Metabolic signatures of Arabidopsis thaliana abiotic stress responses elucidate patterns in stress priming,acclimation,and recovery

Temperature,water,and light are three abiotic stress factors that have major influences on plant growth,development,and *** can be primed by a prior mild stress to enhance their resistance to future *** used an untargeted metabolomics approach to examine Arabidopsis thaliana 11-day-old seedling’s abiotic stress responses including heat(with and without priming),cold(with and without priming),water-deficit and high-light before and after a 2-day-recovery *** of the physiological phenotypes showed that seedlings with stress treatment resulted in a reduction in fresh weight,hypocotyl and root length but remained *** stress responsive metabolites were identified,confirmed with reference standards,quantified,and *** identified shared and specific stress signatures for cold,heat,water-deficit,and high-light *** metabolism including amino acid metabolism,sugar metabolism,glycolysis,TCA cycle,GABA shunt,glutathione metabolism,purine metabolism,and urea cycle were found to undergo changes that are fundamentally different,although some shared commonalities in response to different *** increases in cysteine abundance and decreases in reduced glutathione were observed following multiple stress treatments highlighting the importance of oxidative stress as a general phenomenon in abiotic *** fold increases in low-turnover amino acids and maltose demonstrate the critical role of protein and starch autolysis in early abiotic stress responses.

Electro–optic response in thin smectic C* film with chevron structures

The effects in electrostatic models of chevron surface-stabilized ferroelectric liquid crystals are investigated through numerical modeling. To study smectic C＊ director distribution within the cell, we consider two nonlinear approaches： the chevron interface does not interplay with the electric field; the electric field interplays with the chevron interface. The obtained results of the director field distribution are compared with the earlier linearized studies. We find that whether or not the electric field interplays with the chevron interface, the electro-optic response requires a generalized approach for its description. The threshold electric field, which is necessary for switching between two stable director states in the chevron cell is evaluated. This study suggests that, in many cases of practical interest, electro-optic response to the electric field and the threshold electric field can be precisely estimated. We argue that, beside being numerically efficient, our approach provides a convenient and a novel standpoint for looking at the electro-optic response problem.

Optimization of a continuous ultrasound assisted oxidative desulfurization(UAOD) process of diesel using response surface methodology(RSM) considering operating cost

A continuous-flow ultrasound-assisted oxidative desulfurization(UAOD)of partially hydro-treated diesel has been investigated using hydrogen peroxide-formic acid as simple and easy to apply oxidation *** effects of different operating parameters of oxidation stage including residence time(2–24 min),formic acid to sulfur molar ratio(10–150),and oxidant to sulfur molar ratio(5–35)on the sulfur removal have been studied using response surface methodology(RSM)based on Box–Behnken *** the operating costs of the continuous-flow oxidation stage including chemical and electrical energy consumption,the appropriate values of operating parameters were selected as follows:residence time of 16 min,the formic acid to sulfur molar ratio of 54.47,and the oxidant to sulfur molar ratio of *** these conditions,the sulfur removal and the volume ratio of the hydrocarbon phase to the aqueous phase were 86.90%and 4.34,*** drastic reduction in the chemical consumption in the oxidation stage,the volume ratio of the hydrocarbon phase to the aqueous phase was increased up to ***,the formic acid to sulfur molar ratio and the oxidant to sulfur molar ratio were obtained 23.64 and 3.58,respectively,which lead to sulfur removal of 84.38%with considerable improvements on the operating cost of oxidation stage in comparison with the previous works.

PoreScript:Semi-automated pore size algorithm for scaffold characterization

The scaffold pore size influences many critical physical aspects of tissue engineering,including tissue infiltration,biodegradation rate,and mechanical *** measurements of pore sizes from scanning electron micrographs using ImageJ/FIJI are commonly used to characterize scaffolds,but these methods are both time-consuming and subject to user *** semi-automated analysis tools are limited by a lack of accessibility or limited sample size in their verification *** work here describes the development of a new MATLAB algorithm,PoreScript,to address these *** algorithm was verified using three common scaffold fabrication methods(e.g.,salt leaching,gas foaming,emulsion templating)with varying pore sizes and shapes to demonstrate the versatility of this new *** results demonstrate that the pore size characterization using PoreScript is comparable to manual pore size *** PoreScript algorithm was further evaluated to determine the effect of user-input and image parameters(relative image magnification,pixel intensity threshold,and pore structure).Overall,this work validates the accuracy of the PoreScript algorithm across several fabrication methods and provides user-guidance for semi-automated image analysis and increased throughput of scaffold characterization.

