The aim of the present paper is to characterize bioinspired chitosan （CS）＋ hydroxyapatite （HA） coatings with various components ratio on a zirconium alloy with titanium. The coatings were characterized by FT-IR, SEM, hydrophilic/hydrophobic balance, adherence, roughness, electrochemical stability and in vitro cell response. Electrochemical tests, including potentio- dynamic polarization curves and electrochemical impedance spectroscopy, were performed in normal saline physiological solution. Cell viability of MC3T3-E1 osteoblasts, lactate dehydrogenase, nitric oxide, and Reactive Oxygen Species （ROS） levels, as well as actin cytoskeleton morphology, were evaluated as biological in vitro tests. The results on in vitro cell response indicated good cell membrane integrity and viability for all samples, but an increased cell number, a decreased ROS level and a better cytoskeleton organization were noticed for the sample with a higher CS content. The coating with highest CS concen- tration indicated the best performance based on the experimental data. The highest hydrophilic character, highest resistance to corrosion and best biocompatibility as well recommend this coating for bioapplications in tissue engineering.

One of the factors promoting tumoral progress is the abnormal activation of the epithelial-mesenchymal transition(EMT)program which has been associated with chemoresistance in tumoral *** transcription factor zinc finger E-box-binding homeobox 1(ZEB1),a key EMT activator,has recently been related to docetaxel resistance,the main chemotherapeutic used in advanced prostate cancer *** mechanisms involved in this protective effect are still *** a previous work,we demonstrated that ZEB1 expression induced an EMT-like phenotype in prostate cancer cell *** this work,we used prostate cancer cell lines 22Rvl and DU145 to study the effect of ZEB1 modulation on docetaxel resistance and its possible *** results showed that ZEB1 overexpression conferred to 22Rvl cell resistance to docetaxel while its silencing made DU145 cells more sensitive to *** of resistance markers showed no presenee of ATP-binding cassette subfamily B member 1(MDR1)and no changes in breast cancer resistance protein(BCRP)or ATP-binding cassette subfamily C member 10(MRP7).However,a correlation between ZEB1,multidrug resistance-associated protein 1(MRP1),and ATP-binding cassette subfamily C member 4(MRP4)expression was ***4 inhibition,using MK571,resensitized cells with ZEB1 overexpression to docetaxel *** addition,modulation of ZEB1 and subsequent change in MRP4 expression correlated with a lower apoptotic response to docetaxel,characterized by lower B-cell lymphoma 2(Bcl2),high BCL2-associated X protein(Bax),and high active caspase 3 *** response to docetaxel in our model seems to be mediated mainly by activation of the apoptotic death *** results showed that modulation of MRP4 could be a mediator of ZEBl-related resistance to docetaxel in prostate cancer,making it a possible marker for chemotherapy response in patients who do not express MDR1.

It has been reported that one of the factors that promotes tumoral progression is the abnormal activation of the epithelial-mesenchymal transition program. This process is associated with tumoral cells acquiring invasive and malignant properties and has the transcription factor zinc finger E-box-binding homeobox 1 （ZEB1） as one of its main activators. However, the role of ZEB1 in promoting malignancy in prostate cancer （PC, a） is still unclear. Here, we report that ZEB1 expression correlates with Gleason score in PCa samples and that expression of ZEB1 regulates epithelial-mesenchymal transition and malignant characteristics in PCa cell lines. The results showed that ZEB1 expression is higher in samples of higher malignancy and that overexpression of ZEB1 was able to induce epithelial-mesenchymal transition by upregulating the mesenchymal marker Vimentin and downregulating the epithelial marker E-Cadherin. On the contrary, ZEB 1 silencing repressed Vimentin expression and upregulated E-Cadherin. ZEB1 expression conferred enhanced motility and invasiveness and a higher colony formation capacity to 22Rvl cells whereas DU145 cells with ZEB1 silencing showed a decrease in those same properties. Neonates are at increased risk of viral encephalopathies that can result in neurological dysfunction, seizures, permanent disability and even death. The neurological damage results from the combined effect of the virus and the immune response it elicits, thus finding tools to facilitate viral clearance from central nervous system （CNS） while minimizing neuron damage remains a critical challenge. Neonatal mice inoculated intraperitoneally with Tacaribe virus （TCRV） develop seizures, hindlimb paralysis and death within 15 days of inoculation. TCRV localizes to the CNS within days of challenge, primarily infecting astrocytes in the cerebellum and brain stem. We show that infection leads to inflammation, T cell and monocyte infiltration into the cerebellar parenchyma, apoptosis of astrocytes, neuronal degeneration and loss of Purkinje cells. Infiltrating antigen-specific T cells fail to clear the virus but drive the disease, as T-cell-deficient CD3ε KO mice survive TCRV infection with minimal inflammation or clinical manifestations despite no difference in CNS viral loads in comparison with T-cell sufficient mice. CD8＋ T cells drive the pathology, which even in the absence of CD4＋ T-cell help, infiltrate the parenchyma and mediate the apoptotic loss of cerebellar astrocytes, neurodegeneration and loss of Purkinje cells resulting in loss of balance, paralysis and death. CD4＋ T cells are also pathogenic inducing gliosis and inflammation in the cerebellum and cerebrum that are associated with wasting and death several weeks after CD4＋ T-cell transfer. These data demonstrate distinct pathogenic effects of CD4＋ and CD8＋ T cells and identify them as possible therapeutic targets.

