Introduction: Adolescent HPV vaccination rates remain suboptimal. The purpose of the study was to investigate attitudes about HPV vaccine relative to other adolescent vaccines among clinical staff from primary care offices and school based clinics. methods: We interviewed clinicians in primary care offices and school-based clinics regarding their attitudes about HPV vaccine relative to Tdap and mCV4. Results: Respondents (n = 36) included clinical staff in family medicine (47%), pediatrics (25%), obstetrics/gynecology (19%) and school-based health clinics (8%). Only 3% strongly agreed and 17% agreed that completion of HPV vaccine was more important than completion of pertussis vaccine (Tdap), while 6% strongly agreed and 33% agreed that completion of HPV vaccine was more important than completion of meningitis vaccine (mCV4). Discussion: Providing clinicians with additional information about the cancer prevention benefits of the HPV vaccine and the greater risk for HPV infection/disease relative to other vaccine preventable adolescent diseases may help to increase HPV vaccination rates among adolescents.

琼东南盆地深水区中新世以来构造运动不活跃,自中新世以来沉积物厚度达5 000～9 000m,Ya35-1-2井显示更新世地层沉积物主要以泥质与砂泥互层的细粒沉积物为主,深部流体垂向运移通道不发育。高分辨率地震资料和3D地震资料表明,琼东南盆地的上新世及其以上地层存在大量气烟囱和块体搬运体系。气烟囱为深部流体垂向运移提供通道,沉积物在搬运过程中发生重塑,由于沉积和压实作用使mTD具有相对较高电阻率、高密度异常和渗透率降低,是烃类气体聚集的良好盖层,有利于天然气水合物形成。

Introduction: The rise in antibiotic resistant cases has caused a global concern. Researchers around the world are trying to find a novel alternative to combat this issue. Green tea with its many health benefits, including antibacterial and antiviral activity, has shown to be one of the most promising candidates to be used as an agent to solve this problem. Objective: This study focuses on evaluating the synergistic effects of antibiotics and two green tea polyphenols: epigallocatechin gallate (EGCG), and its modified lipophilic form epigallocatechin gallate stearate (EGCG-S). methods: In this study, twelve antibiotics and eight bacteria: Gram-positive Staphylococcus aureus (S. aureus), Staphylococcus epidermidis (S. epidermidis) and Bacillus megaterium (B. megaterium);Gram-negative Escherichia coli (E. coli), Pseudomonas aeruginosa (P. aeruginosa), Serratia marcescens (S. marcescens), and Enterobacter aerogenes (E. aerogenes);and acid-fast mycobacterium smegmatis (m. smegmatis) were used. Antibacterial synergism profiling of EGCG, EGCG-S and antibiotics has been established using a disk diffusion assay. Results: The results revealed that both 1% of EGCG and 1% EGCG-S enhanced the antimicrobial activities on antibiotics in various bacteria. Antimicrobial susceptibility study indicated that EGCG-S was able to enhance some antibiotics from the resistant category to intermediate or susceptible and/or from intermediate category to susceptible. Both EGCG and EGCG-S worked comparably on Gram-positive bacteria;in S. aureus, both compounds enhanced 5 antibiotics (Am10, CF30, C30, S10 and TE30) activities while EGCG-S had higher efficiency. B. megaterium were susceptible to most of the antibiotic treatment, thus the impact of EGCG and EGCG-S was insignificant. EGCG-S worked better than EGCG on Gram-negative bacteria;converted 9 antibiotics susceptibility in E. coli and P. aeruginosa, and 8 antibiotics in E. aerogenes. 目的通过基因打靶建立c-myb+/+和c-myb+/-胚胎干细胞(ES)模型,了解转录因子c-myb在造血定型和分化中的作用。方法采用去除白血病抑制因子的甲基纤维素体外ES细胞诱导分化体系进行造血分化,收获c-myb+/+和c-myb+/-(表达55%的c-myb)胚胎体进行甲基纤维素克隆形成分析。运用实时定量PCR比较分析c-myb+/+和c-myb+/-造血分化过程的造血相关基因表达。结果c-myb+/+和c-myb+/-ES细胞胚胎体形成相似,但c-myb+/-组胚胎体体积小于c-myb+/+组。两组红系克隆形成单位(CFU-E)克隆形成相似,但c-myb+/-组CFU-E克隆小于c-myb+/+组。两组红系爆式克隆形成单(BFU-E)均出现于第7天,高峰在第10天。两组巨噬系克隆形成单位(CFU-m)形成相似,但c-myb+/-组CFU-m数目多于c-myb+/+组。c-myb+/-组粒-单系克隆形成单位(CFU-Gm)形成数目少于c-myb+/+组。PCR分析显示c-myb+/+和c-myb+/-组β-globin、ζ-globin、GATA3、CD34、Lys、c-fms和c-mpl基因表达没有明显差异。结论亚表达水平的c-myb可以满足祖细胞扩增的需要,但却影响其终末分化。亚表达水平c-myb影响前体细胞向红系和粒系的分化,但不影响巨噬细胞的分化。c-myb表达水平不影响β-globin、ζ-globin、GATA3、CD34、Lys、c-fms和c-mpl基因的表达。