Mining diversity of the natural biorefinery housed within Tipula abdominalis larvae for use in an industrial biorefinery for production of lignocellulosic ethanol

Although they are the largest taxonomic group of animals, relatively few insects have been examined for symbiotic relationships with micro-organisms. However, this is rapidly changing because of the potential for examination of the natural insectmicrobe-lignocellulose interactions to provide insights for biofuel technology. Micro-organisms associated with lignocellulose-consuming insects often facilitate the digestion of the recalcitrant plant diet; therefore these microbial communities may be mined for novel lignocellulose-degrading microbes, or for robust and inexpensive biocatalysts necessary for economically feasible biofuel production from lignocellulose. These insect-microbe interactions are influenced by the ecosystem and specific lignocellulose diet, and appre- ciating the whole ecosystem-insect-microbiota-lignocellulose as a natural biorefinery provides a rich and diverse framework from which to design novel industrial processes. One such natural biorefinery, the Tipula abdominalis larvae in riparian ecosystems, is reviewed herein with applications for biochemical processes and overcoming challenges involved in conversion of lignocellulosic biomass to fuel ethanol. From the dense and diverse T. abdominalis larval hindgut microbial community, a cellulolytic bacterial isolate, 27C64, demonstrated enzymatic activity toward many model plant polymers and also produced a bacterial antibiotic. 27C64 was co-cultured with yeast in fermentation of pine to ethanol, which allowed for a 20% reduction of commercial enzyme. In this study, a micro-organism from a lignocellulose-consuming insect was successfully applied for improvement of biomass-to-biofuel technology.

Paraneoplastic Leukocytosis and Thrombocytosis as Prognostic Biomarkers in Non-small Cell Lung Cancer

Background and Objectives: Search for inexpensive laboratory markers have identified associations between blood counts and lung cancer outcomes. In this study, we evaluated the prognostic value of paraneoplastic leukocytosis(p-Leukocytosis) and paraneoplastic thrombocytosis(p-Thrombocytosis) in patients with non-small cell lung cancer(NSCLC). We also studied their relation to the expression of commonly detected molecular markers. Methods: We conducted a retrospective chart review on 571 consecutive NSCLC patients over a 10 year period. Blood counts were recorded at the time of cancer diagnosis. Kaplan-Meier survival curves were used to compare overall survival(OS) between patients with and without p-Leukocytosis(or) p-Thrombocytosis(p-Leuko/Thrombocytosis). Cox regression was used to determine if leukocytosis/thrombocytosis was a predictor of OS in ***: Patients with p-Leukocytosis and p-Thrombocytosis had a significantly poorer survival compared patients with normal blood counts(P<0.001). In a multivariate survival analysis, both continued to correlate even when adjusted for histology, gender, stage and chemotherapy(P<0.01, 0.03 respectively). Stage I and II NSCLC with p-Leuko/Thrombocytosis did not perform poorly compared to stage I/II NSCLC patients without paraneoplasia. Patients with the combined leukothrombocytosis syndrome did not have worse outcomes compared to those with either paraneoplastic syndrome alone. Conclusions: p-Leuko/Thrombocytosis is an accessible laboratory parameter of prognostic value in NSCLC. Evidence of p-Leuko/Thrombocytosis portends poor survival. The role of various cytokines in tumor pathobiology provides a rationale for identifying cytokine factors responsible for the paraneoplasia and administering anti-cytokine therapies alongside traditional chemotherapy in an attempt to improve survival outcomes in these subset of patients.

Mediterranean dietary components are inversely associated with advanced colorectal polyps:A case-control study

AIM To evaluate the association between the Mediterranean diet(MD) pattern and its components, and advanced colorectal polyps(adenoma and serrated adenoma).METHODS A case-control study among patients undergoing screening, diagnostic or surveillance colonoscopies during 2010-2015 at the Tel-Aviv Medical Center, Gastroenterology Department. Cases with advanced polyps were defined as: Advanced adenoma [> 10 mm, with features of high grade dysplasia(HGD) or villous histology], advanced serrated adenoma(> 10 mm or with dysplasia) or multiple(≥ 3) non-advanced adenomas or serrated adenomas. Cases of non-advanced adenomas were defined as adenomas < 10 mm, without features of HGD or villous histology. Controls were defined as those without polyps at the current colonoscopy and without a history of colorectal polyps. Data collection included: anthropometrics measured according to a standardized protocol, fasting blood tests performed at the same lab, medical history recorded by a structured interview and dietary intake evaluated by a 116-item food frequency questionnaire. Adherence to the MD components was evaluated according to intake above/below the sample median, for potentially beneficial/detrimental components respectively, as accepted. RESULTS We recruited 206 cases with advanced polyps, 192 cases with non-advanced adenoma and 385 controls. The number of adhered MD components was inversely associated with a diagnosis of advanced polyps in a dose-response manner(OR = 0.34, 95%CI: 0.17-0.65; OR = 0.22, 95%CI: 0.11-0.43; and OR = 0.18, 95%CI: 0.07-0.47 for 3-4, 5-7 and 8-10 components, respectively), but not with non-advanced adenomas(OR = 0.54, 95%CI: 0.25-1.13; OR = 0.48, 95%CI: 0.23-0.99; and OR = 0.43, 95%CI: 0.16-1.12 for 3-4, 5-7 and 8-10 components, respectively). Low intake of sugar-sweetened beverages and red meat, as well as high intake of fish, were inversely associated with advanced polyps(OR = 0.56, 95%CI: 0.36-0.87; OR = 0.63, 95%CI: 0.42-0.95; and OR = 0.66
