Transcriptional regulation of the neuronal nitric oxide synthase gene(nNOS)is particularlycomplex as 12.distinct transcripts derived from different first exons are expressed in a tissue- and cell-specific *** exon 1f mRNA is relatively highly expressed in nervous system and relies upon exon1f-specific promoter *** conventional and real-time reverse transcription-polymerase chain reaction,we found exon 1f mRNA was the major transcript of the nNOS gene in human neuroblastoma *** analyzed a 1090 bp fragment of 1f promoter by TRANSFAC-TESS and Match softwares andluciferase assay,and found an important positive transcriptional regulation region that contained a putativenuclear factor(NF)-κB binding ***,using electrophoresis mobility shift and chromatinimmunoprecipitation assays,we identified this site to be the NF-κB responsive element,a crucial positiveregulator in the activation of the nNOS 1f *** together,our study identified an NF-κB responsiveelement within nNOS 1f promoter and showed that it plays an important role in the transactivation of nNOS1f mRNA,the major transcript of nNOS in SK-N-SH cells.

Aim: Anticonvulsant tolerance and dependence are two obstacles that restrict the clinical use of benzodiazepines (BDZ). In order to explore the mechanism of these two adverse reactions, changes of neuropeptide Y (NPY) and its receptors in the hippocampus of rat models, in relation to flurazepam (FZP, a member of BDZ) tolerance and dependence, were investigated. Methods: The mRNA of preproNPY and its receptors (Y1, Y2. and Y5) in the hippocampus were determined by competitive RT-PCR, and the distribution of NPY in the hippocampus was examined by immunohistochemistry. Results: A decrease of preproNPY mRNA in the hippocampus was found in tolerant and dependent rats. The level of preproNPY mRNA in the hippocampus was reversely correlated with the degree of tolerance and dependence, measured by the threshold of pentylenetetrazol-induced seizures. Immunohistochemistry indicated a decrease of NPY-immunoreactive material in neurons of the CA1, CA3, and dentate gyrus regions of both tolerant and dependent rats. The mRNA of NPY receptors Y,and Y5decreased in tolerant rats but did not change in dependent rats. The mRNA of NPY receptor Y2.ncreased in tolerant rats but decreased in dependent rats. Conclusion: A decrease of NPY in the hippocampus might be involved in anticonvulsant tolerance and dependence following long-term treatment with FZP. Y,, Y2. and Y5mRNA were also altered in FZP tolerance and dependence.

Generalized gradient projection neural network models are proposed to solve nonsmooth convex and nonconvex nonlinear programming problems over a closed convex subset of R n . By using Clarke’s generalized gradient, the neural network modeled by a differential inclusion is developed, and its dynamical behavior and optimization capabilities both for convex and nonconvex problems are rigorously analyzed in the framework of nonsmooth analysis and the differential inclusion theory. First for nonconvex optimization problems, the quasi convergence results similar to those achieved by previous work are proved. For convex optimization problems, the global convergence results are proved, i.e. any trajectory of the neural network converges to an asymptotically stable equilibrium point, which is an optimal solution of the primal problem whenever it exists. This result remains valid even if the initial condition is chosen outside the feasible set. In addition, an asymptotic control result involving a Tykhonov like regularization shows that any trajectory of the revised neural network can be forced to converge toward a particular optimal solution of the primal problem. Finally, two simulation optimization algorithms for solving the optimization and control problems are designed respectively, and three typical simulation experiments are illustrated to present the accuracy and efficiency of theoretical convergence results of this paper.

Objective To explore the association between polymorphism in the ACE I/D gene and blood pressure-lowering response to hydrochlorothiazide （HCTZ） in 82. patients. Methods HCTZ 12.5 mg was taken once a day for six weeks. The blood pressure reduction and ratio reaching target blood pressure were compared in different ACE genotype groups. Results The reduction in SBP of patients carrying DD was greater than that in other groups carrying II or ID （12.2.mmHg versus 5.4 mmHg, 12.2.mmHg versus 4.4 mmHg, respectively, P〈0.05）. The reduction in MAP of patients carrying DD was also greater than that in other groups carrying II or ID （6.9 mmHg versus 3.9 mmHg, 6.9 mmHg versus 3.6 mmHg, respectively, P〈0.05）. The ratio reaching target blood pressure in DD groups was significantly higher than that in II or ID groups （P〈0.05）. The pre-treatment SBP, DD genotype, aldosterone levels entered the multi-linear regression model significantly and might affect the reduction of SBP. This paper is concerned with the analysis of a feedback M^[X]/G/1 retrial queue with starting failures and general retrial times. In a batch, each individual customer is subject to a control admission policy upon arrival. If the server is idle, one of the customers admitted to the system may start its service and the rest joins the retrial group, whereas all the admitted customers go to the retrial group when the server is unavailable upon arrival. An arriving customer （primary or retrial） must turn-on the server, which takes negligible time. If the server is started successfully （with a certain probability）, the customer gets service immediately. Otherwise, the repair for the server commences immediately and the customer must leave for the orbit and make a retrial at a later time. It is assumed that the customers who find the server unavailable are queued in the orbit in accordance with an FCFS discipline and only the customer at the head of the queue is allowed for access to the server. The Markov chain underlying the considered queueing system is studied and the necessary and sufficient condition for the system to be stable is presented. Explicit formulae for the stationary distribution and some performance measures of the system in steady-state are obtained. Finally, some numerical examples are presented to illustrate the influence of the parameters on several performance characteristics.

