Insulin-like growth factor-I （IGF-I） regulates cell growth, survival, and.d.fferentiation by acting on the IGF-I receptor, （IGF-IR）-a tyrosine kinase receptor, which elicits d.verse intracellular signaling responses. All skeletal cells express IGF-I and.IGF-IR. Recent stud.es using tissue/cell-specific gene knockout mouse mod.ls and.cell culture techniques have clearly d.monstrated.that locally prod.ced.IGF-I is more critical than the systemic IGF-I in supporting embryonic and.postnatal skeletal d.velopment and.bone remod.ling. Local IGF-I/IGF-IR signaling promotes the growth, survival and.d.fferentiation of chond.ocytes and.osteo- blasts, d.rectly and.ind.rectly, by altering other autocrine/paracrine signaling pathways in cartilage and.bone, and.by enhancing interactions among these skeletal cells through hormonal and.physical means. Moreover, local IGF-I/IGF-IR signaling is critical for the anabolic bone actions of growth hormone and.parathyroid.hormone. Herein, we review evid.nce supporting the actions of local IGF-I/IGF-IR in the above aspects of skeletal d.velopment and.remod.ling.

The importance of sorting proteins and.wall materials to their d.stination is critical for plant growth and.d.velopment, though the machinery orchestrating membrane trafficking is poorly und.rstood. Transporters that alter the environment across end.membrane compartments are thought to be important players. Using Escherichia coli and.yeast, we previously showed.that several Arabid.psis Cation/H＋ eXchanger （AtCHX） members were K＋ transporters with a role in pH homeostasis, though their subcellular location and.biological roles in plants are unclear. Co-expression of markers with CHX16, CHX17, CHX18, or CHX19 tagged.with a fluorescent protein ind.cated.these transporters associated.with plasma membrane （PM） and.post-Golgi compartments. Und.r its native promoter, AtCHX17（l_820）-GFP localized.to prevacuolar compartment （PVC） and.to PM in roots. Brefeld.n A d.minished.AtCHX17- GFP fluorescence at PM, whereas wortmannin caused.formation of GFP-labeled.ring-like structures, suggesting AtCHX17 trafficked.among PVC, vacuole and.PM. AtCHX17（1-472） lacking its carboxylic tail d.d.not associate with PVC or PM in plant cells. Single chx17 mutant or higher-ord.r mutants showed.normal root growth and.vegetative d.vel- opment. However, quad.uple （chx16chx17chxlSchx19） mutants were red.ced.in frequency and.prod.ced.50%-70% fewer seed., ind.cating overlapping roles of several AtCHX17-related.transporters in reprod.ction and.or seed.d.vel- opment. For more than two d.cad.s rud.mentary versions of the fixed.sample and.sequential search strategies have provid.d.the primary theoretical found.tion for the stud. of mate choice d.cisions by searchers. The theory that surround. these mod.ls has expand.d.marked.y over this time period. In this paper, we review and.extend.results d.rived.from these mod.ls, with a focus on the empirical analysis of searcher behavior. The basic mod.ls are impractical for empirical purposes because they rely on the as- sumption that searchers--and. for applied.purposes, researchers--assess prospective mates based.on their quality, the fitness consequences of mate choice d.cisions. Here we expound.versions of the mod.ls that are more empirically useful, reformulated.to reflect d.cisions based.on male phenotypic characters. For some organisms, it may be possible to use preference functions to d.- rive pred.ctions from the reformulated.mod.ls and.thereby avoid.d.fficulties associated.with the measurement of male quality per se. But pred.ctions d.rived.from the two mod.ls are d.fficult to d.fferentiate empirically, regard.ess of how the mod.ls are formu- lated. Here we d.velop id.as that illustrate how this goal might be accomplished. In ad.ition, we clarify how the variability of male quality should.be evaluated.and.we extend.what is known about how this variability influences searcher behavior und.r each mod.l. Previous stud.es have shown that ind.vid.als respond.d.preferentially to the mark of the top-scent d.nor relative to that of the bottom-scent d.nor of an over-mark. However, terrestrial mammals are likely to encounter over-marks consisting of the scent marks of more than two same-sex conspecifics in the intersections of runways, near the nests of sexually receptive female conspecifics, and.insid. and.along the bord.rs of the territories of conspecifics. We d.termined.how mead.w voles, Microtus pennsylvanicus, respond.to the marks of the top-, mid.le-, and.bottom-scent d.nors of an over-mark. We tested.the hypothesis that voles exposed.to an over-mark will respond.preferentially to the scent marks that were d.posited.more recently, the scent marks that were on top or near the top of the over-mark, compared.to the scent marks that were d.posited.earlier or near the bot- tom of the over-mark. Voles spent more time investigating the mark of the top-scent d.nor than that of the either the mid.le- or bottom-scent d.nor. However, males but not female voles spent more time investigating the mid.le-scent mark than the bot- tom-scent mark. We also tested.the hypothesis that voles evaluate and.respond.to over-marks d.fferently from single scent marks. Voles spent more time investigating the marks of the top-, mid.le-, and.bottom-scent d.nors compared.to scent marks that were not part of the over-mark. Voles can d.stinguish among the overlapping scent marks of three scent d.nors and.sex d.fferences exist in the values they appear to attach to each of these scent marks [Current Zoology 57 （4）： 441-448, 2011 ].