Several medical conditions can present as fever and rash in children,including infections,hypersensitivity reactions and immune-mediated diseases[1].Cryopyrin-associated periodic syndrome(CAPS)is a group of inherited autoinflammatory disorders caused by gain-of-function mutations in the NLRP3 gene(CIAS 1).CAPS encompass a spectrum of diseases including Muckle-Wells syndrome(MWS).The A439V mutation is considered a disease-causing mutation,although clinical phenotypes can be diverse[2].MWS is characterized by recurrent episodes of fever,urticarial rash,arthralgia/arthritis,and more rarely,oral ulcers,folliculitis,or erythema nodosum[3].

Home-to-school routes are very sensitive areas: they represent, for children, a learning tool for their everyday activities, but if poorly designed, maintained and equipped they can expose them to traffic risks. Sidewalks' inappropriate level of service and poor maintenance, especially, are main factors contributing to walking unsuitability, thus to poor comfort and safety levels for young pedestrians, and more in general for all the vulnerable non-motorized road users. This paper deals with a methodology specifically developed to highlight the quality of the urban environment where the home-to-school routes are located, according to four main criteria: wellbeing, usage, appearance, and safety and security. Each criterion is associated with a checklist including the most relevant features to assess, with a focus on maintenance as a key parameter to create safe and comfortable routes to school. An application, a case study in Rome, where the lack of regular maintenance results into a network of unsafe sidewalks, is also presented, analyzing three different areas where a number of schools are located. Detours when approaching school premises were surveyed, due to the levels of distresses and linked to the poor comfort and safety levels. As the mutual influence of built environment over road safety and maintenance requirements for home-to-school paths is not largely investigated thus far, the paper's goal is to provide advanced knowledge for studies and applications further afield.

Neglected features of lifestyle: Their relevance in nonalcoholic fatty liver disease

AIMTo investigated in non-alcoholic-fatty-liver-disease (NAFLD), with ultrasound (US)-detected fatty liver, and in a group of non-alcoholic and otherwise healthy subjects, relationship of neglected features of lifestyle with NAFLD and obesity. METHODSFive hundred and thirty-two NAFLD and 667 non-NAFLD healthy subjects, age 21-60 years were studied. Severity of liver steatosis was assessed by US bright liver score. The adherence to mediterranean diet score (AMDS) was assessed on the basis of a 1-wk recall computerized questionnaire which included a detailed physical activity reports (Baecke questionnaire). The western dietary profile score, as a simplified paradigm of unhealthy diet, a questionnaire quantifying sun exposure score and a sleep habits questionnaires provided a further comprehensive lifestyle assessment. RESULTSBody mass index (BMI), insulin resistance (HOMA), and triglycerides, poorer adherence to a mediterranean diet profile, sedentary habits, minor sun exposure and use of “western diet” foods are greater in NAFLD. Multiple linear regression analysis, weighted by years of age, displays BMI, HOMA and AMDS as the most powerful independent predictors of fatty liver severity; however, also the physical activity score, the western diet habit and the sun exposure score are acting inside the model with significant independent effects. CONCLUSIONArticulated clinical intervention, according to our results, are justified in NAFLD and can be pursued addressing by focused intervention nutritional profile, physical exercise mainly in open-air subsets for enhancing sun exposure and healthier sleep duration and rhythm.
