The Role of Phosphorylation in Redox Regulation of Photosynthesis Genes psaA and psbA during Photosynthetic Acclimation of Mustard

The long-term response （LTR） to light-quality gradients improves performance and survival of plants in dense stands. It involves redox-controlled transcriptional regulation of the plastome-encoded genes psaAB （encoding the P700 apoproteins of photosystem I） and psbA （encoding the D1 protein of photosystem II） and requires the action of plastidlocalized kinases. To study the potential impact of phosphorylation events on plastid gene expression during the LTR, we analyzed mustard seedlings acclimated to light sources favoring either photosystem I or photosystem II. Primer extension analyses of psaA transcripts indicate that the redox regulation occurs at the principal bacterial promoters, suggesting that the plastid encoded RNA polymerase （PEP） is the target for redox signals. Chloroplast protein fractions containing PEP and other DNA-binding proteins were purified from mustard via heparin-Sepharose chromatography. The biochemical properties of these fractions were analyzed with special emphasis on promoter recognition and specificity, phosphorylation state, and kinase activity. The results demonstrate that the LTR involves the action of small DNA-binding proteins; three of them exhibit specific changes in the phosphorylation state. Auto-phosphorylation assays, in addition, exhibit large differences in the activity of endogenous kinase activities. Chloroplast run-on transcription experiments with the kinase inhibitor H7 and the reductant DTT indicate that phosphorylation events are essential for the mediation of redox signals toward psaA and psbA transcription initiation, but require the synergistic action of a thiol redox signal. The data support the idea that redox signals from the thylakoid membrane are linked to gene expression via phosphorylation events; however, this mediation appears to require a complex network of interacting proteins rather than a simple phosphorelay.

13-Lipoxygenases(LOXs)initiate the synthesis of jasmonic acid(JA),the best-understood oxylipin hormone in herbivory ***,the roles of 9-LOX-derived oxylipins in insect resistance remain ***,we report a novel anti-herbivory mechanism mediated by a tonoplast-localized 9-LOX,ZmLOX5,and its linolenic acid-derived product,9-hydroxy-10-oxo-12(Z),15(Z)-octadecadienoic acid(9,10-KODA).Transposon-insertional disruption of ZmLOX5 resulted in the loss of resistance to insect ***5 knockout mutants displayed greatly reduced wound-induced accumulation of multiple oxylipins and defense metabolites,including benzoxazinoids,abscisic acid(ABA),and JA-isoleucine(JA-Ile).However,exogenous JA-Ile failed to rescue insect defense in lox5 mutants,while applications of 1 mM 9,10-KODA or the JA precursor,12-oxo-phytodienoic acid(12-OPDA),restored wild-type resistance *** profiling revealed that exogenous 9,10-KODA primed the plants for increased production of ABA and 12-OPDA,but not *** none of the 9-oxylipins were able to rescue JA-Ile induction,the lox5 mutant accumulated lower wound-induced levels of Ca^(2+),suggesting this as a potential explanation for lower wound-induced *** pretreated with 9,10-KODA exhibited rapid or more robust woundinduced defense gene *** addition,an artificial diet supplemented with 9,10-KODA arrested fall armyworm larvae ***,analysis of single and double lox5 and lox10 mutants showed that ZmLOX5 also contributed to insect defense by modulating ZmLOX10-mediated green leaf volatile ***,our study uncovered a previously unknown anti-herbivore defense and hormonelike signaling activity for a major 9-oxylipin α-ketol.