Based on the facts that cations and anions of molten salts and binary basic oxide solid solutions do not separate from each other as well as that the electric conductivities of mattes are greater than that of salts and basic oxide solid solutions, the calculating models of mass action concentrations for binary and ternary mattes involving eutectic have been formulated. The results of calculation agree well with the values measured;this in turn shows that the calculating models can reftect the structural characteristics of the mattes concerned.

Aim:To study the effects(and the mechanisms thereof)of Ganoderma lucidumpolysaccharides(G/-PS)on the proliferation and the anti-tumor activity of cytokine-induced killer(CIK)cells,and to make use of CIK cells as a means to investigatethe interactions between Gl-PS and ***:CIK cells were preparedby using the standard protocol as a positive *** groups alsounderwent the standard protocol,except that Gl-PS(400 mg/L or 100 mg/L)wasadded and the dose of anti-CD3 and interleukin-2.they received was reduced by50% and 75%,*** negative controls,Gl-PS in the experimentalprotocol was replaced with soluble starch or methylcellulose(400 mg/L or 100 mg/L).CIK cell proliferation,cytotoxicity,and phenotype were determined by using theTrypan blue exclusion method,MTT assay,and flow ***:Bysynergizing cytokines,Gl-PS(400 mg/L or 100 mg/L)could decrease the amountof cytokine in lymphokine activated killer(LAK)cells and CIK cells culture,buthad no significant effect on the proliferation,cytotoxicity,or phenotype of LAKcells,or CIK cells induced by cytokines at higher doses alone,in which CIK cellsexpanded about 80-fold and the main effectors,CD3~+NK1.1~+ cells,expanded bymore than 15%.The cytotoxicity of CIK cells in experimental groups was 79.3%±4.7%,76.9%±6.8% versus the positive control 80.7%±6.8% against P815(P>0.05)and 88,9%±5.5%,84.7%±7.9% versus the positive control 89.8%±4.5% againstYAC-1(P>0.05).The activity of GI-PS could mostly be blocked by ***:GI-PS was shown to be a promising biological response modifier andimmune *** effect of GI-PS on CIK cells is possibly mediated prima-rily through complement receptor type 3.

Aim:To investigate the effects of angiotensin receptor blocker (ARB) telmisartanon the expression and distribution of protein kinase C (PKC)-α in the kidneys ofdiabetic ***:Diabetic mice were induced with streptozotocin and agroup of them were randomly selected for treatment with *** 6weeks,the expression and localization of PKC-α in the renal cortex,and the outerand inner medulla were assessed by immunohistochemistry and semiquantitativeWestern *** addition,expressions of PKC-α,transforming growth factor-β1 (TGF-β1),and vascular endothelial growth factor (VEGF) in glomeruli weremeasured by semiquantitative ***:Diabetic and nor-mal mice showed similar distributions of PKC-α in the *** expression ofPKC-α was found in glomeruli,epithelial cells of proximal tubules,and medullary-collecting duct,while not in the medullary and cortical thick ascending limb,andwas different in the epithelial cells of proximal tubules of diabetic nephropathy(DN) mice,PKC-α was mostly translocated from the basement membrane to theapical membrane,whereas it was largely translocated from the apical membrane tothe basement membrane in epithelial cells of the inner medullary-collecting *** blotting detected increased expression of PKC-α in the renal cortex andouter medulla,but not in the inner medulla of DN *** expressions ofPKC-α,TGF-β1,and VEGF were shown in the glomeruli of DN mice,where PKC-αexhibited a correlation to VEGF,but no correlation to TGF-β*** telmisartanattenuated alterations of PKC-α as mentioned earlier in the DN ***:Our findings suggest that PKC-α may play a role in the pathogenesis of DN,andthat the nephroprotective effects of ARB telmisartan may be partly associatedwith its influence on PKC-α.
