Genetic and phenotypic analyses reveal major quantitative loci associated to fruit size and shape traits in a non-flat peach collection ( P. persica L. Batsch)

Fruit size and shape are critical agronomical and pomological attributes and prime targets in peach breeding *** from the flat peach type,a Mendelian trait well-characterized at the genetic level,ample diversity of fruit size and shapes is present across peach ***,knowledge of the underlying genomic loci remains *** this work,fruit size and shape were assessed in a collection of non-flat peach accessions and selections,under controlled fruit load *** architecture of these traits was then dissected by combining association and linkage mapping,revealing a major locus on the proximal end of chromosome 6(qSHL/Fs6.1)explaining a large proportion of phenotypic variability for longitudinal shape and also affecting fruit size.A second major locus for fruit longitudinal shape(qSHL5.1),probably also affecting fruit size,was found co-localizing at locus G,suggesting pleiotropic effects of peach/nectarine *** additional QTL for fruit longitudinal shape(qSHL6.2)was identi fied in the distal end of chromosome 6 in a cross with an ornamental double-flower peach and co-localized with the Di2 locus,controlling flower *** assisting breeding activities,knowledge of loci controlling fruit size and shape paves the way for more in-depth studies aimed at the identification of underlying genetic variant(s).

Endopolygalacturonase (endoPG) plays a pivotal role in determining peach [Prunus persica L. (Batsch)] fruit characteristics. Different Pp-endoPG genes or allelic variants have been described, characterized by different polymorphisms: insertions-deletions (InDels) and single nucleotide polymorphisms (SNPs). Eighty-five peach accessions (comprising commercial cultivars, F1 progenies of selected crosses, and three haploid seedlings) with different flesh softening patterns (Non Melting: NM;Melting: M;Slow Softening: SS;Stony Hard: SH) were screened by exploiting specific polymorphisms, with the aim to characterize their asset at the endoPG locus and evaluate a potential relationship with fruit flesh texture phenotype. The results of InDel analysis allowed to distinguish, by a simple genotyping procedure, NM flesh phenotypes from the others. Further information arose from this analysis, showing that two Pp-endoPG genes, i.e., Pp-endoPGm (Ppa006839m), involved in the determination of the Melting/Non Melting trait, and Pp-endoPG_M (Ppa006857m), involved in the determination of the Clingstone/Freestone trait, always co-segregate, and that SS Big Top possesses a “null” Pp-endoPG allele. Cleaved Amplified Polymorphic Sequence (CAPS) analysis allowed to preliminarily discriminate the Pp-endoPG variants of the SS and SH accessions considered. The integrated use of the considered polymorphisms in a high number of peach accessions proved useful, by individuating the different gene variants and their combinations, to describe the structure of the endoPG locus in different genotypes.

Despite the availability of whole genome sequences of apple and peach,there has been a considerable gap between genomics and *** bridge the gap,the European Union funded the FruitBreedomics project(March 2011 to August 2015)involving 28 research institutes and private *** complementary approaches were pursued:(i)tool and software development,(ii)deciphering genetic control of main horticultural traits taking into account allelic diversity and(iii)developing plant materials,tools and methodologies for *** breakthroughs were made including the making available of ready-to-go DNA diagnostic tests for Marker Assisted Breeding,development of new,dense SNP arrays in apple and peach,new phenotypic methods for some complex traits,software for gene/QTL discovery on breeding germplasm via Pedigree Based Analysis(PBA).This resulted in the discovery of highly predictive molecular markers for traits of horticultural interest via PBA and via Genome Wide Association Studies(GWAS)on several European genebank *** also developed pre-breeding plant materials in which multiple sources of resistance were pyramided and software that can support breeders in their selection *** FruitBreedomics,significant progresses were made in the field of apple and peach breeding,genetics,genomics and bioinformatics of which advantage will be made by breeders,germplasm curators and scientists.A major part of the data collected during the project has been stored in the FruitBreedomics database and has been made available to the *** review covers the scientific discoveries made in this major endeavour,and perspective in the apple and peach breeding and genomics in Europe and beyond.

The ongoing research on pioneering drug candidates for the overactive bladder （OAB） aimed to overcome the limitations of currently licensed pharmacotherapies, such as antimuscarinics, β3-adrenergic agents, and botulinum neurotoxin, has been reviewed performing a systematic literature review and web search. The review covers the exploratory agents alternative to available medications for OAB and that may ultimately prove to be therapeutically useful in the future mana-gement of OAB patients based on preclinical and early clinical data. It emerges that many alternative pharmacological strategies have been discovered or are under investigation in disease-oriented studies. Several potential therapeutics are known for years but still fnd obstacles to pass the clinical stages of development, while other completely novel compounds, targeting new pharmacological targets, have been recently discovered and show potential to translate into clinical therapeutic agents for idiopathic and neurogenic OAB syndrome. The global scenario of investigational drugs for OAB gives promise for the development of innovative thera-peutics that may ultimately prove effective as first, combined or second-line treatments within a realistic timescale of ten years.

The ability to generate complex optical photon states involving entanglement between multiple optical modes is not only critical to advancing our understanding of quantum mechanics but will play a key role in generating many applications in quantum *** include quantum communications,computation,imaging,microscopy and many other novel technologies that are constantly being ***,approaches to generating parallel multiple,customisable bi-and multi-entangled quantum bits(qubits)on a chip are still in the early stages of ***,we review recent advances in the realisation of integrated sources of photonic quantum states,focusing on approaches based on nonlinear optics that are compatible with contemporary optical fibre telecommunications and quantum memory platforms as well as with chip-scale semiconductor *** new and exciting platforms hold the promise of compact,low-cost,scalable and practical implementations of sources for the generation and manipulation of complex quantum optical states on a chip,which will play a major role in bringing quantum technologies out of the laboratory and into the real world.
