The flhDC gene affects motility and biofilm formation in Yersinia pseudotuberculosis

The flagella master regulatory gene flhDC of Yersinia pseudotuberculosis serotype III (YPIII) was mutated by deleting the middle region; replaced by a tetracycline resistant gene,; the subsequent mutant strain named YPIIIΔflhDC was obtained. Swimming assay showed that the swimming motility of the mutant strain was completely abolished. The promoter region of the flagella second-class regulatory gene fliA was fused with the lux box,; was conjugated with the mutant; the parent strains respectively for the first cross. LUCY assay result demonstrated that flhDC regulated the expression of fliA in YPIII as reported in E. coli. Biofilm formation of the mutant strain on abiotic; biotic surfaces was observed; quantified. The results showed that mutation of flhDC decreased biofilm formation on both abiotic; biotic surfaces,; abated the infection on Caenorhabdtis elegans. Our results suggest that mutation of the flagella master regulatory gene flhDC not only abolished the swimming motility, but also affected biofilm formation of YPIII on different surfaces. The new function of flhDC identified in this study provides a novel viewpoint for the control of bacterial biofilm formation.

Climate change significantly impacts human health,exacerbating existing health inequalities and creating new *** study addresses the lack of systematic review in this area by analyzing 2.40 publications,focusing on four key terms:health,disparities,environmental factors,and climate *** inclusion criteria limited the selection to English-language,peer-reviewed articles related to climate health hazards,ensuring the relevance and rigor of the synthesized *** process synthesized 65 relevant *** investigation revealed that recent research,predominantly from developed countries,has broadened its scope beyond temperature-related impacts to encompass diverse climate hazards,including droughts,extreme weather,floods,mental health issues,and the intersecting effects of Coronavirus Disease *** has highlighted exposure as the most studied element in the causal chain of climate change-related health inequalities,followed by adaptive capability and inherent *** most significant vulnerabilities were observed among populations with low socioeconomic status,ethnic minorities,and *** study further reveals research biases and methodological limitations,such as the paucity of attention to underdeveloped regions,a narrow focus on non-temperature-related hazards,challenges in attributing climate change effects,and a deficit of large-scale empirical *** findings call for more innovative research approaches and a holistic integration of physical,socio-political,and economic dimensions to enrich climate-health discourse and inform equitable policy-making.

Objective The activities and products of carbohydrate metabolism are involved in key processes of ***,its relationship with hepatocellular carcinoma(HCC)is *** The cancer genome atlas(TCGA)-HCC and ICGC-LIRI-JP datasets were acquired via public *** expressed genes(DEGs)between HCC and control samples in the TCGA-HCC dataset were identified and overlapped with 3.5 carbohydrate metabolism-related genes(CRGs)to obtain differentially expressed CRGs(DE-CRGs).Then,univariate Cox and least absolute shrinkage and selection operator(LASSO)analyses were applied to identify risk model genes,and HCC samples were divided into high/low-risk groups according to the median risk ***,gene set enrichment analysis(GSEA)was performed on the risk model *** sensitivity of the risk model to immunotherapy and chemotherapy was also *** A total of 8 risk model genes,namely,G6PD,PFKFB4,ACAT1,ALDH2.ACYP1,OGDHL,ACADS,and TKTL1,were ***,the risk score,cancer status,age,and pathologic T stage were strongly associated with the prognosis of HCC *** the stromal score and immune score had significant negative/positive correlations with the risk score,reflecting the important role of the risk model in immunotherapy ***,the stromal and immune scores had significant negative/positive correlations with risk scores,reflecting the important role of the risk model in immunotherapy ***,we found that high-/low-risk patients were more sensitive to 102.drugs,suggesting that the risk model exhibited sensitivity to chemotherapy *** results of the experiments in HCC tissue samples validated the expression of the risk model *** Through bioinformatic analysis,we constructed a carbohydrate metabolism-related risk model for HCC,contributing to the prognosis prediction and treatment of HCC patients.

Internetware can be formed through aggregation of Internetware entities bottom-up to satisfy the user's requirements. In the process, capability description of Internetware entity plays an important role. This paper proposes a capability description of Internetware entity based on environment ontology. The environment ontology includes environment entities and possible interactions with the environment entities in a particular domain. Tree-like hierarchical state machines are also included in the ontology to describe domain life-cycle of the environment entities. On the basis of the ideas, the capability of Internetware entity is embodied by interacting with environment entities and the changes of the environment entities during the interactions. Finally, this paper describes the matchmaking between capability descriptions of Internetware entities to support the discovery of Internetware entity. An example of online education course is given to illustrate the ideas.

By using BCC_RegCM 1.0 (RegCM for short) from beijing Climate Center (BCC), china Meteorological Administration (CMA), the Meiyu season characters over the Yangtze-Huaihe region during 1991-2.05 are simulated. The major conclusions of this study may be summarized as the following: (1) RegCM can reproduce the interannual variation and the spatial distribution of the summertime precipitation and temperature in the Yangtze-Huaihe region. (2. By use of a generalized Meiyu criterion and in accordance with model-calculated precipitation and temperature, the Meiyu onset and en{2. date have been determined. Compared with the observation, RegCM can simulate the interannual variation of the Yangtze-Huaihe Meiyu with preferable capability for most of the normal Meiyu years (such as 1995, 1997, 2.00, 2.01, 2.02.and 2.04), especially for the rich Meiyu years of 1996, 1998 and 1999. (3. In terms of the average simulation for the recent 15 years, the timings of onset and en{2. of Meiyu occur on June 1 and July 13. respectively, which are earlier than the climatological observation. For duration, Meiyu persists for 3.2}d, 3.d shorter than the observation. The index of Meiyu intensity is 2.45, while the climatological one is 3.00. Therefore, RegCM is capable of simulating the climatological Meiyu duration and intensity, while the capability of simulating the onset and en{2. date of Meiyu still needs to be improved.

The crystallization of uniaxial hot drawn syndiotactic polystyrene/multi-walled carbon nanotube （sPS/MWCNT） nanocomposite films was studied by FTIR spectroscopy. The effects of MWCNT content, draw ratio and drawing temperature on the sPS crystallinity were investigated. The sPS/MWCMT nanocomposite films show reduced crystallinities with the increase of MWCNT content. In addition, with the increase of draw ratio, both the pure sPS and the sPS/MWCNT nanocomposite drawn films exhibit increased crystallinity. The effect of drawing temperature on the sPS crystallization is complex. In a temperature range of 100―13. °C, the crystallinity decreases with drawing temperature, whereas it increases at 140 °C for both pure sPS and its nanocomposite films.

Calibrating the HXMT collimators using diffuse illumination

We propose a new method to calibrate the HXMT collimators by measuring the optical point spread function(PSF) of the hard X-ray modulation telescope(HXMT).The light transmission of the collimator at different viewing angles with a camera and a diffuse backlight placed behind the collimator is *** method is much easier to accomplish than measuring the PSF with a parallel optical *** experimental results are very consistent with the *** PSF of the collimator of the high energy X-ray telescope on HXMT is found to be in good agreement with the design,with accuracy better than 1 arcmin.
