to their potential for environmental release and combined that information with the previously reported exposure-based prioritization that considers persistence and long-range transport *** method successfully assigned‘high priority’to known ozone-depleting substances(methyl chloride and dichloromethane)and identified chemicals of concern recently under consideration for nomination as persistent organic pollutants under the Stockholm Convention(octamethylcyclotetrasiloxane and decamethylcyclopentasiloxane).The EFSA scoring system offers a

One extraction tool for in vitro-in vivo extrapolation?SPME-based metabolomics of in vitro 2D,3D,and in vivo mouse melanoma models

Solid phase microextraction(SPME)in combination with high-resolution mass spectrometry was employed for the determination of metabolomic profile of mouse melanoma growth within in vitro 2D,in vitro 3D,and in vivo *** multi-model approach had never been investigated *** to the low-invasiveness of SPME,it was possible to perform time-course analysis,which allowed building time profile of biochemical reactions in the studied *** approach does not require the multiplication of samples as subsequent analyses are performed from the very same cell culture or from the same *** already reduces the number of animals required for experiment;therefore,it is with good concordance with the 3Rs rule(replacement,reduction,and refinement).Among tested models,the largest number of compounds was found within the in vitro 2D cell culture model,while in vivo and in vitro 3D models had the lowest number of detected *** results may be connected with a higher metabolic rate,as well as lower integrity of the in vitro 2D model compared to the in vitro 3D model resulting in a lower number of compounds released into medium in the latter *** terms of in vitro-in vivo extrapolation,the in vitro 2D model performed more similar to in vivo model compared to in vitro 3D model;however,it might have been due to the fact that only compounds secreted to medium were ***,in further experiments to obtain full metabolome information,the intraspheroidal assessment or spheroid dissociation would be necessary.

Diflubenzuron inhibits reproduction of different strains of Drosophila melanogaster

The effect of （1-（4-chlorophenyl）-3-（2,6-difluorobenzoyl）urea） insecticide - diflubenzuron - on wild type and white type fruit flies Drosophila melanogaster （Meigen, 1830） was studied. Adult insects were placed in vials with different concentrations of the insecticide in the nutrient （from 0.048 to 48 mg of diflubenzuron per 1 cm^3 of the nutrient）. In each case, the insects showed some mortality, which was concentrationdependant. When comparing both strains, we could observe different acute toxicities, with wild type being more resistant to diflubenzuron. However, subacute toxicity was similar within both strains. In both strains the prolonged exposure led to the extinction of the majority of the populations （〉 70%）. Although imagoes could freely copulate, we did not observe copulating fruit flies, and we did not find either eggs or larvae in any of the exposed groups. These results indicate that genetic mutation which leads to different eye colour can also affect resistance and survival of insects in pesticide-exposed areas.

Rockfalls are one of the most common instability processes in high *** represent a relevant issue,both for the risks they represent for(infra)structures and frequentation,and for their potential role as terrestrial indicators of climate *** study aims to contribute to the growing topic of the relationship between climate change and slope instability at the basin *** selected study area is the Bessanese glacial basin(Western Italian Alps)which,since 2016,has been specifically equipped,monitored and investigated for this *** order to provide a broader context for the interpretation of the recent rockfall events and associated climate conditions,a cross-temporal and integrated approach has been *** this purpose,geomorphological investigations(last 100 years),local climate(last 30 years)and near-surface rock/air temperatures analyses,have been carried *** research outcomes show that rockfalls occurred in two different geomorphological positions:on rock slopes in permafrost condition,facing from NW to NE and/or along the glacier margins,on rock slopes uncovered by the ice in the last *** thaw of the active layer and/or glacier debutressing can be deemed responsible for slope failure *** regard to timing,almost all dated rock falls occurred in *** the July events,initiation may have been caused by a combination of rapid snow melt and enhanced seasonal thaw of the active layer due to anomalous high temperatures,and *** events are,instead,associated with a significant positive temperature anomaly on the quarterly scale,and they can be ascribed to the rapid and/or in depth thaw of the permafrost active *** to our findings,we can expect that in the Bessanese glacierized basin,as in similar high mountain areas,climate change will cause an increase of slope instability in the *** fasten knowledge deepening,we highlight the need for a growth of a network of high elevation

RET in breast cancer:pathogenic implications and mechanisms of drug resistance

Initiation,progression,outcome and sensibility to therapies in breast cancer(BC),the most frequent cancer in women,are driven by somatic and germline *** the effectiveness of hormonal therapies is well-founded,it is prescribed for cancers which express steroid hormone receptors,such as estrogen receptor(ER).RET is a proto-oncogene encoding a transmembrane tyrosine kinase receptor that is activated by one of its four ligands(GDNF,neurturin,artemin or persephin)and one of its coreceptors(Gfrα1-4).Loss-of-function mutations in RET are responsible for Hirschsprung disease,while gain-of-function mutations for multiple endocrine neoplasia type *** addition,deregulation of its intracellular signaling,due to mutations,gene rearrangements,overexpression or transcriptional upregulation,can cause several neuroendocrine and epithelial *** BC,amplification of receptor tyrosine kinases,such as ERBB2,EGFR,IGFR and FGFR1,and/or their upregulation contribute to cancer initiation and *** can also have an important role in BC,but only in the subset of ER-positive(ER+)tumors,where it is found *** the RET pathway and shedding light on molecular basis of the resistance to hormone therapy may lead to new therapies in ER+BC,improving treatment outcome and preventing tumor-related ***,here,we review the state of the art of RET biology in BC and agents targeting RET tested in the clinical trials and discuss the specificity of the still available RET inhibitors and the molecular mechanisms underlying the BC resistance to endocrine therapy.

The composition of Fusarium species in wheat husks and grains in south-eastern Poland

Fusarium populations were investigated on 53 samples of wheat grains and husks collected approximately three weeks before harvest in 53 wheat fields in south-eastern Poland. A limited area of sample collection was chosen intentionally to avoid the effect of climate and weather variability. The study was conducted to assess the occurrence of 13 Fusarium species using species-specific PCR assays separately on grains and husks of winter wheat. The obtained data suggest that husks could take a protective role of wheat grain against Fusarium spp. The incidence of Fusarium species decreased in grains vs. husks from 29 to 100%. While Fusarium species were present in husks at 11.32% and less, they were absent in the grain. The presence of Fusarium species on husks is inversely proportional to the percentage reduction of Fusarium spp. in grain. There was a correlation of the presence of certain species of Fusarium in husks and in grains. The number of Fusarium species found on husks was about three times higher in comparison to wheat grain. In conclusion, the presented data indicate the importance of Fusarium populations analysis on wheat husk in seeds pathological studies.