Investigation of Wave-Structure Interaction Using State of the Art CFD Techniques

The suitability of computational fluid dynamics (CFD) for marine renewable energy research and development and in particular for simulating extreme wave interaction with a wave energy converter (WEC) is considered. Fully nonlinear time domain CFD is often considered to be an expensive and computationally intensive option for marine hydrodynamics and frequency-based methods are traditionally preferred by the industry. However, CFD models capture more of the physics of wave-structure interaction, and whereas traditional frequency domain approaches are restricted to linear motions, fully nonlinear CFD can simulate wave breaking and overtopping. Furthermore, with continuing advances in computing power and speed and the development of new algorithms for CFD, it is becoming a more popular option for design applications in the marine environment. In this work, different CFD approaches of increasing novelty are assessed: two commercial CFD packages incorporating recent advances in high resolution free surface flow simulation;a finite volume based Euler equation model with a shock capturing technique for the free surface;and meshless Smoothed Particle Hydrodynamics (SPH) method. These different approaches to fully nonlinear time domain simulation of free surface flow and wave structure interaction are applied to test cases of increasing complexity and the results compared with experimental data. Results are presented for regular wave interaction with a fixed horizontal cylinder, wave generation by a cone in driven vertical motion at the free surface and extreme wave interaction with a bobbing float (The manchester Bobber WEC). The numerical results generally show good agreement with the physical experiments and simulate the wave-structure interaction and wave loading satisfactorily. The grid-based methods are shown to be generally less able than the meshless SPH to capture jet formation at the face of the cone, the resolution of the jet being grid dependent.

Using directed mutagenesis and phage display on a soluble fragment of the human immunoglobulin superfamily receptor ILT2(synonyms:LIR1,mIR7,CD85j),we have selected a range of mutants with binding affinities enhanced by up to 168,000-fold towards the conserved region of major histocompatibility complex(mHC)class I *** in a dimeric form,either by chemical cross-linking with bivalent polyethylene glycol(PEG)derivatives or as a genetic fusion with human IgG Fc-fragment,the mutants exhibited a further increase in ligand-binding strength due to the avidity effect,with resident half-times(t1/2)on the surface of mHC I-positive cells of many *** novel compounds antagonized the interaction of CD8 co-receptor with mHC I in vitro without affecting the peptide-specific binding of T-cell receptors(TCRs).In both cytokine-release assays and cell-killing experiments the engineered receptors inhibited the activation of CD8^(+)cytotoxic T lymphocytes(CTLs)in the presence of their target cells,with subnanomolar potency and in a dose-dependent *** a selective inhibitor of CD8^(+)CTL responses,the engineered high affinity ILT2 receptor presents a new tool for studying the activation mechanism of different subsets of CTLs and could have potential for the development of novel autoimmunity therapies.

Effects of transdermal magnesium chloride on quality of life for patients with fibromyalgia: a feasibility study

BACKGROUND： Fibromyalgia is a syndrome characterized by chronic pain, fatigue, depression, and sleep disturbances. Its primary cause is unclear. Several studies have reported decreased intracellular magnesium levels in patients with fibromyalgia and have found negative correlation between magnesium levels and fibromyalgia symptoms.OBJECTIVE： To gather preliminary data on whether transdermal magnesium can improve quality of life for women who have fibromyalgia. DESIGN, SETTING, PARTICIPANTS AND INTERVENTIONS： This is a patient questionnaires and survey in a fibremyalgia clinic at a tertiary medical center. Forty female patients with the diagnosis of fibremyalgia were enrolled. Each participant was provided a spray bottle containing a transdermal magnesium chloride solution and asked to apply 4 sprays per limb twice daily for 4 weeks. Participants were asked to complete the Revised Fibromyalgia Impact Questionnaire, SF-36v2 Health Survey, and a quality-of-life analog scale at baseline, week 2, and week 4. mAIN OUTCOmE mEASURE： Questionnaire and survey scores, evaluated through intent-to-treat and per-protocol analyses. RESULTS： Twenty-four patients completed the study （mean [SD] age, 57.2 [7.6] years; white, 95%; mean body mass index, 31.3 kg/m2）. With intention-to-treat analysis, Revised Fibromyalgia Impact Questionnaire subscale and total scores were significantly improved at week 2 and week 4 （total score, P = 0.001）. Per-protocol analysis results were similar： all subscales of the Revised Fibremyalgia Impact Questionnaire were significantly improved at week 2 and week 4 （total score, P = 0.001）. CONCLUSION： This pilot study suggests that transdermal magnesium chloride applied on upper and lower limbs may be beneficial to patients with fibromyalgia.