Effects of Different Levels of Garlic Powder Incorporated in the Feed Enriched with Stylosanthes guianensis on Reproduction and Pre-Weaning Growth Performances of Cavies (Cavia porcellus)

In order to increase the productivity of the local cavies as a meat animal, a study was carried out between November 2021 and March 2022 at the Research and Experimental Farm, Animal Nutrition and Production Research Unit of the Faculty of Agronomy and Agricultural Sciences (FAAS) and the Microbiology and Anti-Microbial Research Unite of the University of Dschang on 72 cavies aged between 4 and 5 months and weighing an average of 450 ± 50 g produced on the farm, including 60 females and 12 males. The females were distributed according to a completely randomized factorial design in twelve (12) rearing boxes numbered G1 to G12, with 15 females per group distributed in 03 boxes and receiving iso-nitrogenated experimental diet (18% protein) R0 enriched with 20% of Stylosanthes guianensis (0% garlic powder), R0.25 (0.25% garlic powder), R0.50 (0.50% garlic powder) and R0.75 (0.75% garlic powder). Females were identified by numbered ear tags and mated with a sex ratio of 5 females for 1 male. Drinking water enriched with vitamin C (01 tablet of 240 mg in 1.5 liters of water) was served ad libitum and renewed daily. The males were removed after 31 days. A form was established to record the weight of each pregnant female, any abortions during gestation, the date of parturition and the date of birth of each newborn, as well as the number of live-born or stillborn pups. After parturition, the weights of female and newborn cavies were recorded weekly until the 3rd week. The number of cavies that died before weaning and the number of live weaned cavies were also recorded. Reproductive performance significantly increased (p 0.05) fertility rate and net fertility rate, litter size, birth viability rate, and pre-weaning viability rate with rations containing garlic powder, compared to the control ration, especially with the R0.50 ration. Effects of submergence in water on seed germination and vigor of the Copaifera lucens(Fabaceae) seedlings

We analyzed the tolerance of Copaifera lucens seeds to sub- mersion in water to assess the use of this species for direct seeding in riparian forest restoration programs. Seeds were submerged in water for 2, 4, 8, 16 and 32 days or not submerged （control = 0 days of submer- gence）. For the control and at the end of each period of submersion, germination and seedling vigor tests were carried out. For germination tests, seeds were sown in plastic pots containing sand and kept in labora- tory conditions. The percentage of seed germination, the germination rate and the average germination time were analyzed. For seedlings, total biomass, leaf area, leaf mass per area and leaf area ratio were analyzed. Submersion time drastically affected the dissolved oxygen content and seed germination. Between 4 and 8 days of submersion there was a de- crease from 83.8% to 15.6% in the germination percentage. No seed germination occurred after 16 days of submersion. Although there was a significant decrease in the percentage of seed germination between 4 and 8 days of submersion, seedling vigor was not affected. Seeds of this species were partially tolerant to submersion in water, suggesting that C. lucens is a promising species for direct seeding in riparian forest restora- tion projects.

A detailed Pendulum generators analysis was conducted on various architectures including rotational,pendulum,linear,sliding,cantilever,flexible blade,multidimensional and 1 cm magnetoelectric,and the following hybrid technologies. They enable highly e cient ways to harvest electric energy from many forms of vibrational,rotational,biomechanical,wave,wind and thermal sources,among others. Open-circuit voltages up to 75 V,short-circuit currents up to 60 mA and instantaneous power up to 144 mW were already achieved by these nanogenerators. Their transduction mechanisms,including proposed models to make intelligible the involved physical phenomena,are also overviewed here. A comprehensive analysis was performed to compare their respective construction designs,external excitations and electric outputs. The results highlight the potential of hybrid E-TENGs to convert unused mechanical motion into electric energy for both large-and small-scale ***,this paper proposes future research directions toward optimization of energy conversion e ciency,power management,durability and stability,packaging,energy storage,operation input,research of transduction mechanisms,quantitative standardization,system integration,miniaturization and multi-energy hybrid cells.

Analysis of the relation between organic products consumption and box schemes use in Alicante (Spain)

Box-schemes or organic food subscription plans consist on a regular delivery of a box of seasonal fruits and/or vegetables, preferably organic, cultivated in the vicinity at a specified point using subscription, The object of the present work is trying to examine whether there is a relationship between consumer knowledge about these short distribution channels and the consumption of organic products in the province of Alicante. A survey has been conducted among 400 usual food buyers in this Spanish province. It has been found that the consumption of organic products and the knowledge of such initiatives are linked. We recommend to companies that use this type of distribution channel to publicize its benefits.
